Determination of the charge of horse kidney metallothionein by free boundary electrophoresis.

Abstract: Traditionally, the charge of a protein molecule as determined by electrophoresis has been compared to that revealed by pH titration, and any lack of coincidence has been ascribed to ion binding, and the two results have been brought into agreement by adjustment of binding parameters (1). Metallo-thionein allows a unique opportunity to examine the validity of the electrophoretic approach, since the amino acid sequence and metal atom binding studies allow the absolute charge of the molecule to be computed (2). This then can be compared to the charge determined from electrophoretic mobility measurements to allow evaluation of the theories relating charge to mobility.

“The research study explores the method of determining the charge of a protein molecule, specifically the horse kidney metallothionein, using free boundary electrophoresis and comparing the outcomes with traditional methods like pH titration.”

Introduction

  • This research revolves around a unique protein molecule – the horse kidney metallothionein.
  • The charge of a protein molecule like this traditionally has been determined using electrophoretic methods, where the outcomes are compared with pH titration methods.
  • Any inconsistency found between the two methodologies is usually attributed to ion binding and then adjusted accordingly.

Metallothionein and Its Significance

  • Metallothionein, the study’s object, offers a distinctive opportunity to assess the validity of the electrophoretic approach.
  • This is because for metallothionein, both the amino acid sequence and metal atom binding studies permit the absolute charge of the molecule to be calculated.
  • Such calculated charge can provide a reliable basis for comparison with the electrophoretically determined charge.

Evaluating Theories relating to Charge and Mobility

  • By comparing the computed absolute charge of the molecule to its electrophoretically determined charge, this research evaluates the existing theories regarding the relationship between a molecule’s charge and its mobility.
  • This evaluation can help refine the techniques of determining protein molecule charges, improving the reliability and accuracy of such methods.
