Intrageneric amplification of horse microsatellite markers with emphasis on the Przewalski’s horse (E. przewalskii).
Animal genetics    December 1, 1994   Volume 25, Issue 6 401-405 doi: 10.1111/j.1365-2052.1994.tb00530.x
Breen M, Downs P, Irvin Z, Bell K.Primer sequences flanking 13 microsatellite loci isolated from the domestic horse (E. caballus) were successfully used to amplify homologous loci in the Przewalski's horse (E. przewalskii). The results demonstrate that the level of polymorphism at all 13 loci in the Przewalski's horse was comparable to that in the domestic horse and the overall exclusion probability in the Przewalski's horse was calculated to be 0.9994. The results suggest that it should be possible to use E. caballus-derived microsatellite markers to provide parentage verification and additional valuable information to the ca...

Animal genetics    August 1, 1994   Volume 25, Issue 4 215-221 doi: 10.1111/j.1365-2052.1994.tb00196.x
Ishida N, Hasegawa T, Takeda K, Sakagami M, Onishi A, Inumaru S, Komatsu M, Mukoyama H.The D-loop regions in equine mitochondrial DNA were cloned from three thoroughbred horses by polymerase chain reaction (PCR). The total number of bases in the D-loop region were 1114 bp, 1115 bp and 1146 bp. The equine D-loop region is A/T rich like many other mammalian D-loops. The large central conserved sequence block and small conserved sequence blocks 1, 2 and 3, that are common to other mammals, were observed. Between conserved sequence blocks 1 and 2 there were tandem repeats of an 8 bp equine-specific sequence TGTGCACC, and the number of tandem repeats differed among individual horses....

Blood protein polymorphisms in the donkey (Equus asinus).
Animal genetics    June 1, 1994   Volume 25 Suppl 1 109-113 doi: 10.1111/j.1365-2052.1994.tb00413.x
Bell K.Transferrin, albumin, 6-phosphogluconate dehydrogenase and vitamin D-binding protein polymorphisms were detected in 242 feral and domesticated Australian donkeys by polyacrylamide gel electrophoresis, starch gel electrophoresis, autoradiography, immunoblotting with specific antisera and activity staining. All four TF and two ALB variants were donkey specific while only one of the PGD variants was donkey specific. The two GC variants were electrophoretically identical to the Equus caballus F and S proteins. Available evidence suggested that the TF, ALB, PGD and GC systems are controlled by co-d...
Population genetics of Great Basin feral horses.
Animal genetics    June 1, 1994   Volume 25 Suppl 1 67-74 doi: 10.1111/j.1365-2052.1994.tb00405.x
Bowling AT.The genetic make-up of Great Basin wild (feral) horses was investigated by blood typing studies. Blood samples of 975 feral horses from seven trap sites in Nevada and Oregon were tested by serological and electrophoretic techniques for genetic markers at 19 polymorphic loci. The average number of variants for the seven feral populations [72.1 +/- 3.2 (SEM), range 62-85] was not significantly different from that of 16 domestic breeds (75.0 +/- 11.5, range 58-105). The expected average frequency of heterozygotes per locus (average heterozygosity) for the feral populations (0.402 +/- 0.009, range...
Comparison of thoroughbred and Arabian horses using RAPD markers.
Animal genetics    June 1, 1994   Volume 25 Suppl 1 105-108 doi: 10.1111/j.1365-2052.1994.tb00414.x
Bailey E, Lear TL.We compared pools of DNA from 10 Thoroughbred horses and 10 Arabian horses for the presence of randomly amplified polymorphic DNA (RAPD) markers which might be useful in distinguishing between the breeds. Using 212 decamer oligonucleotides and our polymerase chain reaction (PCR) conditions, 173 of the primers produced scoreable bands. The number of bands ranged from 0 to 9 with an average of 3.6. In family studies using 11 arbitrarily selected primers, five of the 11 primers produced polymorphic bands which exhibited Mendelian inheritance as dominant markers. When comparing the pooled DNA from...
Studies on the frequency and associations of equine leucocyte antigens in sarcoid and summer dermatitis.
Animal genetics    June 1, 1994   Volume 25 Suppl 1 75-80 doi: 10.1111/j.1365-2052.1994.tb00406.x
Lazary S, Marti E, Szalai G, Gaillard C, Gerber H.The equine leucocyte antigen (ELA) types and the clinical diagnosis for equine sarcoid and summer dermatitis were evaluated in 2026 horses representing five breeds. Data were analysed in unrelated animals and in family material. In the case of equine sarcoid, a strong association was observed between the ELA class II DW13 antigen and its effect on Swiss (cP < 0.001), French (cP < 0.0001) and Irish (cP < 0.01) Warmblood horses. The class I antigen A3 occurred more frequently in sarcoid-affected French horses (cP < 0.001). These results confirm our earlier findings (Gerber et al. 198...

