Influence of furosemide on the detection of flunixin meglumine in horse urine samples.
Abstract: The possibility of false negative results from TLC when a diuretic is administered concomitantly with flunixin was studied. Samples were subjected to solvent extraction from acidic aqueous solutions; duplicate samples were also subjected to alkaline hydrolysis at pH 12.5. The internal standard was flufenamic acid. The quantification of flunixin was performed by HPLC and the results confirmed by GC/MS. The data show that furosemide influences the urinary concentration of flunixin.

The research article discusses a study exploring the effect of a diuretic called furosemide on the detection of a drug named flunixin in horse urine samples, suggesting that the presence of furosemide could potentially lead to false negative results.
Research Methodology
- The study was initiated due to concerns about potential false negatives arising from Thin Layer Chromatography (TLC) when both a diuretic and flunixin are administered to horses.
- The samples for this experiment were put through a process of solvent extraction from acidic aqueic solutions. Some of the samples were also subjected to alkaline hydrolysis at a level of pH 12.5 in order to duplicate them for further study.
- An internal standard was set up as part of the process, which was flufenamic acid in this case.
Research Findings
- The quantity or amount of flunixin in each sample was measured using High Performance Liquid Chromatography (HPLC).
- Assurance for the results was sought by confirming them through Gas Chromatography/Mass Spectrometry (GC/MS).
- The data collated and analyzed at the end of the research experiment indicated that the presence of furosemide had an impact on the urinary concentration of flunixin in the horse urine samples.
Implications of the Study
- This research suggests that the presence of the diuretic furosemide could potentially alter the concentrations of flunixin in horse urine, thereby impacting the reliability of drug testing results. This is important in various competitive equestrian sports where certain drugs are prohibited.
- The understanding that furosemide can influence the urinary concentration of flunixin provides a basis for further studies to study this interaction more deeply, which may eventually lead to changes in drug testing methods and regulations in competitive horse competitions.
