Hemosiderin; isolation from horse spleen and characterization.
Abstract: 1. A method is outlined for isolating hemosiderin from horse spleen by sedimentation from salt solutions of different specific gravities. 2. The iron, nitrogen, phosphorus and ash concentration varied considerably in hemosiderin samples obtained from different spleens. Fractionation of these preparations by sedimentation in organic liquids showed that fractions could be obtained which varied in their iron concentrations from about 25 to 41% iron. 3. Hemosiderin has been found to contain hexoseamine, galactose, mannose and fucose.

This research outlines a method for isolating a naturally occurring iron-storage substance, hemosiderin, from horse spleen and provides details regarding its varied composition of iron, nitrogen, phosphorus and ash.
Method of Isolating Hemosiderin
- The researchers have detailed a method of extracting hemosiderin – which is a form of stored iron in the body – from horse spleen.
- This process is accomplished through sedimentation using different salt solutions with varying degrees of specific gravity.
Variations In Chemical Concentration
- It was found that the chemical concentration of iron, nitrogen, phosphorous, and ash significantly varied in hemosiderin samples collected from different spleens.
- By segregating these samples through sedimentation in organic liquids, fractions with different iron concentrations, ranging between 25-41%, were revealed.
Hemosiderin Constituents
- Furthermore, the study found that hemosiderin contains several sugar-type substances: hexoseamine, galactose, mannose, and fucose.
- These components suggest a complex biochemical composition and functional role of hemosiderin in the body, which is important for understanding the body’s iron storage system and could have implications for managing iron-related conditions.
