Pharmacological characterization of adrenoceptors in horse corpus cavernosum penis.

Research Methodology

• The first part of the luminary trial revolved around an in vitro analysis of the presence and types of alpha and beta-adrenoceptors. The main site of focus was the corpus cavernosum of the horse, an area integral to erectile function. This was monitored and studied through the usage of select adrenoceptor ligands which bind to these adrenoceptors to stimulate or inhibit their activity, along with isometric tension recording.

Observations and Findings

• Noradrenaline and phenylephrine were observed to induce concentration-dependent contractions in corpus cavernosum preparations, implying the possible involvement of adrenoceptors in mediating these effects in the tissue.
• Phentolamine and prazosin, two alpha-adrenoceptor antagonists were noted to cause a shift rightward in the dose-response curve of noradrenaline, indicating their role in blocking the receptors. The alpha(2)-antagonist, rauwolscine, however, showed no effect on the noradrenaline response.
• Contractions initiated by Phenylephrine were significantly inhibited by the alpha(1)-adrenoceptor antagonist, prazosin, indicating the presence of alpha(1)-adrenoceptors in the horse corpus cavernosum.
• In the case of Isoprenaline and salbutamol, both were recorded to have relaxed the corpus cavernosum preparations in a concentration-dependent manner. The effect of the former was observed to be blocked by stand-alone beta-antagonists – propranolol, practolol, and butoxamine. However, salbutamol’s effect was only blocked by butoxamine.

Conclusion

• The results indicate the presence of postjunctional alpha(1)-adrenoceptors in horse corpus cavernosum. A diverse population of beta-adrenoceptors was also observed in this tissue, from the beta(1) and beta(2) subtypes.

Implications of the findings

• The findings shed light on the influence that adrenoceptors (particularly alpha(1)-adrenoceptors, and beta(1) and beta(2)-adrenoceptors) may play in regulating erectile function in horses, suggesting potential new avenues for research (e.g. development of drugs for erectile dysfunction) and diagnosis of reproductive disorders in horse populations.
