[Preparative isolation of alpha 2-macroglobulin, transferrin, albumin and study of their nonspecific gamma-inhibitory activity].
Abstract: Profiles of distribution of non-specific gamma-inhibitors of influenza A2/Victoria/35/72 in donkey and horse sera were established by gel chromatography in Sephadex G-200. High and low molecular inhibitors were found in 19S and 4S serum fractions. Highly purified preparations of a2-macroglobulin, transferrine and albumin were isolated by a combination of methods of salt precipitation, gel chromatography on Sephadex G-100, G-200 and ion exchange on DEAE-Sephadex A-50. Heating sera resulted in a considerable increase of the antiviral activity of a2-macroglobulin and transferrine and a reduction of albumin activity.

The research article is about the analysis of nonspecific gamma-inhibitors found in animal sera and their antiviral activity after heat processing, focusing on inhibitors such as a2-macroglobulin, transferrin, and albumin.
Distribution Profile of Gamma-Inhibitors
- The study first discusses the creation of distribution profiles for nonspecific gamma-inhibitors located within horse and donkey sera. Gamma-inhibitors are substances within the blood that can prevent or hamper replication of the influenza virus.
- Gel chromatography on Sephadex G-200, a method used for separating proteins based on their size, was used to segregate these inhibitors. The researchers found both high and low molecule inhibitors in 19S and 4S serum fractions respectively.
Isolation of a2-macroglobulin, transferrin, and albumin
- The researchers further isolated specific kinds of inhibitors namely alpha 2-macroglobulin, transferrin, and albumin. The isolation process involved a combination of methods which included techniques such as salt precipitation and gel chromatography on Sephadex G-100 and G-200.
- Additionally, they also used ion exchange on DEAE-Sephadex A-50, a method that separates proteins based on their charge, to achieve highly purified preparations of the inhibitors.
Impact of Heating on Inhibitor Activities
- The team conducted a study on how heat affected the antiviral activities of these inhibitors. They observed that heating the sera (a portion of the blood that includes all proteins not used in blood clotting) led to a significant increase in the antiviral activity for a2-macroglobulin and transferrin.
- However, there was a notable decrease in the corresponding activity for albumin post heat treatment.
