Evaluation of apoptosis markers in different cell lines infected with equine arteritis virus.
Biotechnic & histochemistry : official publication of the Biological Stain Commission    October 23, 2018   Volume 94, Issue 2 115-125 doi: 10.1080/10520295.2018.1521989
Metz GE, Abeyá MM, Serena MS, Panei CJ, Echeverría MG.Equine arteritis virus (EAV) induces apoptosis in infected cells. Cell death caused by EAV has been studied mainly using three cell lines, BHK-21, RK-13 and Vero cells. The mechanism of apoptosis varies among cell lines and results cannot be correlated owing to differences in EAV strains used. We evaluated different markers for apoptosis in BHK-21, RK-13 and Vero cell lines using the Bucyrus EAV reference strain. Acridine orange/ethidium bromide staining revealed morphological changes in infected cells, while flow cytometry indicated the extent of apoptosis. We also observed DNA fragmentation,...
A comparative study of oculomotor, trochlear and abducens nerves in Arabian foals.
Biotechnic & histochemistry : official publication of the Biological Stain Commission    March 1, 2017   Volume 92, Issue 2 149-156 doi: 10.1080/10520295.2017.1288926
Bolat D, Yıldız D, Bahar S, Yürüker S, Kaymaz F, Ilgın C, Bozkurt EÜ, Karahan S, Sabancı SS.We investigated the microscopic structure of transverse sections of the oculomotor, trochlear and abducens nerves of Arabian foals using stereological methods. Bilateral nerve pairs from 2-month-old female Arabian foals were analyzed. The tissues were embedded in plastic blocks, then 1 µm thick sections were cut and stained with osmium tetroxide and methylene blue-azure II. Stereology was performed using light microscopy. Morphometry showed that the right and left pairs of nerves were similar. The transverse sectional areas of the oculomotor, trochlear and abducens nerves were 1.93 ± 0.19 mm...
Age and gender related changes in hematological parameters of thoroughbred foals.
Biotechnic & histochemistry : official publication of the Biological Stain Commission    May 7, 2013   Volume 88, Issue 6 345-349 doi: 10.3109/10520295.2013.788213
Uluisik D, Keskin E, Ozaydın T.Hematological and biochemical profiles commonly are required in equine medicine. We studied hematological parameters including red blood cells (RBC), white blood cells (WBC), hemoglobin (Hb), hematocrit (PCV), differential leukocyte counts, mean cell volume (MCV), mean corpuscular hemoglobin (MCH), and mean corpuscular hemoglobin concentration (MCHC) in thoroughbred foals at different ages and for both sexes. Sixty healthy thoroughbred foals, 1 day, 3 days and 1 year old were used. Each age group consisted of 10 male and 10 female animals. We found significant differences related to age in RBC...
Cytochemistry of peripheral blood leukocytes in thoroughbred foals.
Biotechnic & histochemistry : official publication of the Biological Stain Commission    April 25, 2013   Volume 88, Issue 6 295-301 doi: 10.3109/10520295.2013.782067
Ozaydın T, Celik I, Sur E, Oznurlu Y, Uluısık D.We attempted to characterize the cytochemical staining patterns of leukocytes and to determine the percentages of alpha-naphthyl acetate esterase (ANAE) and acid phosphatase (ACPase) positive lymphocytes in peripheral blood of thoroughbred foals at different ages. Blood samples were obtained from the jugular veins of 60 healthy thoroughbred foals, 1 day, 3 days and 1 year old. Each age group included 10 male and 10 female animals. Peroxidase (PO) activity was detected in neutrophils, eosinophils and monocytes. Lymphocytes were negative for PO staining. Periodic acid-Schiff (PAS) staining was o...
Viability and acrosome staining of stallion spermatozoa by Chicago sky blue and Giemsa.
Biotechnic & histochemistry : official publication of the Biological Stain Commission    November 30, 2006   Volume 81, Issue 4-6 109-117 doi: 10.1080/10520290600931007
Kútvölgyi G, Stefler J, Kovács A.A simple trypan blue-neutral red-Giemsa staining procedure for simultaneous evaluation of acrosome, sperm head, and tail membrane integrity and morphology has been used to evaluate equine spermatozoa. Some special characteristics and problems have arisen in evaluating stallion semen. One problem was the differentiation of intact vs. damaged sperm tails primarily in frozen and thawed samples. After freezing and thawing, a high percentage of spermatozoa with an unstained head and stained tail were observed. These cells are considered immotile. Therefore, unambiguous differentiation of intact vs....
Detection of DNA in ancient bones using histochemical methods.
Biotechnic & histochemistry : official publication of the Biological Stain Commission    August 19, 2000   Volume 75, Issue 3 110-117 doi: 10.3109/10520290009066488
Guarino FM, Angelini F, Odierna G, Bianco MR, Di Bernardo G, Forte A, Cascino A, Cipollaro M.We describe histochemical techniques for detecting DNA within the osteocytic lacunae of ancient bones. The bones examined were fragments of femurs from two human individuals found in the Pompeian C. I. Polybius house and fragments of metacarpals from two horses (Equus sp.) found in the Pompeian "Casti Amanti" house. Both buildings were buried by the 79 A. D. Vesuvius eruption. Fragments of femurs from a modern horse, a modern swine and a modern amphibian also were studied as controls. Some bone sections were stained with two different DNA-specific fluorochromes, 4'-'6-diamidino-2-phenylindole ...
