Microneutralization test in PK(15) cells for assay of antibodies to louping ill virus.

Abstract: A microneutralization test in PK(15) cells was developed to measure the neutralizing antibody response of a group of ponies experimentally challenged with louping ill virus. Viral cytopathic effect was maximal after 6 days of incubation, at which point titration endpoints were clear-cut and readily determinable. The assay compared favorably with the mouse neutralization test for accuracy and ease of performance.

The study outlines the development of a microneutralization test using PK(15) cells to measure the antibody response of ponies when exposed to the louping ill virus. This method was found to be comparable to the mouse neutralization test for accuracy and ease of application.

Development of Microneutralization Test in PK(15) Cells

  • The research primarily focuses on the development of a microneutralization test using Porcine Kidney 15 (PK15) cells.
  • This novel approach was designed to measure the neutralizing antibody response when ponies were experimentally introduced to the louping ill virus.

Experiment Process and Results

  • Throughout the course of the experiment, it was noted that the viral cytopathic effect was at its highest after six days of incubation.
  • It was at this point that the titration endpoints became clearly distinguishable and could be readily determined.
  • The scientists were then able to measure and determine the level of neutralizing antibodies produced by the ponies in response to the louping ill virus.

Comparison with Mouse Neutralization Test

  • The developed PK(15) cell-based microneutralization test was then compared with the traditional mouse neutralization test.
  • In terms of accuracy and ease of performance, the newly developed test proved to be comparable.
  • This conclusion suggests that this novel testing method can be as effective as the conventional mouse neutralization test, potentially expanding research options in this area.
