Rapid detection of alveolar echinococcosis in hepatic nodules of horses by recombinase polymerase amplification assay.

Abstract: Alveolar echinococcosis in slaughtered horses remains a public health issue. This study aimed to develop a Recombinase polymerase amplification (RPA) assay targeting the mitochondrial NADH dehydrogenase subunit 5 () gene of for the rapid detection of equine alveolar echinococcosis. Thirty-six hepatic solid nodules obtained from each horse ( = 36) were evaluated based on histopathological examination and -targeted PCR and then submitted to the RPA assay. The results of the developed RPA assay were 94.4% consistent with those of PCR and Cohen's kappa coefficient value was 0.89 statistically, indicating high agreement. In addition, the RPA assay using the plasmid samples was one hundredfold more sensitive than PCR testing. Consequently, these results suggest that the performance of the RPA assay developed in this study is equal to that of conventional PCR testing.

This research article is about developing a rapid detection method for alveolar echinococcosis in horses using a Recombinase Polymerase Amplification (RPA) assay, which showed high accuracy and greater sensitivity than traditional PCR testing.

Objective of the Study

  • The aim of this research is to establish an RPA assay that targets the mitochondrial NADH dehydrogenase subunit 5 gene of Echinococcus multilocularis, the parasite that causes alveolar echinococcosis in horses. The study is rooted in the continuing public health issue of alveolar echinococcosis found in slaughtered horses.

Methodology

  • 36 hepatic nodules from different horses were collected for the study.
  • All the samples underwent histopathological examination and gene-targeted PCR testing.
  • After these preliminary examinations, the samples were then submitted to the newly developed RPA assay.

Results

  • The results found from the RPA assay matched 94.4% of the time with the PCR test results, indicating high concordance between these two methods.
  • The statistical agreement between the two tests was high with a Cohen’s kappa coefficient value of 0.89.
  • The RPA assay also demonstrated a sensitivity that was 100 times greater than the sensitivity of the PCR testing when the plasmid samples were used.

Conclusion

  • The study concludes that this newly developed RPA assay has a performance equivalent to that of the conventional PCR tests. However, its sensitivity appears to be significantly higher.
  • Suggesting that this new RPA-based method could be an effective diagnostic tool for rapidly detecting alveolar echinococcosis in horses.
