Nitrite and nitrotyrosine concentrations in articular cartilage, subchondral bone, and trabecular bone of normal juvenile, normal adult, and osteoarthritic adult equine metacarpophalangeal joints. Osteoarthritis (OA) is a chronic debilitating joint disorder in which the importance of inflammation is increasingly recognized. In advanced cases, both the articular cartilage and the underlying bony layers are affected, but the exact sequence of events and their localization in the initial phase of pathogenesis remain uncertain. We measured nitric oxide (NO) end products in tissue layers that constitute the bearing surface of the joint, as possible indicators of physiological and pathological processes. Methods: Nitrite as a measure for NO and nitrotyrosine was measured in articular cartilag...
Effect of radiosynovectomy with holmium-166 ferric hydroxide macroaggregate on adult equine cartilage. To analyze the effect of radiosynovectomy with holmium-166 ferric hydroxide macroaggregate (166Ho-FHMA) on articular cartilage in 6 adult horses. Methods: Arthritic changes and mechanical properties of articular cartilage were evaluated with arthroscopy and postmortem microscopic analyses. Glycosaminoglycan content was measured by safranin-O staining combined with digital densitometry, uronic acid analyses, and dimethylene blue binding assay. 35S-sulfate labeling and autoradiography were used to localize proteoglycan synthesis and to characterize proteoglycan structures using SDS-agarose gel e...
Effect of methylprednisolone acetate on proteoglycan and collagen metabolism of articular cartilage explants. The effect of different doses of methylprednisolone acetate (MPA) on proteoglycan and collagen metabolism of articular cartilage from normal equine joints was tested in vitro. Methods: Cultured explants were treated with 0, 0.0004, 0.004, 0.04, 0.4 and 4.0 mg/ml (approximately 10(-6)-10(-2) M) MPA for 72 h. Proteoglycan synthesis was measured by incorporation of sodium [35S]sulfate into proteoglycans and proteoglycan degradation was measured by release of total and radiolabeled proteoglycan into the culture media. The size of the proteoglycans was assessed with size exclusion chromatography un...
The molecular weight of therapeutic hyaluronan (sodium hyaluronate): how significant is it? Various molecular weight hyaluronic acid (HA) preparations have been injected into joints for the treatment of human and equine osteoarthritis. A therapeutic advantage has been claimed for commercial products with a molecular weight in the range found in normal synovial fluid (SF), compared to lower molecular weight products. But a correlation between molecular weight and efficacy is not borne out by an analysis of the available literature on clinical results. SF viscosity, HA concentration, HA molecular weight and rate of synthesis in joint disease. It is proposed that the beneficial effect o...
Oxygen radical production by horse and pig neutrophils induced by a range of crystals. The exposure of animal neutrophils to crystals implicated in joint inflammation induces superoxide and peroxide generation in a concentration and temperature dependent fashion. Active crystals were urate, hydroxyapatite, pyrophosphate and brushite; diamond and cholesterol were inactive. Cytochalasin B increased superoxide yield after addition of brushite, and inhibitors of the PMA induced respiratory burst blocked the crystal induced response. Addition of urate to anaerobic neutrophils causes the reduction of a b-type cytochrome, a likely component of the neutrophil superoxide generating syste...
