Isolation, partial identification and quantitative determination of four guaiphenesin glucuronides in plasma and urine of the horse by high-performance liquid chromatography.
Abstract: The isolation, partial identification and quantitative determination of four guaiphenesin glucuronides in plasma and urine of the horse is described. The identity of the glucuronides was checked by UV and fluorescence spectrophotometry, by NMR spectrometry and by mass spectrometry after permethylation. The applicability of the procedure to pharmacokinetic studies is demonstrated.

The research describes methods for isolating and identifying four different types of guaiphenesin glucuronides in horse plasma and urine using high-performance liquid chromatography. Through various spectrometry tests, the researchers verified the identity of the glucuronides and highlighted the potential for utilizing this methodology in pharmacokinetic studies.
Research Methodology
The researchers implemented a detailed procedure to isolate, identify, and quantitatively measure guaiphenesin glucuronides. These substance are found in both plasma and urine of horses.
- They initiated the process by using high-performance liquid chromatography (HPLC), an analytical chemistry technique primarily used for separating, identifying and quantifying each component in a mixture of compounds. Here, it was used for the extraction and separation of four specific guaiphenesin glucuronides from the horse’s body fluids.
- Following isolation, they utilized UV and fluorescence spectrophotometry for partial identification. Ultraviolet (UV) and fluorescence spectrophotometry are methods that use light to determine the concentration and identity of the separated glucuronides.
- Further confirmation was performed using Nuclear Magnetic Resonance (NMR) spectrometry, a sophisticated and highly sensitive research technique that focuses on the magnetic properties of atomic nuclei for chemical structure analysis. The glucuronides were permethylated, a chemical process that adds a methyl group to a molecule, before undergoing mass spectrometry.
- Mass spectrometry is a technique used for the identification and quantitation of chemical substances. It subjects them to a magnetic field to measure their mass-to-charge ratio, providing further checks to the identity of the glucuronides.
Application to Pharmacokinetics
- The research study describes the demonstrated applicability of their isolation and identification procedure in the realm of pharmacokinetic studies. Pharmacokinetics is a branch of pharmacology dedicated to determining the fate of substances administered to a living organism. The principles of pharmacokinetics are used to understand how drugs are metabolized within diverse biological systems.
- The researchers have indicated that their precisely set up procedure for isolating and identifying guaiphenesin glucuronides can be a fundamental approach in pharmacokinetic studies, particularly for understanding biotransformation processes where substances interact with an organism’s biochemical processes.-
