Evaluation of the antiviral activities of Bacharis dracunculifolia and quercetin on Equid herpesvirus 1 in a murine model.
Abstract: Equid herpesvirus 1 (EHV-1) is a pathogen of high economic importance in equine breeding operations around the world. EHV-1 infection causes respiratory, neurologic and reproductive disease. The absence of an efficient therapy has caught the attention of the scientific community and the therapeutic activities of natural products with its antivirals effects might be effective for the disease's treatment. Herein it was evaluated the prophylactic and therapeutic potential of quercetin and ethanolic extracts of Bacharis dracunculifolia formulations compared to Penciclovir® in an in vivo EHV-1 infection model. Six to seven-week-old female C57BL/6 mice were randomly organized into fifteen groups with six animals each. Ex-1 represents the treatment post-challenge groups to assess morbidity, mortality and weight variation. Ex-2 represents the animals that received treatment for 5 days post-challenge for lesion evaluation. In Ex-3 animals were treated prior to viral challenge to assess morbidity, mortality and weight variation. All mice in the treatment groups were challenged by intranasal inoculation of 3.0 × 105 TCID50 EHV-1. The quercetin and B. dracunculifolia treatment decreased morbimortality in post-challenge treatment (Ex-1) and EHV-1 related lesions (Ex-2). Treatment prior to viral challenge (Ex-3) did not show any significant results. Based on the results of the present study, both tested formulations are promising antiviral agents for the treatment of EHV-1 infection.

The study examines and evaluates the potential antiviral benefits of the plant Bacharis dracunculifolia and the compound quercetin on controlling an equine herpesvirus 1 (EHV-1) infection in a mouse model. It suggests these substances could be promising therapeutic agents for treating EHV-1 infection, although they show limited prophylactic utility.
Study Design
- The research revolved around Equid herpesvirus 1 (EHV-1), a pathogen impacting the equine breeding industry worldwide by causing respiratory, neurological, and reproductive disease.
- To study the possible therapeutic options for this disease, the study tested the potential antiviral benefits of quercetin—a flavonoid found in many plants—and ethanolic extracts of Bacharis dracunculifolia, a plant native to South America.
- The comparison control used was Penciclovir®, a known antiviral medication.
- The study was performed on six to seven-week-old female C57BL/6 mice, arranged into fifteen groups of six.
- Three distinct experiments were performed—Ex-1 to assess post-viral challenge morbidity, mortality, and weight variation; Ex-2 to evaluate lesions five days post-challenge; and Ex-3 to discern any effects of pre-viral challenge treatment on morbidity, mortality, and weight variation.
Results
- Both Quercetin and B. dracunculifolia extract showed a reduction in morbidity and mortality rates when administered after a viral challenge (Ex-1).
- They were also effective in decreasing the severity of EHV-1-related lesions during post-challenge treatment (Ex-2).
- However, the study reported no significant prevention benefits when the substances were used prior to a viral challenge (Ex-3).
Conclusion
- The research concludes that both B. dracunculifolia extract and quercetin have the potential to be developed as antiviral agents for the treatment of EHV-1 infection.
- The results obtained from this study bring hope for the equine industry as they may lead to effective treatment options for diseases caused by EHV-1. The therapeutic potential of these natural compounds against viral infections needs to be further researched to fully understand their mechanism of action and to validate these encouraging results.
