Spi2 gene polymorphism is not associated with recurrent airway obstruction and inflammatory airway disease in thoroughbred horses.

Abstract: The aim was to detect the presence of polymorphisms at exons 1, 2, 3 and 4 of the Spi2 gene, and evaluate a possible association between them and recurrent airway obstruction (RAO) or inflammatory airway disease (IAD) in thoroughbred horses, through single-strand conformational-polymorphism (SSCP) screening. Although polymorphism was not detected in exons 1, 2 and 3, three alleles and six genotypes were identified in exon 4. The frequencies of allele A (0.6388) and genotype AA (0.3888) were higher in horses affected by RAO, although no association was found between polymorphism and horses with either RAO or IAD.

The research examined the potential relationship between variations in the Spi2 gene and chronic respiratory conditions in thoroughbred horses, but ultimately found no such association.

Objective and Methodology

  • The goal of the research was to identify any polymorphisms (variations) in the Spi2 gene, specifically in exons 1, 2, 3 and 4, and to study any links between these variations and recurrent airway obstruction (RAO) or inflammatory airway disease (IAD) in thoroughbreds.
  • To accomplish their objective, the researchers used single-strand conformational-polymorphism screening, a technique that helps detect genetic variation.

Findings and Analysis

  • The screening did not reveal any polymorphisms in exons 1, 2 and 3 of the Spi2 gene.
  • However, within exon 4 of the same gene, three different alleles (versions of a gene) and six genotypes (specific genetic configurations) were discovered.
  • The researchers observed that the frequencies of the allele A (0.6388) and the genotype AA (0.3888) were higher in horses suffering from RAO.

Conclusion

  • Despite some variations in genotype and allele frequencies, the study did not establish any statistically significant association between the Spi2 gene polymorphisms and recurrent airway obstruction or inflammatory airway disease in thoroughbred horses.
  • This suggests that other factors beyond Spi2 gene variation are contributing to these respiratory conditions in the horse population, pointing to the complexity of the genetic and environmental interplay in these diseases.
