Observation on the morphology of contagious equine metritis bacterial colonies isolated from infected pony mares.
American journal of veterinary research    May 1, 1982   Volume 43, Issue 5 796-800
Sahu SP, Wool S, Breese SS.In uterine or cervical specimens obtained from pony mares infected with streptomycin-resistant contagious equine metritis bacteria, several colonies of the bacteria which differed in morphologic characteristics were recognized during their primary isolation on Eugon chocolate agar and tryptose chocolate agar plates. The differences were usually not observed until plates were incubated 10 to 15 days. On Eugon chocolate agar plates, smooth colony, sandy colony with rings, and colony with blebs were recognized. On tryptose chocolate agar plates, only a round smooth convex colony was observed. By ...
Isolation of a retrovirus from cultured equine sarcoid tumor cells.
American journal of veterinary research    May 1, 1982   Volume 43, Issue 5 804-806
Cheevers WP, Roberson SM, Brassfield AL, Davis WC, Crawford TB.A virus with the morphologic and biochemical properties of the family Retroviridae has been isolated from cultured cells explanted from a malignant tumor induced by intradermal inoculation of equine sarcoid cells into a combined immunodeficient Arabian foal. By electron microscopy, intracytoplasmic, extracellular, and budding particles measuring 89 to 120 nm with electron-lucent cores were seen. Virus purified from the medium of cultured cells had a buoyant density of 1.15 g/cm3 in isopycnic sucrose-gradient centrifugation, incorporated radiolabeled uridine but not thymidine, and had constitut...
Effect of glucose administration on equine fasting hyperbilirubinemia.
American journal of veterinary research    May 1, 1982   Volume 43, Issue 5 801-803
Gronwall R, Engelking LR.The effects of several treatments and their routes of administration on the reduction of hyperbilirubinemia in 9 pony mares after a 3-day fast were studied. Treatments were as follows: glucose given at doses of 1.2, 2.4, and 3.7 mg/min/kg of body weight; refeeding the base-line diet; feeding straw; and IV administration of taurocholic acid at a dosage of 0.07 mumol/min/kg. The 3 glucose dosages were each given by 3 different routes: IV, intraduodenal, and intragastric. The smallest dosage of glucose given by IV route reduced the plasma bilirubin concentration only 7%, even though other measure...
Lymphosarcoma with leukemia in a horse.
American journal of veterinary research    May 1, 1982   Volume 43, Issue 5 807-812
Madewell BR, Carlson GP, MacLachlan NJ, Feldman BF.A 17-year-old Arabian gelding was presented because of CNS depression and weight loss. Physical examination revealed generalized lymphadenopathy, pitting edema, and ascites. The diagnosis of lymphosarcoma with leukemic manifestations was based on cytologic and histopathologic examination of lymph node biopsy samples and a peripheral leukocyte count of 98,300/microliters, 94% of which were mature lymphocytes. These circulating lymphocytes lacked B-lymphocyte markers and did not respond to mitogenic stimulation, indicating functional incompetence. Though ultrastructural findings were equivocal, ...
Live temperature-sensitive equine influenza virus vaccine: generation of the virus and efficacy in hamsters.
American journal of veterinary research    May 1, 1982   Volume 43, Issue 5 869-874
Brundage-Anguish LJ, Holmes DF, Hosier NT, Murphy BR, Massicott JG, Appleyard G, Coggins L.Temperature-sensitive (ts) reassortants of an equine influenza virus, subtype A-1, were produced by mating a human influenza ts donor virus with an equine influenza A/Cornell/16/74 wild-type virus and by isolating a ts reassortant virus possessing the equine hemagglutinin and neuraminidase surface antigens. Two equine its reassortant clones, 8B1 and 71A1, were produced which had an in vitro shutoff temperature for plaque formation of 38 and 37 C, respectively. The human ts donor virus had ts mutation(s) on the polymerase 3 (P3) and nucleoprotein genes so that a ts equine reassortant virus coul...
3-methylindole-induced pulmonary toxicosis in ponies.
American journal of veterinary research    April 1, 1982   Volume 43, Issue 4 603-607
Derksen FJ, Robinson NE, Slocombe RF, Hill RE.In unanesthetized ponies, arterial blood gas tensions, pulmonary mechanics, and lung volumes were determined before and 24 to 48 hours after oral administration of 500 ml of corn oil or 100 mg of 3-methylindole (3MI)/kg of body weight in 500 ml of corn oil. In the latter group, variables were also measured after bilateral cervical vagotomy. Respiratory rate and minute ventilation were increased by 3MI treatment and decreased after vagotomy, suggesting that the tachypnea induced by 3MI was vagally mediated. The arterial O2 tension (PaO2) was unaffected but arterial CO2 tension (PaCO2) decreased...
