Data set of the proteome of fresh and frozen thawed stallion spermatozoa.

Abstract: This paper provides the dataset of proteins of stallion ejaculates before and after cryopreservation. The data report the analysis and identification of stallion sperm proteins obtained from the same ejaculates and split in two subsamples. The first aliquot consisted on fresh spermatozoa and the second aliquot was frozen and thawed spermatozoa. Samples were analyzed using a UHPLC/MS/MS system consisting of an Agilent 1290 infinity series UHPLC coupled to an Agilent 6550 Q-TOF mass spectrometer (Agilent Technologies, Santa Clara, CA, USA). We provide a data set of 2226 different proteins, with 2180 aligned to the equine proteome database. The data can be used to identify potential targets to be explored to improve techniques for cryopreservation of spermatozoa. This data article refers to the article "Proteomic profiling of stallion spermatozoa suggests changes in sperm metabolism and compromised redox regulation after cryopreservation" (Martín Cano et al.; 2020) [1].

The study presents a comprehensive dataset of proteins found in stallion sperm before and after the process of freezing and thawing, carried out to explore ways of improving sperm cryopreservation.

Objective of Research

  • The main aim of this research was to meticulously analyze and identify the proteins present in fresh and frozen-thawed stallion spermatozoa with the purpose of enriching current knowledge on cryopreservation techniques.

Methodology

  • The researchers procured the sperm samples from the same ejaculate, divided it into two subsamples – one portion contained fresh spermatozoa while the other portion underwent freezing and thawing.
  • These samples were then subjected to analysis using a sophisticated Ultra High Performance Liquid Chromatography (UHPLC) coupled with Quadrupole Time of Flight (Q-TOF) mass spectrometer – a combination that is capable of conducting high-resolution protein analysis.

Findings

  • The article reveals the discovery of a catalog of 2226 distinguishable proteins in stallion spermatozoa, where 2180 of these proteins aligned with the equine proteome database.
  • This massive data is a rich resource that can be mined for potential targets to boost sperm cryopreservation methods and techniques.

Associated Work

  • The data in this article is connected to another research paper titled “Proteomic profiling of stallion spermatozoa suggests changes in sperm metabolism and compromised redox regulation after cryopreservation” (Martín Cano et al.; 2020), which offers additional insights on changes in sperm structure post-cryopreservation.
