Hemoglobin is an iron-containing protein found in the red blood cells of horses, responsible for the transport of oxygen from the lungs to the tissues and the return of carbon dioxide from the tissues to the lungs. It plays a vital role in maintaining cellular respiration and energy metabolism. The structure and function of hemoglobin can be influenced by various factors, including genetics, age, and health status. Research into equine hemoglobin includes studies on its biochemical properties, the impact of different physiological and pathological conditions on its levels, and its importance in performance and endurance. This page compiles peer-reviewed research studies and scholarly articles that explore the characteristics, regulation, and clinical implications of hemoglobin in equine physiology and health.
HABEEB AF, SMITH DB.Horse globill and its conlponent polypeptide chains obtained by fractional
precipitation and column chroinatography have been ailalyzed for their con- stituent amino acids. The principal difference between the two chains is that
the valyl-leucyl chain is rich in serine and threonine and poor in glutamic acid
and tryptophan compared to the \-alyl-glutaininyl chain.
Komosa M, Flisińska-Bojanowska A, Gill J.1. In six foals and their mothers (of Standard breed) diurnal changes in the haemoglobin level, red blood cell number and mean corpuscular haemoglobin during the first 13 weeks of foal life were studied. 2. Studies begun when a foal reached 7 days of age and were repeated every two weeks till 13 weeks of foal life. Blood was taken every 6 hr in foals and every 4 hr in mares. 3. No diurnal rhythmicity in parameters studied either in foals or in mares was found. 4. Decrease of haemoglobin level in lactating mares (14.0-12.5 g/100 ml) without changes in red blood cell number were observed. Mean c...
Knight PK, Ray SP, Rose RJ.Because the haemoglobin concentration Hb[ during exercise may be at the upper limit of blood viscosity for effective oxygen delivery, we hypothesised that administration of blood would not further enhance oxygen delivery or exercise capacity. Six Thoroughbred geldings were used in 5 incremental treadmill exercise tests over a period of 4 weeks. The first test was performed 6 days prior to phlebotomy, which involved the removal of 20 ml/kg bwt of venous blood. Exercise tests were performed at 1, 8 and 15 days after blood removal. Six days after the 15 day post phlebotomy exercise test, blood fr...
von Rennenkampff F, Kraft H.During 20 weeks blood constituents in peripheral blood (Erythrocyte count, Packed cell volume, Hemoglobin, Sedimentation rate, activities of Aspartate aminotransferase, Alanine aminotransferase, Creatine phosphokinase, Lactate dehydrogenase, Lactate dehydrogenase-1-isoenzyme, Glucose, Lactate, Total bilirubin), pulse and respiration rate were determined at rest and after exercise. Significant increase of blood values after exercise could be noted in relation to the intensity of exercise and the performance of the animals. The well trained horses had an increase of their performance according t...
Soma LR, Uboh CE, Guan F, Luo Y, Moate PJ, Boston RC, Driessen B.Hemoglobin-glutamer-200 (HBOC-200) is a hemoglobin (Hb)-based oxygen carrier (HBOC) comprising glutaraldehyde-polymerized bovine Hb. In this study, we sought to determine the pharmacokinetics of this first generation HBOC after IV infusion of 32.5 g of HBOC-200 solution in horses. Quantification of HBOC-200 in equine plasma and urine was performed using a method recently developed by our laboratory. The elimination from plasma was based on size distribution of the bovine Hb polymer. The decline of plasma concentration-time curve of HBOC-200 was described by a noninterchanging 2-compartmental m...
Smale K, Butler PJ.A new oxygen equilibrium curve is defined for the Thoroughbred horse under standard conditions of 37 degrees C, pH = 7.4 and PCO2 = 5.33 kPa. The "standard" P50 for the Thoroughbred is, at 2.83 +/- 0.04 (SE of mean) kPa, significantly lower than that found for the Hanoverian horse (3.17 +/- 0.03 kPa) by Clerbaux et al. (Can. J. Vet. Res. 50: 188-192, 1986), and lower than other values for horses in the literature. Using data from Butler et al. (J. Exp. Biol. 179: 159-180, 1993), curves were also constructed, in vitro, under simulated conditions of intense exercise to examine the individual eff...
Gallagher K, Stowe HD.Blood samples were obtained from 45 racing Standardbred horses immediately before and immediately after a training jog at a commercial race track to establish reference values for serum selenium and related values of the peroxide reduction system of racing horses. Hemoglobin, hematocrit, and serum selenium increased significantly (P < 0.001), whereas RBC-reduced glutathione decreased significantly (P < 0.01) immediately after exercise. Glutathione peroxidase and glutathione reductase were not altered by exercise. Serum copper and magnesium were significantly increased (P < 0.01) after...
