Characterisation of glycoproteins in the sweat of the horse (Equus caballus).
Abstract: The two major polypeptides H (Mr 49,000) and L (Mr 33,000) of equine sweat have been purified by gel filtration and characterised by gel electrophoresis and compositional analysis. Both H and L are glycoproteins containing sialic acid, neutral sugars, N-acetylglucosamine and N-acetylgalactosamine, but the two polypeptides differ considerably in the extent of glycosylation. H and L also differ in amino acid composition, but both contain only low levels of sulphur containing amino acids and histidine. These glycoproteins may behave as surfactants.

This research focused on the analysis of two major glycoproteins, named H and L, present in the sweat of horses. The team found that these proteins have unique structures and might function as surfactants in the equine body.
Characterising H and L Polypeptides
- The study concentrated on major polypeptides, namely H and L, found in horse sweat. Polypeptides are chains of amino acids, and they are critical components of proteins.
- The researchers used a two-stage process to isolate and purify these polypeptides, including gel filtration (a method used to separate substances based on their size) and gel electrophoresis (a technique for separating proteins based on their size and charge).
- These identified polypeptides were then thoroughly characterised through compositional analysis, which revealed details about the polypeptides’ makeup.
Glycoproteins Composition
- Both H and L were unveiled as glycoproteins. Glycoproteins are proteins that have carbohydrates (or sugars) attached to them. This protein-carbohydrate combo plays crucial roles in various biological functions such as immune response or cell-cell interaction.
- They both contain sialic acid, neutral sugars, N-acetylglucosamine and N-acetylgalactosamine, which are all various types of sugars often found in glycoproteins.
- However, there is a significant difference in the extent of glycosylation, the process of adding carbohydrates to a protein, between the two polypeptides. This suggests that, while they are both glycoproteins, they have notably different structures.
Amino Acid Composition
- H and L also differ when analyzed for their amino acid composition. Amino acids are the building blocks of proteins and are essential for a myriad of biological processes within the body.
- Though they contain only low levels of sulfur-containing amino acids and histidine. Sulfur-based amino acids are essential for a range of bodily functions, including the synthesis of certain proteins, while histidine is necessary for growth and tissue repair.
Potential Role as Surfactants
- The researchers hypothesize that these glycoproteins may behave as surfactants. Surfactants are compounds that lower the surface tension between two substances, generally aiding in mixtures or emulsions.
