Immunoassay detection of drugs in horses. I. Particle concentration fluoroimmunoassay detection of fentanyl and its congeners.

The researchers studied a rapid and sensitive technique, Particle Concentration Fluorescence Immunoassay (PCFIA), for detecting drugs in racehorses. It showed promise in detecting fentanyl and its similar drugs, even in small concentrations, by observing and measuring changes in fluorescence.

Methodology

• The research was centered on the use of Particle Concentration Fluorescence Immunoassay (PCFIA) for drug detection in horse racing. PCFIA involves analyzing a substance by detecting a change in fluorescence brought about by the binding between the substance in question (in this case, drugs) and a fluorescent-marker-labeled antibody.
• An antiserum to a carboxyfentanyl-BSA (bovine serum albumin) conjugate and carboxyfentanyl linked to b-Phycoerythrin were used in the PCFIA system.
• Testing was able to detect levels of Fentanyl—a potent painkiller commonly used in veterinary medicine but also has potential for misuse—as low as 0.1 ng/ml, showing the high sensitivity of the test.

Cross-Reactivity and Pharmacological Effects

• The antiserum used in the assay showed good cross-reactivity with other drugs similar to fentanyl, like carfentanil, sufentanil, and the methylated analogs of fentanyl.
• Researchers evaluated the possible effects of these drugs on horses as well as the test’s ability to detect drug doses that can significantly impact horse performance.
• Each of these drugs triggered noticeable locomotor responses in horses at doses between 0.1 and 10 micrograms/kg, with carfentanyl being the most potent, followed by 3-methylfentanyl, sufentanil, and fentanyl.
• When administered to horses at doses high enough to induce a pharmacological response, all the drugs caused sufficient change in fluorescence in the PCFIA system to be detectable in post-race horse urine samples.

Conclusion

• The PCFIA method was found to be faster than radioimmunoassay, another common technique for drug detection. Furthermore, PCFIA showed similar sensitivity but required less instrumentation.
• Given these advantages, the researchers concluded that PCFIA holds substantial potential as a drug testing technique in horse racing, providing swift, sensitive detection even at lower levels of drug concentrations.
