Studies on equine immunoglobulins. IV. Immunoglobulins of the donkey.
Abstract: Donkey IgGa was isolated in purified form from normal and immune donkey sera by column chromatography on DEAE-cellulose. Isolated donkey IgGa and mixtures of (IgGa+IgGb) were used as antigens to prepare rabbit reagents specific for equine IgGa or IgGb. Antibodies present in sera obtained from a single donkey at various times during the course of hyperimmunization with BSA were isolated by immuno-adsorption. The class or subclass of immunoglobulins present among isolated, donkey anti-BSA antibodies was determined by use of specific rabbit anti-equine immunoglobulin reagents. The homologues of horse IgGa, IgGb, IgGc and IgA were identified in normal donkey, mule, hinny and zebra serum.

This study explores the presence and characteristics of various subclasses of immunoglobulins (antibodies) – specifically IgGa, IgGb, and IgGc – in ordinary and immune donkey serum. The researchers also investigated these immunoglobulins’ presence in the serum of other equine species such as mules, hinnies, and zebras.
Methodology and Objectives
- The researchers isolated IgGa from both normal and immune donkey serum through a column chromatography process on DEAE-cellulose, a kind of ion-exchange resin.
- Using isolated IgGa, and mixtures of IgGa and IgGb, they prepared rabbit reagents. These rabbit reagents are specific to the two immunoglobulin subclasses.
- They also obtained serum from a single donkey at different points during BSA hyperimmunization. Then, they isolated the respective antibodies via immuno-adsorption and determined the class or subclass of immunoglobulins among isolated donkey anti-BSA antibodies.
- The researchers’ primary goal was to identify the presence of the homologues of horse IgGa, IgGb, IgGc, and IgA in normal donkey serum and in the serum of other equine species, such as mules, hinnies, and zebras.
Findings and Importance
- The study makes a significant contribution to understanding the immune system of donkeys and other equine species. It demonstrates the presence of specific antibody subclasses (IgGa, IgGb, IgGc, and IgA) in their serum.
- Insights obtained from these investigations can inform the development of better, species-specific therapies in veterinary medicine, particularly for diseases that affect equine species.
- The study also enhances the knowledge about the immune responses generated in these species in response to hyperimmunization, thereby contributing to immunological research focusing on equine species.
