Kanamycin sulfate in the horse: serum, synovial fluid, peritoneal fluid, and urine concentrations after single-dose intramuscular administration.
Abstract: Six healthy adult mares were given a single dose of kanamycin sulfate (200 mg/ml) IM at a dosage rate of 5 mg/kg of body weight. Kanamycin concentrations in serum, synovial fluid, peritoneal fluid, and urine were measured serially over a 48-hour period. The mean peak serum kanamycin concentration was 12.55 microgram/ml at 1 hour. Mean peak kanamycin concentrations in synovial fluid and peritoneal fluid were 7.25 microgram/ml and 9.27 microgram/ml at 2 hours and 3 hours, respectively. These concentrations decreased steadily in parallel with serum concentrations and were still measurable at 48 hours. Urine concentrations of kanamycin were relatively high, with a mean peak concentration of 80.51 microgram/ml at 1 hour.

The research studies how the antibiotic kanamycin sulfate behaves in equine bodies, specifically looking at concentrations in serum, synovial fluid, peritoneal fluid and urine after a single dosage is administered.
Study Design and Participants
- The study was conducted on six healthy adult mares.
- Each mare was administered a single intramuscular (IM) dose of kanamycin sulfate.
- The dose rate was 5 mg/kg of body weight, making it a considerable dose for a horse.
Methodology and Data Collection
- The researchers measured the concentrations of kanamycin in the horse’s serum, synovial fluid, peritoneal fluid and urine over a 48-hour period.
- The measurement was performed serially, meaning the data was collected continuously over the time period.
Study Findings
- The highest average concentration of kanamycin in the serum was found to be 12.55 microgram/ml at 1 hour after administration.
- For synovial fluid and peritoneal fluid, the highest average concentrations were 7.25 microgram/ml and 9.27 microgram/ml at 2 hours and 3 hours respectively.
- All of these concentrations decreased steadily and in parallel with the serum concentrations during the observation period.
- The concentrations were still measurable at the end of the 48-hour observation period.
- Urine concentrations of kanamycin were significantly higher, peaking at 80.51 microgram/ml at 1 hour after administration.
In summary, the study provides valuable data on how an intramuscular dose of kanamycin sulfate is distributed in the body of a horse and how its concentrations change over time in various body fluids. This information can be useful in understanding the pharmacokinetics of the drug in horses, which can inform appropriate dosing strategies.
