Development of a loop-mediated isothermal amplification assay for rapid detection of Burkholderia mallei.

Abstract: The present study was conducted to establish a Loop-mediated isothermal amplification (LAMP) technique for the rapid detection of B. mallei the etiologic agent of glanders, a highly contagious disease of equines. A set of six specific primers targeting integrase gene cluster were designed for the LAMP test. The reaction was optimized using different temperatures and time intervals. The specificity of the assay was evaluated using DNA from B.pseudomallei and Pseudomonas aeruginosa. The LAMP products were analyzed both visually and under UV light after electrophoresis. The optimized conditions were found to be at 63ºC for 60 min. The assay showed high specificity and sensitivity. It was concluded that the established LAMP assay is a rapid, sensitive and practical tool for detection of B. mallei and early diagnosis of glanders.

This study is about developing a fast and sensitive Loop-mediated isothermal amplification (LAMP) technique for detecting Burkholderia mallei, the causative agent of glanders in equines.

Objective of the Study

  • The goal of this study was to develop and establish a Loop-Mediated Isothermal Amplification (LAMP) technique for fast and efficient detection of Burkholderia mallei. This bacteria is responsible for causing a highly infectious disease known as glanders in equines.

Methodology for the Study

  • The researchers designed six specific primers with focus on the integrase gene cluster for the LAMP test.
  • The test conditions were then fine-tuned by trying different temperatures and time intervals.
  • In order to evaluate the specificity of the test, the researchers used the DNA from B.pseudomallei and Pseudomonas aeruginosa.
  • Furthermore, the LAMP test results were evaluated visually and also under UV light after electrophoresis.

Results from the Study

  • The optimal conditions for the LAMP test observed in the study were found to be 63ºC for 60 min.
  • The LAMP test demonstrated strong specificity and sensitivity during the study.

Conclusion of the Study

  • Based on the results, the researchers concluded that the established LAMP assay provides a fast, sensitive, and practical tool for the detection of B. mallei.
  • This would in turn aid in the early diagnosis of glanders, therefore supporting effective disease management strategies.
