Enzyme-linked immunosorbent assay for detection of antibody to Pseudomonas aeruginosa and measurement of antibody titer in horse serum.
American journal of veterinary research    January 1, 1982   Volume 43, Issue 1 55-60
Ueda Y, Sanai Y, Homma JY.Enzyme-linked immunosorbent assay (ELISA) was used for detection of immunoglobulin (Ig) M and IgG antibodies against a serologically common antigen (original endotoxin protein), protease, and elastase of Pseudomonas aeruginosa. The P aeruginosa antibody in horse sera was measured, using ELISA. Horseradish peroxidase-labeled rabbit anti-horse IgM and IgG antibodies were used for enzyme-labeled antibody conjugate. 5-Aminosalicylic acid and H2O2 were used for substrate. Sera collected from a vaccinated horse, a newborn foal, and 72 healthy racehorses were investigated for antibodies against P aer...
Mesothelial cells of peritoneal fluid.
Equine veterinary journal    January 1, 1982   Volume 14, Issue 1 86-88 doi: 10.1111/j.2042-3306.1982.tb02346.x
Brownlow MA, Hutchins DR, Johnston KG.Cells in the peritoneal fluid from 159 horses were examined in Giemsa stained preparations using light microscopy. Normal mesothelial cells showed an oval nucleus with finely reticular chromatin and pale blue cytoplasm. Activated mesothelial cells occurred in fluids derived from mesothelium under acute or subacute, non-septic stimulus and were remarkable for their pleomorphism and intense basophilia which may mimic neoplasia. Transformed mesothelial cells seen in chronic inflammatory fluids were sometimes phagocytic and showed conspicuous cytoplasmic vacuolation.
Comparison of receptor properties of erythrocyte membrane glycoproteins.
Developmental and comparative immunology    January 1, 1982   Volume 6, Issue 4 765-774
Klimas NG, Caldwell KE, Whitney PL, Fletcher MA.Membrane glycoproteins from horse, sheep, goat and bovine erythrocytes were solubilized and purified. These glycoproteins could be placed in three groups based on their degrees of glycosylation: The major bovine erythrocyte glycoprotein (BGII) had 77% sugar, the minor bovine glycoprotein (BGI) had 27% sugar and the others had approximately 50% sugar. Four of the glycoproteins aggregated in a uniform way in aqueous solution--one, BGII, did not. Four had similar subunit sizes of 25-34,000 daltons, but BGII was larger--55,000 daltons. Receptor functions (for plant and invertebrate lectins, antibo...
Effects of adenosine and deoxyadenosine on PHA-stimulation of lymphocytes of man, horse and pig.
The International journal of biochemistry    January 1, 1982   Volume 14, Issue 5 377-385 doi: 10.1016/0020-711x(82)90023-4
Peters GJ, Oosterhof A, Veerkamp JH.1. Adenosine inhibits thymidine and uridine incorporation of PHA-stimulated lymphocytes of man and horse at concentrations higher than 50 and 10 microM, respectively. Deoxyadenosine is inhibitory at concentrations higher than 100 microM. Thymidine and uridine incorporation of porcine lymphocytes are elevated 5-7-fold by 25-100 microM adenosine, deoxyadenosine, inosine and hypoxanthine. Leucine incorporation of PHA-stimulated lymphocytes was affected by adenosine and deoxyadenosine in the same way, but to a lower extent. 2. Effects of adenosine and deoxyadenosine were more pronounced at shorter...
Chlamydia psittaci induced pneumonia in a horse.
The Cornell veterinarian    January 1, 1982   Volume 72, Issue 1 92-97
McChesney SL, England JJ, McChesney AE.An agent lethal to embryonated chicken eggs was isolated from lung tissues of a quarter horse mare with a fatal respiratory disease. The lesions induced in embryonated chicken eggs, the tinctoral properties, the ultrastructural morphology, the resistance of the organism to sodium sulfadiazine, and the presence of a chlamydial complement fixing antigen, identify this isolate as a member of the family Chlamydiaceae and suggest the agent to be Chlamydia psittaci. Two Shetland ponies experimentally infected with the isolated agent developed subclinical infection as demonstrated by an increase in c...