Pulmonary function tests in standing ponies: reproducibility and effect of vagal blockade.
American journal of veterinary research    April 1, 1982   Volume 43, Issue 4 598-602
Derksen FJ, Robinson NE, Slocombe RF, Riebold TW, Brunson DB.Arterial blood gas tensions, pulmonary mechanics, and lung volumes were measured in 4 sedated ponies every hour for 6 hours and in 5 ponies 4 times at 2-month intervals to assess the short- and long-term reproducibility of pulmonary function measurements. Variability in blood gas tensions was small over the short- and long-term measurement periods, whereas the variability in total respiratory resistance and functional residual capacity was small over the short term but larger over the long term. The variability in tidal volume, minute ventilation, respiratory rate, and dynamic and quasistatic ...
Radiographic characterization of diaphragmatic excursion in halothane-anesthetized ponies: spontaneous and controlled ventilation systems.
American journal of veterinary research    April 1, 1982   Volume 43, Issue 4 617-621
Benson GJ, Manohar M, Kneller SK, Thurmon JC, Steffey EP.A radiograph technique for identification of diaphragmatic segments and quantitation of their contribution to total diaphragmatic function was developed. five anesthetized ponies were studied on 3 separate occasions. Studies were made of the ponies in left lateral recumbency at 2 anesthetic levels (1 and 2 minimal alveolar anesthetic concentrations; halothane) and under spontaneous and controlled ventilation systems. General pattern of diaphragmatic displacement was unchanged by increased depth of anesthesia. Controlled ventilation altered the pattern of diaphragmatic displacement. Diaphragmat...
Chemotaxis of horse polymorphonuclear leukocytes to N-formyl-L-methionyl-L-leucyl-L-phenylalanine.
American journal of veterinary research    April 1, 1982   Volume 43, Issue 4 613-616
Zinkl JG, Brown PD.Horse polymorphonuclear leukocytes (PMN) isolated from horse blood by sedimentation and isotonic lysis and having about 25% accompanying lymphocytes were as effective at chemotaxis as nearly pure PMN isolated by density gradient techniques. N-Formyl-L-methionyl-L-leucyl-L-phenylalanine (FMLP), used as a representative of the formylmethionyl peptides (produced by prokaryocytic organisms), was effective as a chemoattractant only at the high concentration of 10(-4) M. When serum was preincubated with FMLP at concentrations as low as 10(-8) M, the serum attracted horse PMN. This activity was not g...
Urinary production in the healthy horse and in horses deprived of feed and water.
American journal of veterinary research    April 1, 1982   Volume 43, Issue 4 735-737
Rumbaugh GE, Carlson GP, Harrold D.Total daily 24-hour urinary output was obtained from 11 healthy horses fed alfalfa hay with free access to salt during periods of high environmental temperatures. Daily urinary volume averaged 15.6 L, with mean specific gravity of 1.028, osmolality of 1,040 mOsm/kg, and urinary flow rate of 1.24 ml/kg/hr. Total 24-hour sample collections of urine were also obtained from horses held without access to feed or water for periods of 24, 48, and 72 hours during high environmental temperatures. Average urine production under these conditions was 6.3 L during the 1st day; 3.2 L, the 2nd day; and 3.0 L...
Effect of intra-articular injection of orgotein and saline solution on equine synovia.
American journal of veterinary research    April 1, 1982   Volume 43, Issue 4 594-597
Wagner AE, McIlwraith CW, Martin GS.Orgotein was injected into the right intercarpal joint of each of 8 horses; the corresponding left joint was left alone (not injected) or was given an injection of normal saline solution. Injection with orgotein caused a transient, marked inflammatory response, evidenced by clinical signs and by increased leukocytes and total protein in the synovia (synovial fluid). Leukocyte numbers and total protein concentration were increased (P less than 0.010) in the orgotein-injected joints within 24 hours. However, saline solution alone also elicited a marked inflammatory response, manifested by increa...
Renal function of the pony and the horse.
American journal of veterinary research    April 1, 1982   Volume 43, Issue 4 608-612
Zatzman ML, Clarke L, Ray WJ, Garner HE, Traver D.Simultaneous renal clearances of inulin (CIN), p-aminohippurate (CPAH), and creatinine (CCR) were measured in hydrated mares (6 ponies and 2 horses). The CIN and CPAH were determined during steady-state infusion at 3 different infusion rates. A 6-fold change in plasma IN concentration did not produce alteration in CIN, nor was there a difference between the ponies and horses (P greater than 0.2). The overall average (mean +/- SEM) was 190.6 +/- 5.89 ml . min-1 . 100 kg of body weight-1. There was no difference noted between simultaneous CIN and CPAH. Clearance of PAH remained essentially const...

