Follicle profile and plasma gonadotropin concentration in pubertal female ponies. Twelve female ponies were examined daily for 30 days and classified as ovulating (OV; N = 6; 197 +/- 6 kg) or prepubertal (PP; N = 6; 196 +/- 9 kg). Follicles were detected by ultrasound and gonadotropins quantified by radioimmunoassay. The mean diameter of the largest follicles was significantly larger in OV (38 +/- 1 mm) than in PP (26 +/- 2 mm) but there was no difference between groups in the size of the second largest follicle. There were more small follicles (29 mm than the OV fillies. Follicle-stimulating hormone (FSH) levels did not differ between groups but PP fillies had lower lutein...
The erythrocyte cytoskeleton protein 4.2 is not demonstrable in several mammalian species. Erythrocyte membrane proteins from 44 representative mammals were studied. Protein 4.2 was not detected in guinea pigs (Cavia porcellus) (N = 14), Southern Brazilian swamp large rats (Myocastor coypus) (N = 2), cutias (Dasyprocta sp) (N = 4), and horses (Equus caballus) (N = 13). These animals also presented high ankyrin concentrations except for the horse which did not exhibit a sharp band, although minor components located between proteins 2 and 3 could account for the ankyrin family. The rodents studied did present band 6, which was not detectable in other common rodents such as white rats ...
Diversity of genomic electropherotypes of naturally occurring equine herpesvirus 1 isolates in Argentina. The genomes of 10 equine herpesvirus 1 (EHV-1) strains isolated in Argentina from 1979 to 1991, and a Japanese HH1 reference strain were compared by restriction endonuclease analysis. Two restriction enzymes, BamHI and BglII, were used and analysis of the electropherotypes did not show significant differences among isolates obtained from horses with different clinical signs. This suggests that the EHV-1 isolates studied, which circulated in Argentina for more than 10 years, belong to a single genotype.
Is the thoroughbred race-horse under chronic stress? Thoroughbred fillies were divided into three groups according to age: group 1, 7 fillies aged 1 to 2 years (G1) starting the training program; group 2, 9 fillies aged 2 to 3 years (G2) in a full training program; group 3, 8 older fillies 3 to 4 years of age (G3) training and racing. Blood samples were collected weekly from July to December. Cortisol was quantified using a solid phase DPC kit. The intra- and interassay coefficients of variation were 12.5% and 15.65% and sensitivity was 1.9 +/- 0.2 nmol/l. The semester average of cortisol levels varied between groups: G1 = 148.8 +/- 6.7, G2 = 12...
Abortion due to equine herpesvirus in southern Brazil. We report an outbreak of abortion due to equine herpesvirus (EHV) in 5 mares between 9 and 11 months of gestation, from a herd of 22 Thoroughbred mares. Equine herpesvirus was isolated from extracts of the liver, spleen and thymus but not from the lungs of a 9-month fetus grown in Rabbit Kidney (RK13) cells. The virus was identified by electron microscopy, where virus particles could be seen in the nucleus of infected cells, and by the fluorescent antibody technique with polyclonal antibodies against the whole virus. Anamnesis, necropsy, histopathology, bacteriology, and virology data suggest ...
Isolation of horse IgG with protein A. Horse immunoglobulins were obtained from normal serum defatted with dextran sulfate and precipitated with ammonium sulfate. Eight mg of this preparation was submitted to affinity chromatography with protein A-Sepharose CL-4B. Low temperature (4 degrees C) and a starting buffer at pH 8.0 were conditions required for all IgG subclasses to bind to protein A, even those with low affinity. The IgGs bound to protein A were eluted with glycine buffer at pH 2.8. The yield was about 90%. It is suggested that isolated IgG, instead of whole Igs, be used in serum therapy, reducing the amount of Igs and di...
Immunization of horses with Crotalus durissus terrificus (South American rattlesnake) venom. A comparison of four different procedures. 1. A comparative study was carried out on horses immunized with Crotalus durissus terrificus venom using four different inoculation procedures, which included the use of Freund's adjuvant, A1(OH)3 and liposomes as adjuvants. The antibody titer was assessed by enzyme linked immunosorbent assay (ELISA) and the neutralizing potency by the neutralizing median effective dose (ED50). 2. The inoculation schedule used in horses to obtain antivenom serum consisted of sc injections of a 7.5 mg venom starting dose in 5.0 ml sterile saline emulsified with an equal volume of Freund's complete adjuvant. One...
Immunization of equines with phospholipase A2 protects against the lethal effects of Crotalus durissus terrificus venom. Equines (2 horses and 2 donkeys) immunized with whole Crotalus durissus terrificus venom or its phospholipase A2 component either presented an increased survival time determined 3 days after challenge or were totally resistant to a challenging lethal dose of 200 mg crude venom 270 days after the initial immunization or 90 days after the last booster injection. The resistance was demonstrable on the basis of a good correlation with antibody titers determined by the ELISA method but not with the flocculation and neutralization assays. Since phospholipase A2 is essentially nontoxic, it can be use...