The Shwartzman phenomenon in equine species.
Experimental pathology    January 1, 1982   Volume 22, Issue 3 165-171 doi: 10.1016/s0232-1513(82)80004-2
Sprouse R, Garner H, Eyestone W.The occurrence of the Local Shwartzman Reaction (LSR) in equine species has not previously been reported. The molecular mechanism appears identical to that reported for the rabbit and other species. The immunopathologic and histopathologic similarities of the experimentally induced LSR in horses and ponies to that of the hoof-laminae (an extension of the skin) lesion in naturally-occurring and/or carbohydrate induced laminitis may offer insight into the pathogenesis of this complex disease.
Application of a chemically inactivated, adjuvanted vaccine to control abortigenic infection of mares by equine herpesvirus I.
Developments in biological standardization    January 1, 1982   Volume 52 493-498
Bryans JT, Allen GP.A chemically inactivated, adjuvanted vaccine prepared from a virulent strain of Equine herpesvirus I (EHV-I) was used to immunize pregnant Thoroughbred broodmares during a five-year field test designed to determine its safety and efficacy. Each mare in the vaccinated groups received 3 intramuscular injections of vaccine beginning immediately prior to and during the last half of pregnancy. Vaccine was injected at approximately 60-day intervals. The accumulated incidence of EHV-I abortions among vaccinated mares during the field trial period was 1.6/1000 as compared with an incidence of 6.8/1000...
Maternal anti-fetal cytotoxic antibody responses of equids during pregnancy.
Journal of reproduction and fertility. Supplement    January 1, 1982   Volume 32 361-369
Kydd J, Miller J, Antczak DF, Allen WR.The maternal immunological response to the developing equine fetus was investigated in 69 pregnancies of various genotypes. In normal intraspecies horse pregnancy (N = 16), 94% of mares produced strong cytotoxic antibody responses to paternal histocompatibility antigens which were first detectable between 44 and 70 days after ovulation. In all other types of pregnancy examined (intraspecies donkey, N = 19; interspecies mule, N = 6; interspecies hinny, N = 2; extraspecies horse-in-donkey, N = 3; and extraspecies donkey-in-horse, N = 21), antibody production was observed less frequently, and som...
Isolation and identification of steroids from gonadal vein blood of the fetal horse.
Journal of reproduction and fertility. Supplement    January 1, 1982   Volume 32 383-387
Raeside JI, Gofton N, Liptrap RM, Milne FJ.Direct connection of the artery of a fetal ovary to the carotid artery of the mare allowed collection of a large volume of blood over a 30-min period. Extraction of steroids and their fractionation was followed by separation of the steroids by alumina adsorption chromatography, and Sephadex LH-20 and Celite partition chromatography. Further resolution of the material by HPLC led to the identification of dehydroepiandrosterone (DHA) by nuclear magnetic resonance and mass spectrometry. Other compounds were isolated, which remain to be identified fully, but in the 8th month of pregnancy the princ...
Ultrasonic echography in the mare.
Journal of reproduction and fertility. Supplement    January 1, 1982   Volume 32 423-430
Chevalier F, Palmer E.Ultrasonic echography in the mare allows pregnancy diagnosis as early as Day 14 after ovulation. In the 1980 and 1981 breeding seasons, a total of 7438 examinations of 4688 mares were performed on several farms with the same apparatus. The accuracy of positive pregnancy diagnosis was estimated to be greater than 95% and of non-pregnancy diagnosis greater than 84%. When abnormal pictures of vesicles are found, it has to be decided whether it is a conceptus or a cystic structure. Most small vesicles do not develop, whereas large ones, deformed shapes, presence of some echoes, or a division insid...
Body fluid concentrations of ampicillin trihydrate in 6 horses after a single intramuscular dose.