Isolation of an adenovirus antigenically distinct from equine adenovirus type 1 from diarrheic foal feces.
American journal of veterinary research    March 1, 1982   Volume 43, Issue 3 543-544
Studdert MJ, Blackney MH.Adenovirus was isolated in equine fetal kidney cell cultures from the feces of 2 foals with diarrhea that also had large numbers (greater than 10(6)/g) of rotavirus particles in their feces. Unlike equine adenovirus type 1 (EAdV1), the fecal EAdV did not hemagglutinate human O, rhesus macaque, or equine RBC. By serum neutralization, the fecal viruses were identical with each other, but showed no relationship to EAdV1. Antiserum prepared against the fecal viruses did not contain hemagglutination-inhibiting antibody to EAdV1. It is proposed that the fecal viruses be considered prototypic of EAdV...
Modifications of a force plate system for equine gait analysis.
American journal of veterinary research    March 1, 1982   Volume 43, Issue 3 538-540
Steiss JE, Yuill GT, White NA, Bowen JM.A force plate system for measurement of the vertical component of the force applied by a horse's limb on ground contact was modified. The modifications included use of steel supporting posts for improved durability and an additional strain gauge on each arm of the Wheatstone bridge for increased sensitivity and temperature compensation. Data from clinically normal horses are provided to indicate the performance obtained with these modifications of the force plate system.
Retropulsion-propulsion in equine large colon.
American journal of veterinary research    March 1, 1982   Volume 43, Issue 3 390-396
Sellers AF, Lowe JE, Drost CJ, Rendano VT, Georgi JR, Roberts MC.The circular and longitudinal muscle coats of equine "midcolon" were found to be directly electrically coupled. They appear to act in concert, in healthy animals, as a pacemaker in the area of the large colon pelvic flexure, for retropulsive-propulsive myoelectrical events. The retropulsive events keep the cecum and right ventral and left ventral divisions of the colon filled, imposing a delay time for fermentation of cellulose and for bacterial protein synthesis. Point-to-point involvement of adjacent colon sections was slowed by cooling the intestinal contents with no adverse clinical signs....
Chemotaxis of radiolabeled equine neutrophils.
American journal of veterinary research    March 1, 1982   Volume 43, Issue 3 397-401
Camp CJ, Leid RW.A method for the isolation of equine neutrophils was developed using metrizamide cushions. A purity of greater than 95% was routinely obtained with greater than 90% viability. These cells were radiolabeled and tested for their chemotactic response in Boyden chambers to zymosan-activated equine serum, the partially purified equine complement component C5a, and formyl-L-methionyl-L-leucyl-L-phenylalanine. The time and ionic requirements for chemotaxis of radiolabeled equine neutrophils were investigated and maximal movement was observed at 2 hours' incubation and 1.0 mM Ca and 0.5 mM Mg. Dinitro...
Effects of dexamethasone on endotoxin shock in the anesthetized pony: hematologic, blood gas, and coagulation changes.
American journal of veterinary research    March 1, 1982   Volume 43, Issue 3 405-411
Frauenfelder HC, Fessler JF, Moore AB, Bottoms GD, Boon GD.The effects of dexamethasone (1 mg/kg of body weight) on hematologic, blood gas, and blood coagulation values in anesthetized ponies during endotoxin-induced shock were evaluated. Fifteen ponies were assigned to 3 groups of 5 ponies each: group 1, anesthetized nontreated and dexamethasone-treated controls; group 2, endotoxin, nontreated; group 3, endotoxin, dexamethasone treated. The hematologic changes in this endotoxin shock model included leukopenia and hemoconcentration. Significant hematologic effects were not seen in ponies after administration of dexamethasone. However, dexamethasone tr...
Controlled tests of fenbendazole against migrating Strongylus vulgaris in ponies.
American journal of veterinary research    March 1, 1982   Volume 43, Issue 3 541-542
Slocombe JO, McCraw BM.Sixteen pony foals were reared worm-free and inoculated with Strongylus vulgaris. On day 7 after inoculation, 12 ponies were given a fenbendazole 10% suspension at dose rate of 50 mg/kg of body weight by stomach tube. On day 8 after inoculation, 8 of these ponies were given the 2nd treatment of the anthelmintic and on day 9, 4 of these ponies were given the 3rd treatment. (The other 4 of the 16 ponies were given only tap water, as controls.) The ponies were necropsied at death or on day 28 after inoculation. Fenbendazole was effective in minimizing the appearance of clinical signs associated w...