Sneddon JC, Van der Walt J, Mitchell G.1. The resistance of sub-tropical horses, and desert-dwelling horses to 72 hr dehydration/24 hr rehydration was investigated via changes in red cell parameters and plasma protein concentration. 2. Red cell count, haemoglobin and haematocrit increased up to 48 hr dehydration. Between 48 and 72 hr dehydration these parameters decreased, implying a fluid shift onto the intravascular space from the interstitium/hindgut. Most parameters had regained baseline values by 24 hr rehydration. 3. Mean cell volume, mean cell haemoglobin, mean cell haemoglobin concentration and total plasma protein were not...
Corradini I, Georges K, Jose-Cunilleras E.To determine the effects of time after sampling on CO-oximetry measurements of equine blood samples and the effects of adding ascorbic acid (AscAc) and methylene blue (MetBlue) to samples with methemoglobinemia. Methods: Experimental study. Methods: University teaching hospital. Methods: Thirty healthy adult horses assigned to 5 groups. Methods: Repeated CO-oximetry determinations were performed on venous (n = 6) and arterial blood samples (n = 7) stored at 0°C for 48 hours. Methemoglobinemia was induced in vitro in 17 additional blood samples. Six were used as untreated controls, 6 had ...
Krzywanek H, Mohr E, Mill J, Scharpenack M.Until the age of about 2 years, trotters normally grow up on pasture without any kind of training. In the stud farm Lindenhof (Templin, Germany), however, these first 2 years are used for a special fitness training for the young animals: 2-3 times a week, a group of the yearlings is forced to run a distance of about 1700 m on a track at an average speed of up to 10 m/s. Until now, little was known about changes of blood parameters which may occur during such special exercise. This study therefore investigated the activity of selected serum enzymes (aspartate-amino-transferase (AST), alanine-am...
Zobel G, Kolb FE.Acid phosphatase of erythrocytes of several species was investigated, with three isozymes having been recorded from swine (three types), three (two types) from horse, four (one type) from dog, two (two types) from cat, two (three types) from duck, and two (one type) from fowl. The Michaelis constant of the enzyme varied between 3.5 and 5 X 10(-4) M for the species involved. The species, however, differed slightly for the optimum pH of the enzyme. The average enzymatic activities were (5.68 +/- 0.42 for dog, 4.46 +/- 1.0 for horse, 3.8 +/- 0.24 for swine, 3.72 for cat, 2.5 +/- 0.62 for duck, an...
Bodurov N, Filipov Zh.Comparative clinical and experimental investigations were carried out on 18 clinically normal horses. It was found that the single transfusion of in vitro UV-treated autogenous blood, following a previously described method, was tolerated well by horses and did not lead to any deviations from the normal indices. Under the effect of the treated blood there was an increase in the hemoglobin content, rise in the erythrocyte and leukocyte count with slightly expressed hyperleukocytosis and weak neutrophilia (simple regeneration of the blood). The sedimentation of erythrocytes also showed higher ra...
Allen BV.Results of blood counts have been analysed in three-year-old racehorses in training comprising 77 colt stayers, 27 colt sprinters, 61 filly stayers and 35 filly sprinters. The distributions of haemoglobin, erythrocyte count and haematocrit were significantly higher in colt stayers compared to the other three groups. In fillies these values were also significantly higher in stayers compared to sprinters. The erythrocyte count was significantly higher in filly stayers compared to colt sprinters but there were no significant differences between haemoglobin or haematocrit values in these groups. N...
Milne EM.Equine serum haptoglobin was separated by polyacrylamide gel isoelectric focusing and visualized by protein staining or Western blotting. Conventional protein staining revealed up to three bands in the pI range 4.17 to 4.44. The blotting technique, however, showed an anodal group of 8 to 10 bands with a pI range of 4.11 to 4.52 and a cathodal group of 4 to 6 bands with a range of 4.55 to 5.14. The blotting method revealed that equine haptoglobin migrates outside the prealbumin area, in contrast to previous reports.
Osterhoff DR, Groenewald J.By using isoelectric focusing in thin agarose slab gels 1049 Thoroughbred, 82 Nooitgedachter, 45 Percheron and 244 horses of other breeds were examined. The numbers of other Equidae tested were 107 donkeys, 50 mules, 4 common zebras (Equus burchelli boehmi) and 8 mountain zebras (Equus zebra hartmannae). Phenotypic data are presented for all tested animals and gene frequencies are calculated for the horses.
