Light and electron microscopy of Ag-NORs in domestic horse chromosomes identified after R-banding.
Abstract: Silver staining shows the presence in the domestic horse of six NORs located on chromosomes 1, 26 and 31 as identified after R-banding. Following electron microscopy, the argyrophilic material was observed outside the terminal secondary constrictions (satellite stalks) on the terminal portion of the short arm of chromosome 1, outside the secondary constrictions on the proximal region of the long arms of chromosome 31, and beside the proximal region of the long arms of chromosome 26. Satellite staining applied to these chromosomes appears to reveal only the active NORs.

This study explores the location of six nucleolus organizer regions (NORs) in the chromosomes of a domestic horse using silver staining and R-banding techniques. These NORs were found on chromosomes 1, 26 and 31 with electron microscopy showing the silver stain located in specific areas of these chromosomes.
Methodology
- The researchers used silver staining, a histological technique, to visualize the nucleolus organizer regions (NORs) within the chromosomes of a domestic horse.
- With the help of R-banding, another staining technique, they accurately identified the NORs on specific chromosomes.
- Once visualized and identified, they used electron microscopy to examine these regions in closer detail. The electron microscopy provided a more high-resolution image of these regions, allowing the researchers to precisely locate where the silver-stained, argyrophilic material was located within the chromosomes.
Findings
- The researchers identified six NORs located on chromosomes 1, 26, and 31 of the domestic horse.
- With electron microscopy, the researchers observed the silver-stained, argyrophilic material (indicative of NORs) were found outside of certain regions of these chromosomes.
- On Chromosome 1, the NORs were found outside the terminal secondary constrictions, specifically on the terminal portion of the chromosome’s short arm.
- On Chromosome 31, the argyrophilic material was observed outside the secondary constrictions on the proximal region of the chromosome’s long arms.
- On Chromosome 26, the researchers identified the NORs located beside the proximal region of the long arms.
- The application of satellite staining further revealed that the identified NORs were active. They interpret this to mean these NORs are in use during the transcription process where genetic material is actively being used to create RNA, and ultimately proteins, within the cell.
Implications
- The results of this research provide insight into the genetic structure of domestic horses, specifically the location and activity of NORs within their chromosomes.
- NORs are significant as they play a vital role in the organization of the nucleolus and participate in synthesizing ribosomal RNA (rRNA), a critical player in the production of proteins within the cells.
- By understanding the specifics about the structure and activity of these NORs, scientists can better understand the genetic makeup of domestic horses. It can also serve as a foundation for further research into genetic variations or abnormalities that may influence equine health and traits.
