Optimization of solid-phase extraction for the liquid chromatography-mass spectrometry analysis of harpagoside, 8-para-coumaroyl harpagide, and harpagide in equine plasma and urine. Solid-phase extraction cartridges among those usually used for screening in horse doping analyses are tested to optimize the extraction of harpagoside (HS), harpagide (HG), and 8-para-coumaroyl harpagide (8PCHG) from plasma and urine. Extracts are analyzed by liquid chromatography coupled with multi-step tandem mass spectrometry. The extraction process retained for plasma applies BondElut PPL cartridges and provides extraction recoveries between 91% and 93%, with RSD values between 8 and 13% at 0.5 ng/mL. Reversed-phase liquid chromatographic method for estrogen determination in equine biological samples. Equine unsaturated estrogens are the main components of brand formulations indicated for hormonal replacement therapy in both hypogonadic and postmenopausal women. These hormones are produced by the fetoplacental unit during equine gestation. A method is described for the quantitative determination of equilenin (EL), equilin (EQ), 17alpha-dihydroequilin (17dEQ), and estrone (El) in the plasma of a pregnant mare. Immunoaffinity chromatography in the detection of dexamethasone in equine urine. Due to the widespread use of dexamethasone in racing horses, mostly in low doses by intra-articular administration for the treatment of inflammatory processes, a method is developed to detect this drug in horse urine samples using liquid-liquid extraction followed by immunoaffinity chromatography. Liquid chromatography with diode-array detection is used for the identification of the drug. The use of immunoaffinity columns enhances the selectivity of the analysis, and the results show that dexamethasone can be detected up to 28 h after intra-articular administration.
Gas/liquid chromatographic analysis of pemoline in biological fluids using electron capture detection. An analytical gas/liquid chromatographic (GLC) protocol is described for the quantitation of pemoline in biological fluids of the horse. Plasma samples containing known quantities of pemoline and its analog as an internal standard (IS) were deproteinized with 5-sulfosalicylic acid, heated at 80 degree C, and centrifuged. 5-Phenyl-2,4-oxazolidinedione, the hydrolytic product of pemoline in acid medium, was extracted with dichloromethane (DCM). Mefanamic acid blood and urine levels in the horse determined by derivative gas-liquid chromatography-electron capture. Mefenamic acid is extracted from biological fluids and is acylated with pentafluoropropionic anhydride to form a derivative possessing high electron affinity. The derivative is analyzed by gas-liquid chromatography with an electron capture detector. The method is particularly valuable for determining drug levels in blood where small sample and/or drug concentrations are available.
