Seasonal functional relevance of sperm characteristics in equine spermatozoa.

Abstract: A group of stallions with different reproductive indexes were used to study seasonal variations in sperm quality (Equus caballus). Semen samples were collected from late September to July and analyzed according to four seasonal periods: late September-December, January-March, late March-May, and June-July. Parameters monitored included sperm concentration, sperm motility, sperm morphology, sperm viability, acrosomal status, plasma membrane stability, and sperm mitochondrial membrane potential. Overall, seminal parameters monitored are affected mostly by time period, followed by animal and lastly by fertility, stressing the importance of individual variations in out-bred animal models. The analysis of multiple ejaculates from the same animals showed clear seasonal-based differences (P<0.05) with poor semen quality in winter and a noticeable improvement in sperm quality with increasing photoperiod. Better semen quality was observed between late March and May. Interactions between month period, animal, and fertility were evident (P<0.05) for sperm concentration, head and tail sperm anomalies, and acrosomal integrity. Thus, it may be advisable to adjust the use of stallion semen according to seasonal variations.

This research explores how the quality of stallion sperm changes with the seasons and how these variations in sperm concentration, motility, morphology, and other parameters can potentially impact fertility. The study’s findings underline the importance of considering these seasonal changes when using stallion semen, particularly in breeding programs.

Research Methodology

  • The researchers conducted the study on a group of male horses (stallions) with different reproductive indexes to understand how the season could affect the quality of their sperm.
  • The stallions’ semen samples were collected from late September to July. The collection periods were divided into four season-based categories: late September-December, January-March, late March-May, and June-July.

Sperm Analysis Parameters

  • The parameters of the sperm that were monitored included concentration, motility (movement abilities), morphology (structural features), viability (their capability to live and reproduce), acrosomal status (condition of the sperm’s acrosome- a cap-like structure necessary for fertilization), plasma membrane stability, and sperm mitochondrial membrane potential.

Key Findings

  • Some major findings of the study were that these parameters are chiefly affected by the time period or the season. Variations between individual animals (stallions) were secondary causes for differences in sperm parameters, while differences in fertility parameters were the least significant factor causing variations.
  • The study found clear seasonal variations as there was poor semen quality during the winter and an improvement in sperm quality was observed with increasing photoperiod (the length of the day).
  • Increased sperm quality was particularly noticeable between late March and May. The changes in sperm concentration, head and tail sperm anomalies, and acrosomal integrity were evidently influenced by the interaction of month period, individual animal variation, and fertility.

Implications of the Study

  • From these findings, it’s suggested that if stallion semen is being used, particularly in breeding programs, the seasonal changes should be accounted for to optimize the results and ensure the highest possible quality of the semen.
