Sirius red is able to selectively stain eosinophil granulocytes in bovine, ovine and equine cervical tissue.

Abstract: The aim of the present study was to examine the suitability of sirius red staining for selective light microscopic demonstration of eosinophil granulocytes in cervical tissue of mares, cows and sheep. For this purpose, tissue was fixed in 4% neutral buffered formol or in Bouin's solution. Paraffin sections of 5-microm thickness were stained with sirius red. In cows, mares and sheep a selective distinction of eosinophilic infiltration is successful after both fixation methods.

This research examines the efficacy of sirius red staining for highlighting the presence of eosinophil granulocytes in cervical tissue of cows, sheep and horses.

Objective of the Study

  • The primary focus of this research was to investigate the effectiveness of sirius red as a stain for selectively demonstrating eosinophil granulocytes in the cervical tissue of cows, sheep, and mares (female horses) through light microscopic observation.

Methodology

  • The research was conducted on tissues that were fixed using two primary methods. The tissues were fixed either in a 4% neutral buffered formol or in Bouin’s solution.
  • Paraffin sections of the cervical tissue maintained at a thickness of about 5 micrometres were then stained using sirius red.

Findings

  • The results of the study showed that, irrespective of the fixation method employed (whether 4% neutral buffered formol or Bouin’s solution), sirius red staining successfully enabled a selective distinction of eosinophilic infiltration in the cervical tissue of cows, mares, and sheep.
  • This indicates that sirius red is effective in selectively staining eosinophil granulocytes in the cervical tissues of these three species, thereby providing a beneficial tool for future research studies relating to eosinophilic infiltration in such tissues.
