[The effect of a single dose of dexamethasone on the production of superoxide anion and on the chemotaxis of blood polynuclear neutrophils in the horse].
Abstract: Chemotaxis and superoxide anion production of neutrophils in healthy horses were investigated before and 8 h after, a single injection of dexamethasone at a dose of 0.045 mg/kg. Chemotaxis was studied by the technique of migration under agarose and superoxyde production was measured by ferricytochrome c reduction. Superoxide anion production was not changed, but the chemotactic index, with zymosan activated horse serum, was increased. The FMLP chemoattractant effect, at 10(-4) M, was slightly enhanced.

The research paper investigates the effect of a single dexamethasone dose on the neutrophil’s chemotaxis and superoxide anion production in healthy horses.
Objective of research
- The major aim of this research is to understand how a single dose of dexamethasone, a type of corticosteroid medication, impacts the chemotactic function and superoxide anion production of polynuclear neutrophils in horses. The study was conducted on healthy horses and measurements were taken before and 8 hours following the dexamethasone injection.
Methods and techniques
- The test dose of dexamethasone was set at 0.045 mg/kg, which was administered to the horses. The resulting effects on neutrophils were recorded after eight hours of injection.
- Leukocytes’ chemotaxis was studied by the technique of migration under agarose. This technique is often used in cell biology to study the movement and behavior of cells.
- The production of superoxide anion molecules (a type of reactive oxygen species involved in the immune response) was determined by analyzing the reduction of ferricytochrome c. This is a method that effectively gauges the rate of superoxide production.
Findings and conclusions of the study
- Superoxide anion production remained unchanged post-dexamethasone injection.
- The chemotactic index, indicating the responsiveness of cells to chemical attractants, showed an increase when tested with zymosan activated horse serum. This suggests heightened chemotactic activity in response to dexamethasone injection.
- The effects of the chemoattractant formyl-methionyl-leucyl-phenylalanine (FMLP) were slightly enhanced at a 10(-4) M concentration following the corticosteroid intervention.
- Overall, the study indicates that a single dose of dexamethasone impacts the chemotactic activity, but not the superoxide anion production, of blood polynuclear neutrophils in a horse. The implications of this discovery require further research to ascertain any potential therapeutic or adverse effects related to the immunomodulatory properties of dexamethasone.