Serum gamma-glutamyltransferase in equids: reference physiologic values.
American journal of veterinary research    February 1, 1982   Volume 43, Issue 2 339-340
Braun JP, Bardies J, Thouvenot JP, Benard P, Rico AG.Gamma-Glutamyltransferase reference values in 277 clinically healthy equine indicated a log-gaussian distribution with an upper limit of physiologic val. uea at 24 U/L. Reference values were about 2 U/L lower in males then in females and gelding, and values were slightly higher in horses used for m-ing than in horses used for riding, draft homes, and ponies. Age, preg-nancy, and season of the year seemed to have no or minor effect on serum gamma-glutamyltransferase values.
Radioimmunoassay for the detection of antigen-specific IgM, IgG, and IgA in equine sera.
American journal of veterinary research    February 1, 1982   Volume 43, Issue 2 294-298
Rearden TP, Sprouse RF, Garner HE.A radioimmunoassay was developed to discriminate immunoglobulin (Ig) classes specific for the J-5 mutant of Escherichia coli (serotype O:111-B4). Adult horses were periodically inoculated IM with a nonviable suspension of the J-5 mutant emulsified in Freund's incomplete adjuvant. Before and after the horses were inoculated, sera were collected sequentially and examined by radioimmunoassay. Rabbit anti-(horse) Ig and [125I]protein A served as the indicator system. Antigen-specific IgM, IgG, and IgA were observed to follow a classic immune response. The radioimmunoassay offers a valuable tool fo...
Effect of diethylcarbamazine on Strongylus vulgaris infection in ponies.
American journal of veterinary research    February 1, 1982   Volume 43, Issue 2 341-345
Hofing GL, Bennett DG.Shetland ponies (n = 4) were given diethylcarbamazine orally at a dose level of 22 mg/kg/day for 1 week before they were inoculated with 800 third-stage larvae of Strongylus vulgaris. Treatment was continued for 86 (1 pony) or 200 days (3 ponies) after the inoculation. As compared with the changes seen in a similarly inoculated group of ponies (group 2) which were not treated, diethylcarbamazine did not prevent the clinical or pathologic changes due to the migrating larvae. Fewer adult parasites were recovered at necropsy from treated ponies than from nontreated (group 2) ponies, even when tre...
Enzyme-linked immunosorbent assay for detection of antibody to Pseudomonas aeruginosa and measurement of antibody titer in horse serum.
American journal of veterinary research    January 1, 1982   Volume 43, Issue 1 55-60
Ueda Y, Sanai Y, Homma JY.Enzyme-linked immunosorbent assay (ELISA) was used for detection of immunoglobulin (Ig) M and IgG antibodies against a serologically common antigen (original endotoxin protein), protease, and elastase of Pseudomonas aeruginosa. The P aeruginosa antibody in horse sera was measured, using ELISA. Horseradish peroxidase-labeled rabbit anti-horse IgM and IgG antibodies were used for enzyme-labeled antibody conjugate. 5-Aminosalicylic acid and H2O2 were used for substrate. Sera collected from a vaccinated horse, a newborn foal, and 72 healthy racehorses were investigated for antibodies against P aer...
Erythrocyte volume distribution analysis in healthy dogs, cats, horses, and dairy cows.
American journal of veterinary research    January 1, 1982   Volume 43, Issue 1 163-166
Weiser MG.Erythrocyte volume distribution curves (erythrograms) were determined on a total of 300 blood samples from healthy dogs, cats, horses, and cattle (dairy cows). An index of anisocytosis was determined for these animals. Erythrograms were highly reproducible, and the mean corpuscular volumes determined from erythrograms compared well with those determined from hemograms. Bovine and equine erythrocyte volumes were found to be stable after the blood was stored at 4 C for 24 hours. Under the same conditions, canine and feline erythrocytes increased slightly in volume. After incubation of blood dilu...
Correct occlusive bladder width for indirect blood pressure measurement in horses.