Equine veterinary journal    January 1, 1982   Volume 14, Issue 1 83-85 doi: 10.1111/j.2042-3306.1982.tb02345.x
Brown MP, Stover SM, Kelly RH, Farver TB.Six adult mares were given a single dose of ampicillin trihydrate (250 mg/ml) intramuscularly at a dosage of 20 mg/kg body weight. Serum, synovial fluid, peritoneal fluid and urine ampicillin concentrations were measured serially over a 48 h period. The mean peak serum ampicillin concentration was 2.49 micrograms/ml at 6 h. Ampicillin was found in synovial fluid and peritoneal fluid, which obtained mean peak ampicillin concentrations of 1.65 micrograms/ml and 1.81 micrograms/ml at 6 h and 4 h respectively. These concentrations declined in parallel with serum concentrations and were still detec...
Identification of stage-specific and hormonally induced polypeptides in the uterine protein secretions of the mare during the oestrous cycle and pregnancy.
Journal of reproduction and fertility    January 1, 1982   Volume 64, Issue 1 199-207 doi: 10.1530/jrf.0.0640199
Zavy MT, Sharp DC, Bazer FW, Fazleabas A, Sessions F, Roberts RM.Uterine secretions were obtained on Days 4, 8, 12, 14, 16, 18 and 20 of the oestrous cycle and early pregnancy. Acid phosphatase activity was significantly affected by day of the cycle, reaching a maximum at days 12-14 during the luteal phase and then declining to almost undetectable levels, by Day 20. In pregnant animals, activity continued to increase beyond Day 14. Two-dimensional polyacrylamide gel electrophoresis showed that albumin was a major component. However, a number of unique proteins of non-serum origin appeared in mid-cycle but had disappeared by Day 20. One of these was a basic ...
Ovarian activity in the mare between latitude 15 degrees and 22 degrees N.
Journal of reproduction and fertility. Supplement    January 1, 1982   Volume 32 261-267
Saltiel A, Calderon A, Garcia N, Hurley DP.Ovarian activity, as measured by the presence of small (10-15 mm diam.), medium (15-30 mm) and large (greater than 30 mm) follicles, corpora haemorrhagica and corpora lutea (CL), was determined in a total of 3584 ovaries during 1 year using post-mortem specimens. There were significant correlations of the incidence of ovulated follicles (ovulations), large and medium follicles (P less than 0 . 001) and of the mean number of small follicles (P less than 0 . 05) per mare with respect to season. The yearly averages of ovulated, large and medium follicles were 30 . 1, 13 . 9 and 73 . 9%, respectiv...
Clinical and radiological aspects of stifle bone cysts in the horse.
Equine veterinary journal    January 1, 1982   Volume 14, Issue 1 40-46 doi: 10.1111/j.2042-3306.1982.tb02333.x
Jeffcott LB, Kold SE.Thirty-three cases with subchondral bone cysts in the stifle are reported. The condition was most commonly seen in young Thoroughbreds and produced intermittent lameness of varying degree. Radiographically distinct areas of radiolucency were found in the distal femur or proximal tibia adjacent to the femorotibial joint. Lesions were usually unilateral but 5 horses had cysts in both stifles. The cases could be divided into 2 distinct groups. Horses in Group A (28 cases) had a large circular or dome-shaped cyst in the medial femoral condyle with a distinct communication with the femorotibial joi...
A quantitative study of the histological morphology of the endometrium of normal and barren mares.
Canadian journal of comparative medicine : Revue canadienne de medecine comparee    January 1, 1982   Volume 46, Issue 1 17-20
Leishman D, Miller RB, Doig PA.The density of uterine glands, height of surface epithelium, numbers of hemosiderin laden macrophages, inflammatory cells and layers of periglandular fibrosis were evaluated in uterine biopsies from 40 mares. These features were found to be highly variable in normal equine endometrium. Minor pathological changes appeared to be masked by this normal variability. Atrophy of uterine glands was recognized in mares which had been barren for more than three years. No significant differences were found between barren and normal mares in the height of epithelium, number of hemosiderin laden macrophage...