Characterization of a red blood cell antigen in donkeys and mules associated with neonatal isoerythrolysis.
Animal genetics    April 1, 1994   Volume 25, Issue 2 119-120 doi: 10.1111/j.1365-2052.1994.tb00091.x
McClure JJ, Koch C, Traub-Dargatz J.A red cell antigen of donkeys and mules was identified using antibodies in serum from a mare which produced a mule foal affected with neonatal isoerythrolysis (NI). Subsequently antibodies with similar activity were identified in the sera of other mares which had produced mule foals and were produced by immunization of horses with blood from donkeys. The antigen detected by these antibodies does not correspond to any recognized horse red cell alloantigen. This may be a xenoantigen since all donkeys (and mules) tested have shared this antigen and all horses tested have lacked the antigen. The r...
A new phenogroup in the horse D system of red cell alloantigens found in the Caspian pony.
Animal genetics    February 1, 1994   Volume 25, Issue 1 49-50
Cothran EG, Long YG.A new D blood group phenogroup consisting of the specificities adeo was observed in a stallion family of Caspian ponies from Iran. An additional six undescribed genetic variants were seen in a total of 82 Iranian horses. This result suggests extensive new genetic variation may be present in domestic horses from geographic regions that have not yet received scientific attention.

Animal genetics    February 1, 1994   Volume 25, Issue 1 19-23
Marklund S, Ellegren H, Eriksson S, Sandberg K, Andersson L.Ten (TG)n positive clones, isolated from an equine genomic library and sequenced, contained 12-19 uninterrupted TG repeats. Primers for polymerase chain reaction (PCR) were synthesized and nine of these (TG)n loci (HTG7-15) were successfully amplified and utilized in this study together with five previously reported equine microsatellite loci (HTG2-6). The PCR products were analysed by polyacrylamide gel electrophoresis followed by automated laser fluorescence detection or autoradiography. All microsatellites showed polymorphism and stable Mendelian inheritance. Differences in microsatellite v...
Genetic blood markers in Arabian, Barb and Arab-Barb horses in Morocco.
Animal genetics    February 1, 1994   Volume 25, Issue 1 45-47
Ouragh L, Mériaux JC, Braun JP.Gene frequencies at 16 blood group and protein polymorphism loci (A, C, D, K, P, Q, U, Al, Gc, Es, A1B, Tf, PGD, PGM, GPI and Pi) are given for three horse breeds in Morocco (Arabian, Arab-Barb and Barb). These data are used to calculate average heterozygosity (h), Nei's standard genetic distance (DN) and probability of exclusion (PE). Variability expressed as the average heterozygosity was lower in the Arabian (0.330 +/- 0.066), while it was higher and almost the same in the Arab-Barb (0.413 +/- 0.071) and the Barb (0.414 +/- 0.070). The shortest genetic distance was found between Barb and Ar...

Silent blood chimaerism in a mare confirmed by DNA marker analysis of hair bulbs.
Animal genetics    August 1, 1993   Volume 24, Issue 4 323-324 doi: 10.1111/j.1365-2052.1993.tb00322.x
Bowling AT, Stott ML, Bickel L.Microsatellite DNA markers in a mare's hair bulbs not concordant with markers in her blood confirmed the hypothesis of chimaerism which had been proposed to explain the apparent parentage exclusion of the mare from her suckling foal. Parentage analysis for this foal based on genetic markers not originating from blood cells of its dam supported a parentage verification conclusion.
Synteny mapping in the horse using horse-mouse heterohybridomas.
Animal genetics    August 1, 1993   Volume 24, Issue 4 257-260 doi: 10.1111/j.1365-2052.1993.tb00308.x
Williams H, Richards CM, Konfortov BA, Miller JR, Tucker EM.In a study of 35 horse-mouse heterohybridoma cell lines, synteny in the horse was found between LDHB, PEPB and IGF1 and between NP, MPI and IDH2. A synteny between ADA and PEPC was also indicated. The loci for horse immunoglobulin light chain (IgL) genes and for LDHA were independent.
DNA sequence analysis of serologically detected ELA class II haplotypes at the equine DQ beta locus.
Animal genetics    June 1, 1993   Volume 24, Issue 3 187-190 doi: 10.1111/j.1365-2052.1993.tb00285.x
Szalai G, Bailey E, Gerber H, Lazary S.The genetic diversity at the ELA DQ beta locus was investigated using polymerase chain reaction and DNA sequencing. Based upon serological methods 16 class II homozygous animals were selected and their genomic DNA was used. A DQ beta gene from an equine cDNA library was also sequenced. Our methodology and the similarity between the genomic and the cDNA sequences suggest that the studied locus is expressed on equine lymphocytes. In the predicted amino acid sequence the most extensive variation is located at residues 56-60. The pattern of these five amino acids is strongly correlated to the sero...

