Local, systemic and immunologic safety comparison between xenogeneic equine umbilical cord mesenchymal stem cells, allogeneic canine adipose mesenchymal stem cells and placebo: a randomized controlled trial.
Abstract: Mesenchymal stem cells are multipotent cells with a wide range of therapeutic applications, including, among others, tissue regeneration. This work aims to test the safety (EUC-MSC) of intra-articular administration of equine umbilical cord mesenchymal stem cells in young healthy dogs under field conditions following single and repeated administration. This was compared with the safety profile of allogenic canine adipose derived mesenchymal stem cells (CAD-MSC) and placebo in order to define the safety of xenogeneic use of mesenchymal stem cells when administered intra-articular. Twenty-four police working dogs were randomized in three groups in a proportion 1:1:1. EUC-MSCs and CAD-MSCs were obtained from healthy donors and were manufactured following company SOPs and under GMP and GMP-like conditions, respectively, and compliant all necessary controls to ensure the quality of the treatment. The safety of the treatment was evaluated locally, systemically and immunologically. For this purpose, an orthopedic examination and Glasgow test for the assessment of pain in the infiltrated joint, blood tests, clinical examination and analysis of the humoral and cellular response to treatment were performed. No adverse events were detected following single and repeated MSC administration despite both equine and canine MSC generate antibody titres in the dogs. The intra-articular administration of equine umbilical cord mesenchymal stem cells in dogs has demonstrated to be safe.

This research summary has been generated with artificial intelligence and may contain errors and omissions. Refer to the original study to confirm details provided. Submit correction.
The study compares the safety of locally injecting stem cells from horse umbilical cords and dog fat tissue into healthy working dogs. The research proved that such treatment showed no adverse effects, although the dogs started to produce antibodies in response to both types of cells.
Introduction
- The field of regenerative medicine is exponentially expanding, with mesenchymal stem cells (MSCs) being one of the most promising forms of therapy. They have extensive therapeutic potential—particularly in tissue regeneration.
- This study aimed to assess the safety of a novel treatment involving the intra-articular (within the joint) injection of equine umbilical cord mesenchymal stem cells (EUC-MSCs) into healthy dogs, with repeated administration. This was done under typical field conditions.
Methods
- To make a meaningful comparison, this EUC-MSC treatment was compared with two others: the current standard treatment using canine adipose (fat) derived MSCs (CAD-MSCs) and a placebo treatment.
- The researchers used 24 working police dogs randomized in a 1:1:1 ratio to ensure a balanced distribution of subjects across the three treatments.
- The MSCs were obtained from healthy donors and prepared according to Standard Operating Procedures (SOPs) under Good Manufacturing Practices (GMP) conditions.
- The effectiveness of the treatment was assessed through local, systemic, and immunological evaluation—we tested on the local area, the full body, and the immune response to treatment.
Results and Discussion
- They performed extensive tests, such as an orthopedic examination and the Glasgow test for assessing pain in the injected joint. Blood tests, clinical exams, and analysis of the humoral and cellular response to the treatment were also conducted.
- Remarkably, there were no adverse events following either single or repeated administration of the MSC treatment, which signifies a high level of safety associated with this treatment.
- In an interesting finding, the study showed that both the equine and canine MSCs triggered the production of antibodies in the dogs. This offers valuable insights into the dogs’ immunological response.
- Thus, the research convincingly demonstrated that the intra-articular injection of EUC-MSCs in dogs is a safe procedure.
