[A multiplex PCR method based on nuclear and cytoplasmic inheritance to identify the horse and donkey-derived components of Asini Colla Corii and the hide].
Abstract: To identify the original components of Asini Colla Corii and its raw material hides provides a guarantee for authenticity of Asini Colla Corii. It is urgent for Asini Colla Corii production enterprises and market supervision departments to develop effective identification methods of Asini Colla Corii and hides derived from horses, donkeys, mules and hinnies. This study screened species-specific DNA sequences of nuclear and mitochondrial genomes as detection targets, designed horse and donkey specific primers and established multiple PCR identification methods for identifying the animal hides (including the horse, donkey, mule and hinny) and Asini Colla Corii containing horse-derived and donkey-derived components. Our method can identify the horse, donkey, mule and hinny hides and horse, donkey-derived components of Asini Colla Corii with high species specificity (no crossed amplification was observed ). The limit of detection was 0.2 ng DNA. The method developed in this study provides technical support for Asini Colla Corii production enterprises and market supervision departments. .

This research focuses on effectively identifying the original components of Asini Colla Corii and the hides used as its raw materials. It has established a multiplex PCR method to distinguish horse, donkey, mule, and hinny hides, thus ensuring the authenticity of Asini Colla Corii.
Identification of Asini Colla Corii Components
- The objective of the study is to enhance the authenticity of Asini Colla Corii, a product made from equine hides, by establishing effective identification methods.
- Asini Colla Corii production businesses and market supervision departments need effective methods for identifying original components such as horse, donkey, mule, and hinny hides.
Use of Multiplex PCR Method
- To achieve this, the study uses a multiplex PCR method, a process that has been developed to identify multiple targets in a single PCR experiment.
- This method focuses on distinguishing between specific DNA sequences of different equine species’ nuclear and mitochondrial genomes.
- The researchers designed specific primers for horse and donkey hides that serve as key identifiers.
Results of the Study
- Through the multiplex PCR method, the study successfully identifies and differentiates among horse, donkey, mule, and hinny hides.
- This method determines the species of hide used in Asini Colla Corii with a high level of specificity, reliable enough to prevent cross-amplification – a problem that arises when the PCR process unintentionally multiplies non-target DNA sequences.
- The limit of detection of this method stands at 0.2 ng DNA, indicating its high sensitivity.
Significance of the Study
- This research provides technical guidance and infrastructure for Asini Colla Corii production enterprises and market supervision departments, helping them authenticate the original components of their product.
- By ensuring the validity of raw materials and the finished product, this work aims to maintain quality control and build consumers’ trust in Asini Colla Corii.
