Seminal freezing in pure breed andalusian horse: difference in individual stallions and correlation between pre and post-freezing sperm parameters.
Abstract: The aim of this study was the optimization of the sperm freezing protocols for the Pure Breed Andalusian Horse (AH) stallions. The study was performed in 84 ejaculates from 14 stallions (6 ejaculates per stallion). We examined the effect of individual stallion, centrifugal force and centrifugation extender on post-thaw sperm quality. Neither centrifugal force nor centrifugal extender had any significant effect on post-centrifugation or post-thawing sperm quality. Stallion was the principal source of variation in our experiments, showing individual significant differences (p < 0.05) in all parameters. Individual differences were more extreme prior to freezing than after freezing-thawing. There are significant positive correlations (p < 0.05 and p < 0.01) between all post-centrifugation and post-thawing parameters.

This research study aims to optimize sperm freezing protocols for Pure Breed Andalusian Horse stallions by examining the effect of individual stallion, centrifugal force, and centrifugation extender on post-thaw sperm quality. While no significant effects are found for centrifugal force or extender, the individual stallion differences are significant.
Background of the Study
- The study was designed with the aim of optimizing the protocols for the freezing of sperm in Pure Breed Andalusian Horses (AH).
- The emphasis on this particular breed of horse could imply its genetic value and the need to preserve its genetic line through artificial insemination.
- The investigators did the study with 84 ejaculates from 14 AH stallions, with each stallion contributing 6 ejaculates to the study.
Methodology
- The research examined the effect of three parameters on the quality of thawed sperm: the individual stallion, the centrifugal force applied during the freezing process, and the type of extender used in the process.
- Centrifugal force refers to the force applied to the sperm during the freezing process to separate it into layers based on density, with denser components moving outward. An extender is a solution used to maximize the volume of diluted semen for freezing.
Findings
- The research found that neither the centrifugal force nor the type of centrifugal extender used had any significant effect on the quality of sperm post-centrifugation or post-thawing.
- However, the individual stallion appeared to play a significant role in the post-thawing quality of the sperm, indicating that genetic factors of individual stallions can impact the outcomes of the freezing and thawing process.
- These individual differences were more extreme before freezing than after the freezing-thawing process.
Conclusions
- The researchers highlighted that there were significant positive correlations between all post-centrifugation and post-thawing parameters.
- This finding suggests that improving the freezing process could consequently improve the thawing results, boosting the overall success of sperm preservation.
- Therefore, future research in this field may focus on refining the technical aspects of the sperm freezing and thawing process, while taking into account the genetic variation among individual stallions.
