Advancing maternal age predisposes to mitochondrial damage and loss during maturation of equine oocytes in vitro.

Abstract: In many mammalian species, reproductive success decreases with maternal age. One proposed contributor to this age-related decrease in fertility is a reduction in the quantity or functionality of mitochondria in oocytes. This study examined whether maternal age or (in vitro maturation). IVM affect the quantity of mitochondria in equine oocytes. Oocytes were collected from the ovaries of slaughtered mares categorized as young (<12 years) or aged (≥12 years) and either denuded and prepared for analysis immediately (not-IVM) or matured in vitro for 30 hours before preparation (IVM). The mean oocyte mitochondrial DNA copy number was estimated by quantitative polymerase chain reaction and found to be significantly lower in oocytes from aged mares and that had been subjected to IVM than in any other group. Transmission electron microscopy demonstrated that mitochondria in aged mare oocytes subjected to IVM experienced significantly more swelling and loss of cristae than in other groups. We conclude that maternal aging is associated with a heightened susceptibility to mitochondrial damage and loss in equine oocytes, which manifests during IVM. This predisposition to mitochondrial degeneration probably contributes to reduced fertility in aged mares.

The research explores the link between an increase in maternal age and a decrease in the quality and quantity of mitochondria in horse eggs. This decrease is thought to contribute to lower fertility rates in older mares.

Objective of the Study

  • The objective of the study is to understand if maternal age and the in-vitro maturation (IVM) process contribute to a reduction in the number or functionality of mitochondria in the oocytes of equine species. The researchers hypothesize that this could be a possible reason behind the decrease in fertility observed with advancing maternal age in mammals.

Research Methodology

  • The study involved collecting oocytes from the ovaries of mares that had been slaughtered, categorized into two groups based on age: young (less than 12 years old) and aged (12 years and older).
  • The oocytes collected were divided into two groups: one that was directly prepared for analysis (not-IVM) and another which underwent in-vitro maturation (IVM) for 30 hours before preparation.

Findings from the Study

  • Through a quantitative polymerase chain reaction, the researchers estimated the average mitochondrial DNA copy number in the oocytes. It was found to be significantly lower in the oocytes derived from older mares and those that had undergone IVM.
  • Transmission electron microscopy was employed to observe the mitochondria in the oocytes. It demonstrated a significantly higher amount of swelling and loss of cristae (inner foldings of a mitochondrion) in the oocytes of the older mares, particularly when subjected to IVM.

Conclusions from the Study

  • The study concluded that as a mare ages, its oocytes show a greater predisposition towards mitochondrial damage and loss, particularly noticeable during in-vitro maturation.
  • Such mitochondrial degeneration likely contributes to the decrease in fertility seen in older mares.
