Studies on fetal, neonatal and maternal cortisol metabolism in the mare. Fetal plasma cortisol concentrations (mean +/- S.E.M.) in ten animals with indwelling umbilical catheters ranged from 13-9 +/- 1-5 ng/ml (227 to 244 days) to 18-1 +/- 2-2 ng/ml (290 to 310 days). Maternal values did not change over this period (15-9 +/- 1-7 ng/ml). Fetal cortisol production rates in two fetuses were 3-6 and 3-8 mg/kg/day at 291 days; there was little placental transfer of cortisol. A histochemical study of steroid metabolism in the equine fetus and placenta. The distribution of several hydroxysteroid dehydrogenases (HSD) was examined in a variety of equine placental and fetal tissues. The main points of interest were: (a) the lack of dehydrogenase activity in the gonad, (b) the appearance of a variety of HSDs in the trophoblast as early as 13 days of gestation, (c) the histochemical similarity between the cells of the trophoblast and the endometrial cup, and (d) the restriction of the strong endometrial 17beta-HSD reaction to those parts of the uterine epithelium directly apposed to the trophoblast.
Mammalian erythrocyte glutathione reductase: kinetic constants and saturation with cofactor. Glutathione reductase (GR) was studied in erythrocytes of horses, cats, dogs, and man. Glutathione reductase activity was measured in hemolysates with and without preincubation of hemolysates with flavinadenine dinucleotide. The percentage saturation of GR apoenzyme with cofactor (flavin-adenine dinucleotide) was lower in cats and dogs than in horses or man. The fine structure of the glycocalyx of equine spermatozoa: a high-resolution cytochemical study. Equine spermatozoa were obtained from ejaculates of young stallions. The seminal plasma was removed and the sperm pellets washed three times with 0-15 M-NaCl solution before final centrifugation at 4500 g for 15 min. [The effect of pregnant mare’s serum on the level of K+ ions in the immature rat ovary]. Pregnant Mare Serum Gonadotrophin enhanced potassium level in immature rat ovaries in comparison with controls, three hours after intravenous injection. In vivo 42K uptake was also 36% higher (p less than or equal to 0,01) in PMSG primed rat ovaries. This response was specific to the ovary. Experiments are carried out to determine correlation between K+ level and macromolecule biosynthesis.
Periovulatory changes in peripheral plasma progesterone and estrogen concentrations in the mare. Concentrations of progesterone and estrogen were measured in peripheral blood plasma samples from mares around the time of ovulation. Samples were collected every 2 hours from 36 hours before, to 26 hours after, ovulation and assayed by radioimmunoassay. Progesterone concentrations were between 60 and 100 pg/ml for the period 24 hours before ovulation through 8 hours after ovulation. By 10 hours after ovulation, concentrations increased to 140 pg/ml and, by 26 hours after ovulation, reached 346 pg/ml. Plasma estrogen concentrations did not change significantly throughout the same period.
Bile acid kinetics and bile secretion in the pony. Horse liver alcohol dehydrogenase. A study of the essential lysine residue. 1. The inactivation of horse liver alcohol dehydrogenase by pyridoxal 5'-phosphate in phosphate buffer, pH8, at 10 degrees C was investigated. Activity declines to a minimum value determined by the pyridoxal 5'-phosphate concentration. The maximum inactivation in a single treatment is 75%. It has been found that NADPH-dependent hydroxylation of dimethylaniline, aniline, p- and o-nitroanisol and lipid peroxidation is inhibited by the tyrosine-copper (II) complex (low molecular weight analog of superoxide dismutase), which is indicative of a possibility of superoxide radicals formation in these reactions. Immunological and chemical correlation between alpha-fetoproteins from human and several mammalian species. Alpha-Fetoproteins of several animals were purified and their molecular weights, amino acid compositions and peptide maps were compared, demonstrating the close similarities. Differences in subunit composition and iron content of isoferritins. Horse spleen ferritin was fractionated into its constituent isoferritins by isoelectric focusing. Separated isoferritins were stable and showed no tendency to redistribute when re-examined by analytical gel focusing. All of the isoferritins were immunologically indistinguishable when tested with antibodies raised against unfractionated horse spleen ferritin. Myosin was isolated from leucocytes in horse arterial blood by the same procedures used for the isolation of myosin from skeletal muscle. The Ca2+-, EDTA-, and Mg2+-ATPase [EC 3.6.1.3] activities of the protein was 0.148, 0.147, and 0.001 mumoles/min/mg, respectively, in 0.5 M KCl at pH 7.0 and 25 degrees. Pregnant mare serum gonadotrophin: rate of clearance from the circulation of sheep. The process involved in the disappearance of PMSG from the blood of sheep, following a single intravenous injection, has been separated into two exponential components. Values (mean plus or minus S.E.) calculated from experiments on five animals were: metabolic clearance rate (37.8 plus or minus 1.6 ml hr-minus 1); rate constant of disposal (0.0315 plus or minus 0.0016 hr-minus 1); half-time of disposal (21.2 plus or minus 1.1 hr). The stage of the oestrous cycle, ovariectomy and the dose of PMSG used had no apparent effect on these values.
Effects of histamine and acetylcholine on equine digital lymph flow and composition. We measured the flow rate and protein concentration of lymph collected from a digital lymphatic in eight anesthetized ponies. Additionally, we recorded systemic arterial pressure (Part), and small vein pressure (Psv). Control lymph flow averaged 0.068 ml/min, and contained 3.11 g/100 ml of protein with albumin/globulin ratio of 0.75. The influence of exercise on serum enzyme levels in the horse. A group of clinically normal horses was subjected to controlled strenuous exercise. Elevated serum concentrations of lactic dehydrogenase, aldolase and creatine kinase were observed after exercise but no significant change in serum glutamic-oxalacetic transaminase was noted. These changes were reduced by repeated exposure to exercise suggesting that measurement of serum enzyme elevations, particularly creatine kinase, might be a useful index of fitness in the horse. Nitrite and nitrate pharmacokinetics in the dog, sheep, and pony. Elimination kinetics of nitrite and nitrate in the dog, sheep, and pony were determined. The elimination half-lives of nitrite were 0.499, 0.475, and 0.566 hours in the dog, sheep, and pony, respectively; those of nitrate were 44.681, 4.233, and 4.821 hours.
