1H NMR investigation of normal and osteo-arthritic synovial fluid in the horse.

Abstract: Proton magnetic resonance spectroscopy (1H NMR) has been successfully used in the study of many biological fluids. The data presented here report on the metabolic profiles of normal equine synovial fluids compared with osteoarthritic (OA) fluids. Twenty-five OA synovial fluid samples and eight normal ones were collected from the forelimb fetlock joint in 22 horses, aged between five and 24 years. 1H NMR spectroscopy was carried out with a Bruker Avance DRX 500 equiped with a cryo-magnet working at 11 Tesla, and 'Mestre-C 4.9.9.6' software was used to analyze the spectra. The study assessed the increase of lactate, alanine, acetate, N-acetylglucosamine, pyruvate, citrate, creatine/creatinine, glycerol, HDL choline, and a-glucose in OA synovial fluid. The variations observed in samples from horses with OA compared to those in the control group, and similar data found in other studies, confirm that this technique may be useful in the study of joint metabolism. Its practical application may be in the evaluation of the treatment of OA in athletic horses.

The research article explores the use of Proton magnetic resonance spectroscopy (1H NMR) to study the metabolic profiles of normal horse synovial fluids in comparison to fluids from horses suffering from Osteoarthritis (OA). The findings from the study confirm that this technique could be useful in studying joint metabolism and evaluating OA treatment in athletic horses.

Introduction of the Research

  • The researchers used proton magnetic resonance spectroscopy (1H NMR), a popular method for studying biological fluids, to examine the metabolic profiles of synovial fluid in horses. Synovial fluid is crucial in lubricating and providing nutrients to the joints.
  • The focus was on comparing the synovial fluid of healthy horses with that from horses suffering from Osteoarthritis (OA), a common joint disorder in horses.

Methodology of the Research

  • The researchers collected 25 OA synovial fluid samples and 8 normal samples from the forelimb fetlock joint of 22 horses, aged between 5 and 24 years.
  • A Bruker Avance DRX 500 equipped with a cryo-magnet was used along with ‘Mestre-C 4.9.9.6’ software for 1H NMR spectroscopy. This setup allowed the researchers to meticulously analyze the metabolic content of the fluids.

Findings of the Research

  • The study identified increased levels of lactate, alanine, acetate, N-acetylglucosamine, pyruvate, citrate, creatine/creatinine, glycerol, HDL choline, and a-glucose in the OA synovial fluid compared to normal joint fluid.
  • This finding, in combination with similar results discovered in other research, confirms the effective application of 1H NMR spectroscopy in examining joint metabolism and the characteristics of OA.

Implications of the Research

  • The metabolic variations found in the synovial fluid of OA horses compared to healthy ones might help in diagnosing OA in horses earlier and more accurately.
  • The practical application of the technique may lay in evaluating the effect of treatments for OA in performance horses, which could potentially lead to improved management and rehabilitation strategies.
