[Method of isolating and controlling fluorescent Fab fragments of antibodies against horse serum proteins].

Abstract: For the first time the lyophilized fluorescent Fab-fragments of rabbit antibodies to horse serum protein, suitable for detecting different antigens and antibodies to these antigens, have been obtained by the specially developed method. The criteria to be used in the control of the antispecific fluorescent fragments of antibodies have been described and the methods of their control before and after lyophilization have been developed. The use of the antispecific fluorescent Fab-fragments of antibodies has been shown to considerably accelerate and simplify the indirect immunofluorescent assay.

This research article presents a method for isolating fluorescent Fab fragments from antibodies that target horse serum proteins. These fluorescent fragments have potential applications for detecting various antigens and antibodies. The authors also devise protocols for their quality control before and after lyophilization.

Isolation of Fluorescent Fab Fragments

  • The study primarily revolves around a novel method to extract fluorescent Fab fragments from rabbit antibodies that are reactive against horse serum proteins.
  • Fab fragments are parts of an antibody that bind to specific antigens. They are crucial for diagnostic and therapeutic applications due to their selective binding capacities.
  • The process derived in this study enables the researchers to obtain these fragments in a lyophilized (freeze-dried) state, which adds to their long-term stability.

Quality Control of Fab Fragments

  • The researchers also establish the standards for assessing the quality of the isolated fluorescent Fab fragments.
  • These guidelines provide a framework to evaluate the antispecific fluorescent antibody fragments reliability, based on which the quality control procedures before and after lyophilization can be executed.

Applications of Fab Fragments

  • The study demonstrates that these fluorescent Fab fragments can be crucial for indirect immunofluorescent assays.
  • Immunofluorescent assays detect the presence of certain substances (like antigens or antibodies) based on the principle of specific binding between an antibody and an antigen. The use of these fluorescent Fab fragments could enhance the speed and convenience of such assays, leading to quicker disease detection rates.