Recovery of contagious equine metritis organisms and development of lesions in experimental infection of mares.
Journal of reproduction and fertility. Supplement    January 1, 1982   Volume 32 187-191
Acland HM, Allen PZ, Kenney RM.Twenty-three of 24 mares were infected experimentally with contagious equine metritis organisms by intrauterine inoculation, and killed 2-116 days later. From mares killed within 14 days after infection the organism could be recovered from many sites in the uterus, and most sites in the cervix, a few sites in the vagina and oviduct and from one clitoral sinus. At this time the endometrial folds were swollen and there were 10-20 ml of fluid in the uterus. In mares killed after 14 days, the organism was recovered from the ovarian surface (1 mare), oviduct (4 mares), uterus (2 mares) and the clit...
Use of ultrasound echography for early diagnosis of single and twin pregnancy in the mare.
Journal of reproduction and fertility. Supplement    January 1, 1982   Volume 32 431-439
Simpson DJ, Greenwood RE, Ricketts SW, Rossdale PD, Sanderson M, Allen WR.In 1981 a trial was conducted on 6 Newmarket Thoroughbred stud farms into the use of real-time ultrasound echography (scanning) as an aid to the early diagnosis of pregnancy in the mare. The procedure was found to be practicable and interpretation of the findings was relatively straightforward. Pregnancy was positively diagnosed from as early as 14 days after ovulation and a number of twin pregnancies were recognized, some in which the conceptuses were situated in separate horns of the uterus but an almost equal number in which the conceptuses were together in the same horn. It was concluded t...
Radioimmunoassay and in-vitro bioassay of serum LH throughout the equine oestrous cycle.
Journal of reproduction and fertility. Supplement    January 1, 1982   Volume 32 253-260
Alexander S, Irvine CH.Mares were bled once daily throughout a cycle, or 3 times daily from the first day of oestrus to the 2nd day after ovulation. LH was measured by heterologous radioimmunoassay and by an in-vitro bioassay based on LH-stimulated testosterone production by mouse Leydig cells. The patterns of bio- and immuno-active LH during the oestrous cycle were similar but not identical, so that in both groups of mares the ratio of biological: immunological (B:I) activity during the LH surge was significantly higher before than after ovulation (P less than 0 . 001). Considerable individual variation in cycle me...
Bronchoscopy and cytological examination of bronchial secretions of horses with chronic obstructive pulmonary disease (COPD).
Le Poumon et le coeur    January 1, 1982   Volume 38, Issue 5 311-316
Deegen E, Müller P, Petzoldt K.On the basis of cytological similarities in chronic obstructive disease (COPD) in human beings and in horses during 1981 a total number of 144 horses with COPD were examined. Bronchial secretions were taken under endoscopic control and stained with a modified Hansel staining procedure. Whereas 26 horses did not exhibit signs of allergic reactions cytologically and 94 horses showed low amounts of eosinocytes and/or mast cells in their secretion; only the secretions of 24 horses (16,67%) were characterized by fairly high contents of these allergy associated cells and low numbers of neutrophilic ...
Erythrocyte volume distribution analysis in healthy dogs, cats, horses, and dairy cows.
American journal of veterinary research    January 1, 1982   Volume 43, Issue 1 163-166
Weiser MG.Erythrocyte volume distribution curves (erythrograms) were determined on a total of 300 blood samples from healthy dogs, cats, horses, and cattle (dairy cows). An index of anisocytosis was determined for these animals. Erythrograms were highly reproducible, and the mean corpuscular volumes determined from erythrograms compared well with those determined from hemograms. Bovine and equine erythrocyte volumes were found to be stable after the blood was stored at 4 C for 24 hours. Under the same conditions, canine and feline erythrocytes increased slightly in volume. After incubation of blood dilu...
Characteristics and fertility of stallion semen.
