Acid-soluble phosphorus compounds in mammalian semen.
Abstract: 1. A method is described for the extraction, purification and separation of acid-soluble phosphorus compounds from mammalian semen. [8-(14)C]ATP and [8-(14)C]AMP were used as internal recovery standards to measure the breakdown and loss of these nucleotides in the procedure. 2. Bull, ram, boar and stallion semen was separated into seminal plasma and spermatozoa and the two fractions were examined separately. The overall composition of the mixture of the phosphorus compounds extracted from the two fractions was similar for the four species. 3. Glycerylphosphorylcholine and glycerylphosphorylinositol were the two phosphorus compounds identified in extracts of seminal plasma. ATP, ADP, AMP, GTP, GDP, NAD, fructose 1,6-diphosphate and glucose 6-phosphate were identified in extracts prepared from spermatozoa.

The researchers have developed a method for extracting, purifying, and separating acid-soluble phosphorus compounds from mammalian semen. They used this method to examine the composition of these compounds in both seminal plasma and sperm cells, identifying specific compounds in each.
Methodology
- The researchers devised a method for extraction, purification, and separation of acid-soluble phosphorus compounds from mammalian semen. This method involved using [8-(14)C]ATP and [8-(14)C]AMP as internal recovery standards to track the degradation and loss of these nucleotides during the process.
Separation of Semen Samples
- The team collected semen samples from four different mammalian species: bull, ram, boar, and stallion. The semen from these animals was then divided into seminal plasma and spermatozoa – the two main components of semen. These fractions were then examined separately.
- The researchers observed that the overall composition of the phosphorus compounds extracted from both the seminal plasma and spermatozoa was similar across all four species.
Identification of Phosphorus Compounds
- In their examinations, they identified two specific phosphorus compounds in the extracts of seminal plasma: glycerylphosphorylcholine and glycerylphosphorylinositol. Both of these compounds are important for semen function: glycerylphosphorylcholine improves the motility of spermatozoa, while glycerylphosphorylinositol plays an important role in cellular signaling.
- From the spermatozoa extracts, they identified eight phosphorus compounds: ATP, ADP, AMP, GTP, GDP, NAD, fructose 1,6-diphosphate, and glucose 6-phosphate. These compounds are all involved in the energy metabolism of the sperm cells, helping to fuel their movement and activity.
