Recombinant horse interleukin-4 and interleukin-10 induced a mixed inflammatory cytokine response in horse peripheral blood mononuclear cells.

Abstract: Interleukin (IL)-4 and IL-10 activate plethora of immune cells and induce the humoral immune response. However, recombinant version of horse IL-4 and IL-10 has not been investigated to understand their immunomodulating activities. This study aimed to produce recombinant horse mature IL-4 and IL-10 in . Immune-modulating activities of recombinant horse IL-4 and IL-10 were investigated in peripheral blood mononuclear cells (PBMCs). Methods: Equine PBMCs were stimulated with recombinant IL-4 and IL-10. A proliferation of PBMCs was measured by XTT assay and cytokines induction was measured by enzyme-linked immunosorbent assay and real-time polymerase chain reaction. Results: Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis displayed a molecular weight of 15 kDa for IL-4 and 19 kDa for IL-10. Recombinant IL-4 and IL-10 significantly induced cell proliferation at 250 ng/ml. The results demonstrated that IL-4 enhanced expression of interferon-gamma (IFN-γ), IL-6, tumor necrosis factor-alpha (TNF-α), and IL-10, while recombinant horse IL-10 induced expression of IL-6, IFN-γ, and TNF-α. Conclusions: The present study demonstrated that biologically active horse IL-4 and IL-10 could be produced in .

Summary

This research summary has been generated with artificial intelligence and may contain errors and omissions. Refer to the original study to confirm details provided. Submit correction.

This research focuses on the production and examination of the immune-modulating activities of lab-produced horse interleukin-4 and interleukin-10 (IL-4 and IL-10), two types of immune cells. The researchers found that these cells caused a mixed inflammatory cytokine response in peripheral blood mononuclear cells, indicating their potential use in immunomodulatory therapy.

Methods

  • The study started with the production of recombinant horse IL-4 and IL-10.
  • These cells were then introduced to peripheral blood mononuclear cells (PBMCs), a type of immune cell found in the bloodstream,
  • After stimulation of the PBMCs with these recombinant IL-4 and IL-10, a few measurements were taken:
  • A proliferation (increase in number) of PBMCs was noted using an XTT assay, a test used to measure cell viability and proliferation.
  • The induction of cytokines, small proteins that are crucial in cell signaling during immune responses, was measured using an enzyme-linked immunosorbent assay (ELISA) and real-time polymerase chain reaction (PCR).

Results

  • After analyzing the samples using sodium dodecyl sulfate-polyacrylamide gel electrophoresis, the researchers found that the molecular weights of IL-4 and IL-10 were 15 and 19 kDa respectively.
  • More importantly, the researchers discovered that both the lab-produced IL-4 and IL-10 significantly induced cell proliferation at a concentration of 250 ng/ml.
  • On a more specific level, IL-4 was found to increase the expression of interferon-gamma (IFN-γ), IL-6, tumor necrosis factor-alpha (TNF-α), and IL-10. Meanwhile, the lab-produced horse IL-10 induced expression of IL-6, IFN-γ, and TNF-α.

Conclusion

  • The research successfully showed that biologically active horse IL-4 and IL-10 could be produced in a lab setting and are able to stimulate immune reactions in horse peripheral blood mononuclear cells.
  • This finding may have important implications for the development of immunomodulatory medicine, specifically for horses. However, further research is needed to ascertain the potential therapeutic uses and understand any possible side effects.
