Immunohistochemical Expression of Collagens in the Skin of Horses Treated with Leukocyte-Poor Platelet-Rich Plasma.

Abstract: This study evaluated the immunohistochemical expression of type I (COL I) and III (COL III) collagens during the healing process of skin treated with leukocyte-poor platelet-rich plasma (LP-PRP). Seven healthy gelding crossbred horses aged 16 to 17 years were used. Two rectangle-shaped wounds were created surgically in the right and left gluteal regions. Twelve hours after wound induction, 0.5 mL of the LP-PRP was administered in each edge of the wounds of one of the gluteal regions. The contralateral region was used as control (CG). Three samples were obtained: after wound induction (T0), 14 days (T1) of healing process, and after complete closure of the skin (T2). The normal skin (T0) showed strong staining for type III and I collagen in papillary and reticular dermis, respectively. In the scar of the treated group, COL III showed important (p < 0.05) increase in immunoreaction in T2 compared with T1. The administration of a single dose of LP-PRP 12 h after induction of wound in horses does not influence formation of collagens I and III. However, the intense labeling for COL III suggests that the tissue was still weak during the macroscopic closure of the wound, demonstrating that healing was not completely finished.

Summary

This research summary has been generated with artificial intelligence and may contain errors and omissions. Refer to the original study to confirm details provided. Submit correction.

The research article reports on the effects of using leukocyte-poor platelet-rich plasma (LP-PRP) to aid in the wound healing process of horses, particularly by examining the development of type I and III collagens.

Objective of the Study

  • This analysis investigates the role played by type I (COL I) and type III (COL III) collagens in wound healing in horses. The researchers also look at how these collagens are impacted by the use of leukocyte-poor platelet-rich plasma (LP-PRP) in the process.

Design and Execution of the Study

  • Seven healthy gelding crossbred horses, ages 16 to 17 years, formed the test group. Surgical rectangle-shaped wounds were inflicted on the right and left gluteal regions of each animal.
  • The LP-PRP was administrated on one of the wounds 12 hours after it was induced, while the other wound was kept as a control (CG). The researchers tested three samples: immediately after the wounds were created (T0), 14 days into the healing process (T1) and following complete skin closure (T2).

Results of the Study

  • The study findings suggest that the LP-PRP does not affect the production of collagen types I and III during the healing process. The normal skin (T0) exhibited significant staining for type III and I collagen in the papillary and reticular dermis, respectively.
  • Interestingly, for the treated group, there was a notable increase in COL III immunoreaction in T2 as compared to T1, pointing towards that the tissue was still weak during the macroscopic wound closure. This might suggest that healing was not fully achieved at the macroscopic level at the time of wound closure, implying a delayed or extended healing period.

Conclusions of the Study

  • The concluded that the administration of LP-PRP does not necessarily accelerate the production of COL I and COL III collagen during the wound healing process.
  • However, the researchers suggest that further studies are required to ascertain whether the weak tissue observed during macroscopic wound closure is a normal incident or are there other factors at play.