Rubio MD, Muñoz A, Santisteban R, Tovar P, Castejón FM.Exercise-induced hematological alterations were studied in 20 four-year old foals, 11 Andalusian and 9 Arabian. They were subjected to a test exercise program consisting of 4 levels of gradually increasing intensity (15, 20, 25 and 30 km/hr) with a duration of 5 min each. Blood samples were taken during resting, after each exercise level and at 10 and 30 min of recovery from exercise. The following hematic parameters were analyzed: red blood count, hematocrit value, hemoglobin, volumetric indexes (MCV, MCH and MHCH) and total plasma proteins. The alterations as a result of the physical effort ...
Paradis MR, Breeze RG, Laegreid WW, Bayly WM, Counts DF.Eight ponies were allotted to 2 groups of 4. Group-1 ponies (1-4) were given 0.2 g of indole/kg of body weight orally and group-2 ponies (5 to 8) were given 0.1 g of indole/kg. Various physical, hematologic, and physiologic measurements were obtained after administration of indole. Intravascular hemolysis and hemoglobinuria were detected in both groups within 24 hours of dosing. Hemolysis was reflected by decreases in PCV, hemoglobin concentration, and RBC count, and an increase in indirect bilirubin. Erythrocyte fragility appeared to increase in both groups at 8 hours after dosing and peaked ...
Yamada S, Suruga K, Ogawa M, Hama T, Satoh T, Kawachi R, Nishio T, Oku T.The appearance of NO2- reducing activity of cytochrome c (Cyt c) upon heat denaturation was investigated with equine heart Cyt c. Denatured equine heart Cyt c (dCyt c), which was treated at 100 degrees C for 30 min, had NO2- reducing activity in the presence of dithionite and methylviologen in an aqueous solution under anaerobic conditions. In contrast, hemoglobin and myoglobin had no such activity under the same conditions. Using spectroscopic methods, we found that the appearance of this activity in the Cyt c was due to the following intramolecular changes: unfolding of the peptide chain, ex...
Tennent-Brown BS, Goetz TE, Manohar M, Hassan AS, Freeman DE, Bundy JS, Evans MR.Dehydration and the associated impairment of cardiovascular and thermoregulatory function comprise major veterinary problems in horses performing prolonged exercise, particularly under hot and humid conditions. For these reasons, there is considerable interest in using pre-exercise hyperhydration to help maintain blood volume in the face of the excessive fluid loss associated with sweat production during prolonged exertion. However, recently it was reported that pre-exercise hyperhydration causes arterial hypoxemia in horses performing moderate intensity exercise simulating the second day of a...
Goldztein S, Carreras Vescio LA, Salamone HJ, Calahonra R, Kohan AI, Sánchez Avalos JC.A 24-year-old male patient with a severe aplastic anemia (SAA) was treated with equine-antilymphocyte globulin (ALG). As complication of this treatment he developed a severe heteroimmune hemolytic anemia mediated by anti-species pan-agglutinin antibodies present in ALG. In spite of the fact that ALG is absorbed with red-cell stroma and platelets to remove anti-erythrocyte and anti-platelet contaminating antibodies, often only partial absorption is achieved, and the remaining antibodies are passively acquired by the recipient. Neutropenia and especially thrombocytopenia are usual complications ...
Rifkind JM.The oxidation of horse hemoglobin by Cu(II) has been followed by the changes in the electron spin resonance spectra of copper. By stopped-flow and freeze-quenching techniques, it is shown that the second-order rate constant for the binding of Cu(II) to hemoglobin is greater than 5 X 10(5) mol-1 s-1 and the apparent first-order rate for the reduction of Cu(II) to Cu(I) is 0.051 s-1. It is also shown that the binding of Cu(II) to hemoglobin is followed by an alteration of the Cu(II) spectrum, decreasing the g values. This process has an apparent rate constant of 17 s-1 and presumably involves a ...
Khittoo G, Vermette L, Nappert G, Lariviere N.In mammalian species studied previously, pepsinogen consisted of biochemically different groups of isozymogens. By use of gel filtration chromatography and electrophoresis, we isolated a predominant pepsinogen from the gastric mucosa of a horse. Peptide mapping with V8 protease revealed differences with its porcine homologue. However, porcine and equine pepsinogens, when activated to pepsin, had a similar pattern of activity when hemoglobin was used as substrate. Those results suggest that differences must exist in the primary structure of the pepsinogens of the 2 species.
Alben JO, Bare GH.Infrared absorption spectra of the alpha-104 (G11) cysteine SH group have been observed for aqueous solutions of hemoglobin derivatives from humans, pigs, and horses. The center frequencies ((nu)SH) show ligand sensitive patterns that are similar for the three species, with (nu)SH (HbCO) <(nu)SH (HbO(2) ~ HbCN) < (nu)SH (Hb(+)) <<(nu)SH (deoxyHb) for human and pig hemoglobins. The alpha-104 SH group is most strongly H-bonded (smallest (nu)SH), has the greatest range of (nu)SH (Hb ? HbCO) in human hemoglobin, and is least strongly H-bonded and has the smallest range of (nu)SH (Hb ? ...
