Pharmacokinetics of ketoprofen in the donkey (Equus asinus).
Abstract: The pharmacokinetic parameters of ketoprofen were determined in four donkeys after a single intravenous injection of a dose of 2.2 mg/kg body weight. The total body clearance (ClB) was 414.0 +/- 98.70 ml/h/kg (mean +/- SD), the volume of distribution at steady state (Vss) 263.10 +/- 55.43 ml/kg and the elimination half-life 1.30 +/- 0.75 h. These values were compared to those obtained in horses.

The research article discusses the effect of the drug Ketoprofen on donkeys after a single dosage was given intravenously. The study measures the body’s processing of the drug—how it’s distributed and eliminated.
Research Objectives and Methodology
- The purpose of this research was to investigate how the drug Ketoprofen behaves in the body of donkeys after being administered a single dose intravenously. The researchers aimed to provide values for the total body clearance, the volume of distribution in a stable state and the elimination half-life of the drug.
- Four donkeys were used in the experiment where each was given an intravenous injection with a 2.2 mg/kg body weight dose of Ketoprofen. The researchers then monitored and recorded the pharmacokinetic parameters of the drug.
Findings
- The total body clearance, which measures the body’s ability to remove the drug, was found to be 414.0 +/- 98.70 ml/h/kg. This value implies how quickly the drug disappears from the body after administration.
- The volume of distribution at steady state was measured to be 263.10 +/- 55.43 ml/kg. This value illustrates the extent to which the drug has spread out within the body.
- The elimination half-life of Ketoprofen in donkeys was found to be 1.30 +/- 0.75 hours. This value represents the time it takes for the drug concentration in the body to be reduced by half.
- The research compared these values with similar studies conducted in horses. Though the results on the comparison have not been explicitly stated in the abstract, this comparison is significant in veterinary pharmacology, potentially guiding dosage adjustments across different equine species.
Implications
- This study provides crucial information in understanding how ketoprofen behaves in the body of donkeys. As a non-steroidal anti-inflammatory drug (NSAID), ketoprofen is commonly used in veterinary medicine for pain relief. Thus, understanding its pharmacokinetics can aid in optimizing dosing schedules and predicting potential side effects.
- The findings also offer a basis for comparison with how the drug behaves in the bodies of other equine species like horses. This comparison can help determine if dosages need adjustment when prescribing Ketoprofen for different species within the equine family.