American journal of veterinary research    January 1, 1982   Volume 43, Issue 1 50-54
Parry BW, McCarthy MA, Anderson GA, Gay CC.The influence of occlusive bladder width on blood pressure, measured indirectly using a doppler ultrasound technique at the middle coccygeal artery, was studied in 6 anesthetized horses. The relationship was investigated on tails with and without hair, and the optimum bladder width (BW)/tail girth (TG) ratio was determined for systolic pressure (SP) and diastolic pressure (DP), with the data grouped as unclipped tails (TT-1), clipped tails (TT-2), and both unclipped and clipped tails collectively (TTB). The optimum BW/TG ratios for SP and DP were 0.296 and 0.866 for TT-1, 0.376 and 1.156 for T...
Anthelmintic efficacy of ivermectin given intramuscularly in horses.
American journal of veterinary research    January 1, 1982   Volume 43, Issue 1 145-148
DiPietro JA, Todd KS, Lock TF, McPherron TA.The anthelmintic activity of ivermectin was evaluated in 18 female horses with naturally acquired parasitic infections. Horses were treated once (IM) with vehicle only (n = 6), 200 microgram/kg of body weight (n = 6), and 300 microgram/kg (n = 6). Efficacy of both dosages of ivermectin was greater than 99% against Gasterophilus spp, 100% against Trichostrongylus axei, Habronema muscae, H majus, and Draschia megastoma, 98% to 99% against adult cyathostomes, 86% to 97% against 4th-stage cyathostomes, and 100% against adult large strongyles. Although ivermectin was incomplete in its activity agai...
Endotoxin-induced change in hemograms, plasma enzymes, and blood chemical values in anesthetized ponies: effects of flunixin meglumine.
American journal of veterinary research    January 1, 1982   Volume 43, Issue 1 140-144
Fessler JF, Bottoms GD, Roesel OF, Moore AB, Frauenfelder HC, Boon GD.A study was made of flunixin meglumine (FM), an analgesic agent with antiprostaglandin activity, in the management of endotoxin-induced changes in ponies. Three groups of 5 ponies each were used: A--controls, B--nontreated ponies with endotoxin-induced shock, and C--ponies with endotoxin-induced shock treated with FM. Shock was induced in anesthetized ponies with IV injections of Escherichia coli endotoxin. Disruption of glucose homeostasis, insulin levels, hemograms, aerobic metabolism, and cell damage as indicated by plasma enzymes were observed. Treatment with FM (5 minutes) after shock was...
Plasma and tissue concentrations of oxytetracycline in the horse after intravenous administration.
American journal of veterinary research    December 1, 1981   Volume 42, Issue 12 2165-2166
Larson VL, Stowe CM.Oxytetracycline (OTC) was administered IV to 3 clinically normal horses at a dosage of 10 mg of OTC/kg of body weight. Plasma OTC concentrations were determined at 30-minute intervals until postinjection minute (PIM) 240. At PIM 240, the mean OTC concentration in pulmonary tissue was 3.96 microgram/g of tissue (wet weight) and in renal tissue was 25.47 micrograms/g. diluted bronchial fluid had a mean concentration of 0.288 microgram of OTC/ml at PIM 240. The data demonstrated that OTC has adequate tissue distribution in horses.
Bacteriologic examination of equine fecal flora as a diagnostic tool for equine intestinal clostridiosis.
American journal of veterinary research    December 1, 1981   Volume 42, Issue 12 2167-2169
Wierup M, DiPietro JA.The fecal flora of 56 clinically healthy and 23 sick horses were examined bacteriologically for counts of Clostridium perfringens, molds, coliforms, alpha- and beta-hemolytic streptococci, and microbes belonging to genus Bacillus, as well as for the presence of Salmonella spp. Of the healthy horses, 85.7% had a C perfringens count less than 10(1) colony-forming units/g of feces. Of the healthy horses, lowest counts were found in race-horses. Of the sick horses, equine intestinal clostridiosis was diagnosed in 2 horses with large C perfringens counts (10(4) to 10(7) colony-forming units/g) and ...
Coagulation studies f plasmas from healthy domesticated animals and persons.
American journal of veterinary research    December 1, 1981   Volume 42, Issue 12 2170-2177
Greene CE, Tsang VC, Prestwood AK, Meriwether EA.Optimal reaction conditions for the activated partial thromboplastin time (APTT), 1-stage prothrombin time (PT), and Russell's viper venom time were studied for pooled plasmas of horses, cattle, sheep, goats, swine, dogs, cats, and persons. Changes in CaCl2 and NaCl concentrations had significant effects on the APTT and PT reactions. The APTT was more sensitive than the PT to changes in CaCl2 concentration. The CaCl2 concentration recommended by the manufacturer for the APTT was suboptimal for some of plasmas of domesticated animals in ths study. Infusorial earth (Celite, activator) concentrat...