Animal genetics    January 1, 1992   Volume 23, Issue 3 279-281 doi: 10.1111/j.1365-2052.1992.tb00143.x
Zurkowski M, Kuryl J.A new genetic variant in the horse Pi system, designated Z2, was reported in Polish Arabs by using two-dimensional agarose polyacrylamide gel electrophoresis. The frequencies of Pi alleles F, G, L, L2, N, S, U, W, Z and Z2 were found to be 0.036, 0.005, 0.171, 0.013, 0.008, 0.237, 0.416, 0.003, 0.107 and 0.004 respectively.
Cloning of highly polymorphic microsatellites in the horse.
Animal genetics    January 1, 1992   Volume 23, Issue 2 133-142 doi: 10.1111/j.1365-2052.1992.tb00032.x
Ellegren H, Johansson M, Sandberg K, Andersson L.We have isolated equine microsatellites by screening a genomic library with (TG)n and (TC)n probes. TG microsatellites were found to be more abundant than TC repeats, with an estimated frequency of one per 100,000bp. Sequence analysis of eight TG-positive clones revealed varying structures of the repeat regions; perfect stretches of TG repeats, imperfect stretches of TG repeats and compound regions of TG and TC repeats. Five loci were analysed by PCR and showed extensive polymorphism; three to seven alleles and heterozygosities of 0.40-0.76 were observed when screening 20-30 unrelated individu...
Localization of the calcium release channel gene in cattle and horse by in situ hybridization: evidence of a conserved synteny with glucose phosphate isomerase.
Animal genetics    January 1, 1992   Volume 23, Issue 1 43-50
Chowdhary BP, Harbitz I, Davies W, Gustavsson I.In situ hybridization techniques were used to localize regionally the calcium release channel (CRC) gene on cattle and horse chromosomes, using a porcine CRC cDNA probe. In cattle, the hybridization signal peaked on the 18q23-q26 bands and in horse on the 10pter region. Previous studies have shown that the glucose phosphate isomerase (GPI) gene localizes at the same site in both species, indicating that the two loci are syntenic. As CRC and GPI are syntenic in human, pig and mouse, the present results in cattle and horse represent another example of synteny conservation in the evolution of mam...
DNA fingerprinting in horses using a simple (TG)n probe and its application to population comparisons.
Animal genetics    January 1, 1992   Volume 23, Issue 1 1-9
Ellegren H, Andersson L, Johansson M, Sandberg K.A synthetic polynucleotide (TG)n was hybridized to equine DNA digested with HinfI and hypervariable hybridization patterns were obtained. Mendelian inheritance of these DNA fingerprinting patterns was confirmed by pedigree analysis. Estimates of the probabilities of identical band patterns in unrelated individuals of different breeds (Swedish Trotters, North Swedish Trotters, Thoroughbreds and Arabians) were in the range 1 x 10(-4) - 7 x 10(-6). The variability derived with the (TG)n probe in horses was higher than what we obtained with several other commonly used probes for DNA fingerprinting...
One-dimensional isoelectric focusing and immunoblotting of equine major histocompatibility complex class I antigens.
Animal genetics    January 1, 1992   Volume 23, Issue 2 87-95
Schuberth HJ, Anders I, Pape U, Leibold W.The cells of 60 randomly selected Hannoveranian warm-blooded horses were subjected to one-dimensional isoelectric focusing and immunoblotting with a cross-reacting monoclonal antibody (Bo 1) recognizing bovine class I antigens. The banding patterns were correlated with the serologically defined specificities of the ELA-A locus. ELA-A2 was correlated with four bands, while ELA-A5, ELA-W18, ELA-A6, ELA-A14 and ELA-A9 were correlated with a single band each. The complexity of the pattern and additional polymorphic bands which could not be correlated to any of the known ELA specificities may indic...

Bowling AT, Nickel LS.Family data from Paso Fino horses support the existence of a new allele (Aabdf) in the A system of red cell alloantigens. Considering breeds throughout the world, the A system now consists of 13 alleles defined by reagents which serologically detect seven factors.
Linkage of hyperkalaemic periodic paralysis in quarter horses to the horse adult skeletal muscle sodium channel gene.
Animal genetics    January 1, 1992   Volume 23, Issue 3 241-250 doi: 10.1111/j.1365-2052.1992.tb00136.x
Rudolph JA, Spier SJ, Byrns G, Hoffman EP.A genetic disease observed in certain Quarter horses is hyperkalaemic periodic paralysis (HYPP). This disease causes attacks of paralysis which can be induced by ingestion of potassium. Recent studies have shown that HYPP in humans is due to single base changes within the adult skeletal muscle sodium channel gene. A large Quarter horse pedigree segregating dominant HYPP was studied to determine if mutations of the sodium channel gene are similarly responsible for HYPP in horses. We used cross-species, PCR-mediated, cDNA cloning and sequencing of the horse adult skeletal muscle sodium channel a...
