Identification by gas-liquid chromatography-mass spectrometry of 4-O-acetyl-9-O-lactyl-N-acetyl-neuraminic acid, a new sialic acid from horse submandibular gland.

Abstract: The novel sialic acid 4-O-acetyl-9-O-lactyl-N-acetylneuraminic acid has been identified as a constituent of horse submandibular gland glycoproteins in addition to the already known equine sialic acids, N-acetylneuraminic acid, 4-O-acetyl-N-acetylneuraminic acid, 9-O-acetyl-N-acetylneuraminic acid, 4,9-di-O-acetyl-N-acetylneuraminic acid, N-glycolylneuraminic acid, 4-O-acetyl-N-glycolylneuraminic acid and 9-O-acetyl-N-glycolylneuraminic acid. The structure has been established by combined gas-liquid chromatography-mass spectrometry.

This research article highlights the identification of a new sialic acid, named 4-O-acetyl-9-O-lactyl-N-acetylneuraminic acid, present in horse submandibular gland glycoproteins, by utilizing the method of combined gas-liquid chromatography-mass spectrometry.

Overview of the Research

  • The study focuses on the identification of a novel sialic acid named 4-O-acetyl-9-O-lactyl-N-acetylneuraminic acid. This acid has been discovered as a component of horse submandibular gland glycoproteins.
  • Researchers have used gas-liquid chromatography-mass spectrometry, a common technique for the identification and analysis of complex mixtures, to reach their conclusion.

Context of the Research

  • The new sialic acid is found in addition to other previously known equine sialic acids including N-acetylneuraminic acid, 4-O-acetyl-N-acetylneuraminic acid, 9-O-acetyl-N-acetylneuraminic acid, 4,9-di-O-acetyl-N-acetylneuraminic acid, N-glycolylneuraminic acid, 4-O-acetyl-N-glycolylneuraminic acid and 9-O-acetyl-N-glycolylneuraminic acid.
  • Sialic acids are a group of acidic monosaccharides found in animal tissues, most commonly known for their role in cellular recognition, interactions, and adhesion.

Methodology

  • The method utilized for the determination of the new sialic acid is combined gas-liquid chromatography-mass spectrometry.
  • Gas-liquid chromatography allows for the separation of volatile compounds, which can then be detected and identified by mass spectrometry, a technique sensitive and descriptive enough to detect the unique spectral patterns of individual molecular species, enabling the identification of the new sialic acid.

Implications of the Research

  • The discovery of a new sialic acid expands our understanding of the complex molecular make-up of horse submandibular gland glycoproteins and potential roles in equine health and disease.
  • Moreover, this new finding could contribute to the broader field of biochemistry and medical research, as it may influence the understanding of cellular interactions and biological processes involving sialic acids and glycoproteins.
