Pharmacokinetics and tolerability of a new formulation of omeprazole in the horse.
Abstract: A new formulation of omeprazole in gastro-resistant granules was tested with regard to its pharmacokinetics and tolerability. Twenty-four horses were randomly divided into three groups (8 horses/group) and treated, according a parallel study design, as follows: Group A untreated (control group), Group B received 4 mg/kg of omeprazole, and Group C received 12 mg/kg of omeprazole, both of which were treated orally once a day for 90 days. Blood samples, taken from Group B subjects during the 1st and the 29th day of treatment at pre-established time points, were used to determine the concentration-time curves of omeprazole. The treatments were found to be safe and well tolerated by the horses. The serum hematological and biochemical values were within reference ranges for the entire observational time. No accumulation of the drug was found after 29 days of treatment. Lower C and AUCs were obtained at the 29th day of treatment.

This research focuses on the pharmacokinetics and tolerability of a newly formed gastro-resistant granule formulation of omeprazole in horses. It concludes that the formulation is safe and well tolerated by the horses, with no accumulation of the drug found after 29 days of treatment.
Objective of the Research
- The main objective of this research was to test a new formulation of omeprazole, commonly used for treating gastrointestinal issues, in horses to understand its pharmacokinetics and tolerability.
Research Design
- A total of 24 horses were used in this study.
- The horses were randomly divided into three groups, each comprising 8 horses.
- Group A was the control group, which was left untreated. Group B and Group C were the test groups, which received 4 mg/kg and 12 mg/kg of omeprazole, respectively. Both groups B and C were treated orally once a day for 90 days.
Procedure and Observations
- Blood samples were collected from the horses in Group B during the 1st and 29th day of treatment at pre-established time points.
- These samples helped ascertain the concentration-time curves of omeprazole.
- Findings established that the treatments were safe and well tolerated by the horses.
- All serum hematological and biochemical values remained within reference ranges throughout the entire observation period.
- There was no accumulation of the drug in the horses’ bodies found after 29 days of recurring treatment. By further calculating the total amount of drug underneath the concentration-time curve (AUC), it was reported to be lower on the 29th day of treatment.
Conclusion of the Research
- The research concluded that the new preparation of omeprazole in the form of gastro-resistant granules is safe for use in horses, considering their pharmacokinetic behavior and tolerability in the specimens used in this study.
- The study further revealed that the drug did not accumulate in the horse’s body over time, which implies minimal long-term side effects.
