Effect of a specific platelet-activating factor antagonist on cardiovascular and peripheral cellular responses to colonic ischemia and reperfusion in anesthetized ponies.

This research investigates the effects of a specific type of drug called a Platelet-Activating Factor (PAF) antagonist on responses in the cardiovascular system and cells in the extremities of anesthetized ponies, during a procedure of induced colonic ischemia and reperfusion. The outcomes indicate that while the drug did not impact the cardiovascular responses, it affected specific responses in the cells located in the peripheral areas.

Methods and Findings

• The researchers experimented on twelve anesthetized ponies. They induced a condition called large-colon torsion, which is a twist in the large colon causing interruption of blood flow (ischemia), followed by restoration of blood flow (reperfusion).
• They divided the ponies into two groups. One group of six ponies was given a specific PAF antagonist (a kind of drug that blocks the action of PAF, named WEB 2086). The other group (control group) of six ponies were given an equivalent volume of saline solution.
• The study involved monitoring the ponies during the reperfusion period. The onset and progression of two conditions – hypotension (low blood pressure) and metabolic acidosis (excessive acidity in the body fluids) – were observed in all ponies following colonic torsion correction.
• The cardiac index initially increased but then significantly decreased over the study duration.
• The time between the correction of torsion and the onset of cardiac failure and death showed no noticeable difference between the two groups.
• There was a significant drop in the platelet count (thrombocytopenia) during the reperfusion phase in the control ponies, which was not observed in the ponies treated with WEB 2086.
• Similarly, the concentration of white blood cells (leukocytes) in the control group ponies was seen to fall immediately upon reperfusion, compared to a more stable concentration in the WEB-treated ponies.

• The study concluded that using a PAF antagonist like WEB 2086 does not prevent cardiovascular responses to colonic ischemia and reperfusion.