Journal of reproduction and fertility. Supplement    January 1, 1982   Volume 32 1-8
Dowsett KF, Pattie WA.A study of stallion fertility was conducted on commercial studs in eastern Australia over 4 breeding seasons (1974/75-1977/78). Data from 47 stallions (66 stallion seasons) aged 2-26 years and representing 7 breeds and 1664 mares were used to relate seminal characteristics to fertility. Percentage pregnancies per service was the most sensitive measure of fertility with stallions on commercial studs. Breed and age of stallion, breeding season and the referral status of the stallion ('suspect' or 'normal') had significant (P less than 0.01) effects on fertility. The semen characteristics that we...
[Properties of alpha-1,4 leads to -glucosyltransferase from the muscles and blood serum of horses].
Polskie archiwum weterynaryjne    January 1, 1982   Volume 23, Issue 3 73-85
Dzongowska-Dzongu T, Kotoński B, Hutny J.Alpha-1,4 leads to 1,4-glucosyltransferase preparations from horse muscles and serum were studied. The enzyme proteins from both tissues are very similar. Both proteins have a molecular weight of 240 000 and consist of four subunits of 60 000 daltons each. pH 5,0 is optimal for the activity. Only substrates with alpha-1, 4-linkages can serve as glucosyl donors for transferase reactions. Km values for both enzymes differ very slightly. At low substrate concentrations the hydrolytic activity can be found in addition to transferase reactions. At the concentration of the substrate higher than 40 m...
[Maduromycotic mycetoma in a horse].
Veterinarni medicina    January 1, 1982   Volume 27, Issue 1 37-43
Otcenásek M, Mátl J, Vítovec J, Vladík P, Wohlman J.A case of maduromycotic mycetoma (eumycetoma) in seven years old draught horse is described. The disease was localized in anal region and healed after surgical treatment. Attention is drawn to the necessity of distinguishing three types of mycosis in horses, characterized by the origin of tumor lesions - mycetomas, hyphomycosis and entomophthoromycosis - and information was gathered on their etiology and geographical occurrence. On the basis of the morphology of fungal elements traced in inflammated changed tissues and with regard to the existing findings on the origin of eumycetoma in animals...
Anthelmintic efficacy of ivermectin given intramuscularly in horses.
American journal of veterinary research    January 1, 1982   Volume 43, Issue 1 145-148
DiPietro JA, Todd KS, Lock TF, McPherron TA.The anthelmintic activity of ivermectin was evaluated in 18 female horses with naturally acquired parasitic infections. Horses were treated once (IM) with vehicle only (n = 6), 200 microgram/kg of body weight (n = 6), and 300 microgram/kg (n = 6). Efficacy of both dosages of ivermectin was greater than 99% against Gasterophilus spp, 100% against Trichostrongylus axei, Habronema muscae, H majus, and Draschia megastoma, 98% to 99% against adult cyathostomes, 86% to 97% against 4th-stage cyathostomes, and 100% against adult large strongyles. Although ivermectin was incomplete in its activity agai...
An investigation of sperm migration into the oviducts of the mare.
Journal of reproduction and fertility. Supplement    January 1, 1982   Volume 32 59-64
Bader H.A total of 23 mares were inseminated once within 0-6 h after clinical detection of ovulation, 14 with fresh and 9 with deep-frozen semen containing 0.1 x 10(9) to 4.7 x 10(9) motile spermatozoa. Within these two groups, the mares were slaughtered 2, 4 or 6 h after insemination and their genital tracts removed. The utero-tubal junction, isthmus and ampulla ipsilateral to the ovary in which ovulation occurred were flushed separately for sperm recovery. In 1 or 2 mares of each group, the uterine horn and corpus uteri, the cervix and vagina were also flushed. Tissue samples were collected from the...
Changes in plasma progesterone levels during storage of heparinized whole blood from cow, horse, dog and pig.
