Genetic characteristics of feral Misaki horses based on polymorphisms of microsatellites and mitochondrial DNA.

Abstract: The Misaki horse is a Japanese native horse, known as the "feral horse of Cape Toi". In this study, we acquired the genetic information to establish their studbook, and analyzed their genetic characteristics for conservation. We genotyped 32 microsatellites and a mitochondrial D-loop region in 77 Misaki horses (80.2% of the population). The average number of alleles, observed heterozygosity, and expected heterozygosity were 3.4, 0.509, and 0.497, respectively. A neighbor-joining phylogenetic tree of individuals was constructed. Moreover, the results suggested that Misaki horses experienced a bottleneck, but it was neither severe nor recent. In addition, three mitochondrial haplotypes were confirmed. Consequently, we clarified the genetic background of Misaki horses that have been resident at Cape Toi for a long time.

This research explores the genetic characteristics of Japanese native Misaki horses. DNA analysis is conducted with the goal of establishing their genetic diversity and history while also setting the groundwork for proper conservation efforts.

Research Objective

  • The study aimed to enhance the understanding of the genetic makeup of the Misaki horses using microsatellite and mitochondrial DNA polymorphisms.
  • Results from this research intended to guide future conservation efforts and breeding plans to preserve the genetic uniqueness of Misaki horses.

Methodology

  • The research involved 77 Misaki horses, equivalent to 80.2% of the living Misaki horse population.
  • The scientists used 32 microsatellites and a mitochondrial D-loop region for genotyping each horse.
  • The number of alleles, observed heterozygosity, and expected heterozygosity were calculated, with averages of 3.4, 0.509, and 0.497 respectively being obtained.
  • A neighbor-joining phylogenetic tree was constructed to map the genetic relationships among individuals.

Investigation Findings

  • The results suggested that the Misaki horses experienced a genetic bottleneck, a process that occurred when a significant percentage of their population was wiped out.
  • The bottleneck event, however, was neither severe nor recent, implying a sustained size of the Misaki horse population over more recent times.
  • Additionally, three different mitochondrial DNA haplotypes were confirmed in the horses, indicating the presence of different maternal lineages within the population.
  • The study thus elucidated on the genetic background of the Misaki horses that have been residing at Cape Toi for many generations.
