Characterization of semen collected by pharmacologically induced ejaculation from a stallion with seminal vesiculitis.

The study explores and finds that pharmacological initiation of ejaculation can successfully obtain sperm of better quality and minimal contamination in stallions with seminal vesiculitis when compared to the conventional artificial vagina method.

Comparison of Semen Collection Methods

• This investigation involved sperm collection from a ten-year-old thoroughbred stallion with seminal vesiculitis, a reproductive disease in male horses that results in inflammation of the seminal vesicles.
• Two semen collection methods were compared: the traditional artificial vagina technique and a new method of pharmacologically induced ejaculation.
• The pharma-induced ejaculation involved the intravenous administration of two substances: detomidine, a sedative used in veterinary medicine, and oxytocin, a hormone known to stimulate uterine contractions and enhance social bonding. The dosage for the former was 0.01 mg/kg and for the latter was 20 IU.

Evaluation of Semen Characteristics

• Various aspects of the semen samples were evaluated including sperm motility, plasma membrane and acrosome integrity (PMAI), reactive oxygen species (ROS) levels, polymorphonuclear neutrophil (PMN) percentage, and bacterial profiles of both fresh and cool-stored semen at 5°C for 24 hours.

• The study found that semen obtained by the pharmacologically induced method displayed lower volume yet superior sperm motility in cooled samples when compared to semen obtained through the artificial vagina technique.
• In addition, the semen collected using the pharmacological method demonstrated a fewer PMN percentage, a type of white blood cell, hinting at a lower inflammation in the seminal fluid.
• Semen from this approach also showcased increased levels of PMAI, suggesting a higher degree of viable and healthy sperm cells, and lower levels of ROS, which are damaging oxygen molecules.
• The research therefore concludes that pharmacologically induced ejaculation offers an effective alternative for obtaining higher quality and longer-lasting semen with lower contamination from stallions suffering from seminal vesiculitis.