Acta veterinaria Scandinavica    January 1, 1982   Volume 23, Issue 1 1-8 doi: 10.1186/BF03546815
Oltner R, Edqvist LE.Progesterone concentrations in heparinized plasma harvested immediately after blood collection were compared with levels obtained after storage of the corresponding whole blood for 2 h, 4 h, 6 h, 1 day, 2 days and 5 days at room temperature and in a refrigerator. The blood was taken during the luteal phase from 4 dogs, 4 horses, 4 pigs and 8 cows. For 4 cows the storage time was extended to 9 and 20 days. No significant effect of whole blood storage time on plasma progesterone concentrations could be shown for dogs or pigs. For the horse a slight but significant decrease was demonstrated when ...
Studies on the composition and antibacterial activity of uterine fluid from mares.
Journal of reproduction and fertility. Supplement    January 1, 1982   Volume 32 143-149
Blue MG, Brady AA, Davidson JN, Kenney RM.Forty uterine fluid samples were obtained during oestrus and dioestrus of successive cycles from 4 mares classified as resistant to bacterial infection of the uterus. The flushings were assayed for peroxidase activity and ability to influence phagocytosis by polymorphonuclear neutrophils. Uterine fluid volumes and protein concentrations and serum oestrogen values were similar during oestrus and dioestrus. Serum progesterone concentrations were depressed during oestrus. Equine neutrophils adherent to glass slides were exposed to a genital strain of Streptococcus zooepidemicus suspended in salin...
Dehydroepiandrosterone synthesis by the fetal foal and its importance as an oestrogen precursor.
Journal of reproduction and fertility. Supplement    January 1, 1982   Volume 32 389-397
Pashen RL, Sheldrick EL, Allen WR, Flint AP.The gonads of the fetal horse were found to be relatively devoid of 3 beta-hydroxysteroid dehydrogenase and other enzymes which metabolize dehydroepiandrosterone (DHA). In short-term in-vitro incubation experiments fetal liver converted DHA to the potential equilin precursor, 7 alpha-hydroxy DHA. DHA was converted to oestrone when incubated with extracts of horse placenta but 7 alpha-hydroxy DHA was not converted to equilin. Levels of DHA measured in peripheral blood of mares throughout pregnancy paralleled those of equilin and oestrone, and DHA concentrations fell rapidly after fetal gonadect...
A summary of antibody titration experiments in some animal species treated with ERA vaccine and an inactivated rabies vaccine.
Comparative immunology, microbiology and infectious diseases    January 1, 1982   Volume 5, Issue 1-3 139-141 doi: 10.1016/0147-9571(82)90027-3
Mantovani A, Caporale V, Ciuchini F, Di Trani L, Irsara A, Prosperi S.The results of antibody titrations in different animal species vaccinated against rabies are reported. The following points are considered: (1) antibody titration may be used to detect an immunity status in dogs, (2) equines should be vaccinated in infected areas, (3) experiments in progress are comparing ERA vaccine and an inactivated vaccine in bovines, and (4) the vaccination of fallow deer (Dama dama) and moufflons (Ovis ammon musimon) produced results suggesting an extension of the experiment with the purpose of vaccinating wild ruminants whenever possible.
Effect of PGF-2 alpha on LH receptors in the equine corpus luteum.
Journal of reproduction and fertility. Supplement    January 1, 1982   Volume 32 235-245
Roser JF, Evans JW, Mikuckis GM, Adams TE, Hughes JP.As quantified by Scatchard analysis, a 27 000 g crude luteal membrane fraction contained a single population of unoccupied LH receptors characterized by high affinity, ka = 0.647 +/- 0.158 X 10(11) M-1 and low binding capacity, Rt = 4.91 +/- 0.78 X 10(-11) M/mg membrane fraction. Acceptable hormonal specificity, reversibility, saturability, high affinity and tissue specificity indicated that the binding protein was a physiological receptor. To ensure that the methods used for Scatchard analysis were valid, hCG was characterized for specific activity and maximum bindability, non-specific bindin...