Kedzierski W, Bergero D.The objective of the present study was to compare changes of blood parameters induced by the same work and performed in the same conditions in two racehorse breeds, Thoroughbred and Purebred Arabian. The effect of moderate-intensity exercise was studied in 20 stallions--ten Thoroughbreds, aged 2-3 years and the same number of Purebred Arabians, 3-4 years old. All the horses were administrated the same effort test consisting in 1200 m gallop at a speed typical of the daily training sessions. Three jugular venous blood samples were collected for each horse: at rest, just after the end of the gal...
Frietsch G, Weigand E, Prüstel N.The postnatal iron status of 20 trotter foals (7 female, 13 male) was studied by analyzing different parameters in blood drawn on the day of birth, and 14 +/- 2 and 28 +/- 2 days later. Hemoglobin hematocrit, plasma iron and transferrin saturation averaged 14.9 g/dl, 41.8%, 300 micrograms/dl and 59.4%, respectively, on the day of birth. These values were significantly reduced after 14 days, but had approached again the initial levels by day 28. There was a slight, statistically nonsignificant rise in the mean total iron-binding capacity over the four-week period. Iron status was not affecting ...
Franken P, Schotman JH.The activities and concentrations of a number of erythrocytic enzymes and intermediate products of erythrocyte metabolism were determined in twenty-one normal standard-bred horses which were studied clinically and biochemically. These studies showed that equine anaerobic glycolysis is characterized by a biochemical pattern similar to that observed in human PK deficiency. The greater sensitivity of equine haemoglobin to oxidants is attributable either to low stability of GSH, which may be due either to the low activity of GR or that of 6PGD as observed in the studies. In addition, the saturatio...
Ayo JO, Ake AS, Abimbola AA.The study aimed to evaluate effects of packing (load-carrying) and trekking on hematological and biochemical parameters in donkeys during the hot-dry season. Ten pack donkeys, aged 2-3 years, weighing 93 ± 2.7 kg were enrolled in the study and randomly divided into two groups of five animals each. Group 1 donkeys were subjected to packing + trekking while group 2 trekked only. All the donkeys trekked 20 km thrice, 1-day apart. Blood was collected before and after packing or trekking for hematological evaluation; and serum analyzed for electrolyte concentrations, and aspartate aminotransfera...
Zinchenko AV, Govorova YS.Critical to the understanding the mechanism of destruction and protection during cryopreservation of biological objects is the knowledge of the conformational transitions of biopolymers experiencing low temperatures in the presence of cryoprotective agents. This information may be derived from the kinetic and thermodynamic parameters of macromolecular thermal denaturation kinetics under different environmental conditions. Objective: The study deals with the influence of cryoprotective agents (glycerol, 1.2-propanediol (1.2-PD), and dimethyl sulfoxide (DMSO)) on thermodynamic and kinetic parame...
Steinhardt J, Hiremath CB.Many of the stability characteristics of horse ferrihemo-globin (Hb+) in acid solutions, such as pH dependence and susceptibility to stabilization by iron ligands, are shared by human ferrihemoglobin, but striking differences between the two proteins exist. The most noticeable is the much greater rate of denaturation of the human protein at all pH values. Other differences include a shift to higher pH in the equi-librium between native and acid-denatured forms, differ-ences in the temperature at which the temperature effect on the equilibrium-pH curve reverses, a complete absence in human Hb+ ...
Mirck MH, Franken P.Experimental infections using 600,000 infective larvae of Strongyloides westeri were carried out in seven worm-free Shetland ponies, four foals and thee yearlings. In the foals, the prepatent period varied from ten to fourteen days, the patent period ranging from forty-three to eighty-three days. Within approximately two months after infection, a decrease in the concentration of haemoglobin and an increase in the beta-globulin fraction of the protein pattern of the serum, practically coinciding with the maximum S. westeri faecal egg counts, were recorded. In the yearlings, the prepatent period...
Sant C, Lima DM, d'Abadie R, Pargass I, Georges KC.This study aimed to develop a sound database for the hematological reference intervals of thoroughbred foals in Trinidad, West Indies from birth to 1 month of age. Methods: 89 foals. Methods: Whole blood samples were taken from 89 foals throughout Trinidad at approximately 1 day, 1 week, and 1 month of age. These foals were examined to be classified as healthy or free from disease. Complete blood count (CBC), microscopic analysis of blood smears, and conventional PCR for Theileria equi and Babesia caballi were performed. Results: Of the 89 foals, 67 were deemed healthy and suitable for establi...
