Regulation of mitogen- and TCGF-induced lymphocyte blastogenesis by prostaglandins and supernatant from equine embryos and endometrium. Immunosuppressive substances which interfere with lymphocyte blastogenesis are released in vitro by embryos and endometrium from mares in early pregnancy. Immunosuppression was not evident when tissues were cultured in the presence of indomethacin (a prostaglandin-synthesis inhibitor). Various prostaglandins (PGs) were added to equine lymphocytes and lymphocyte proliferation was measured after the addition of concanavalin A (Con A) or phytohaemagglutinin A (PHA). Technique for continuous stride analysis of horses exercising on a treadmill. A technique for continuous computerised recording and analysis of the stride of horses on a treadmill is described. Advantages of the system are low cost, the use of normal shoes and the calculation of stride parameters in real time.
Amplification and differentiation of the DNA of an abortigenic (type 1) and a respiratory (type 4) strain of equine herpesvirus by the polymerase chain reaction. Unpurified DNA derived from cultures of equine fetal kidney cells infected with either equine herpesvirus type 1 or equine herpesvirus type 4 was amplified by the polymerase chain reaction using one pair of oligonucleotide primers. Restriction endonuclease digestion of the amplified segments with PvuII, followed by electrophoresis, revealed restriction fragment length polymorphisms which enabled the two virus types to be differentiated.
Fibronectin concentration in plasma of mares and neonatal foals. Plasma fibronectin concentrations were measured in clinically healthy mares and their neonatal foals, using a modified human fibronectin competitive enzyme-linked immunosorbent assay. Ranges of plasma fibronectin were established in clinically healthy horses, and the assay was reliable and reproducible. Plasma fibronectin concentrations were similar in mares and foals, both before and after colostrum ingestion.
Acute phase proteins in grass sickness (equine dysautonomia). Four acute phase proteins were assayed in the serum of normal horses and those with acute, subacute and chronic grass sickness, colic and inflammatory conditions, in order to investigate their diagnostic value in grass sickness. Electromyography of some respiratory muscles in the horse. To investigate activity in respiratory muscles, insulated wire electrodes were used to record electromyographic activity in the costal diaphragm and in the intercostal, serratus ventralis, internal abdominal oblique, transversalis and rectus abdominis muscles in conscious horses and in the same animals when anaesthetised. Isolation of equine peripheral blood mononuclear cells using Percoll. The concentration of Percoll required for isolating equine peripheral blood mononuclear cells has been reinvestigated. A poor cell yield was obtained at the 60 per cent concentration already reported. It is recommended that workers specifically interested in high yields of mononuclear cells, for investigation of lymphocyte and monocyte functions, use a concentration of 65 per cent Percoll. However, workers wishing to isolate pure populations of equine neutrophils might consider a concentration of 70 per cent in the upper layer of Percoll used to retain the mononuclear cells.
Late-stage mediators of the inflammatory response: identification of interleukin-1 and a casein-degrading enzyme in equine acute inflammatory exudates. Interleukin-1 and a casein-degrading enzyme have been identified in an experimental system for studying acute inflammation in the horse. The levels of both the cytokine and the proteinase increased over the first 24 hours following initiation of the inflammatory response, and remained at high levels through to the last sample collected at 48 hours. This is in marked contrast to prostaglandin E2 concentrations which were low initially, peaked at four to eight hours and had returned to low levels by 12 to 24 hours. Effects of an infusion of dopamine on the cardiopulmonary effects of Escherichia coli endotoxin in anaesthetised horses. Horses with colic may be endotoxaemic and subsequently develop hypotension during anaesthesia for surgical operation. The aim of this study was to evaluate the efficacy of dopamine as a means to improve cardiovascular function in anaesthetised endotoxaemic horses. Characterisation of compounds isolated from the sera of horses with acute grass sickness. Isolates were prepared from the sera of 12 horses with acute grass sickness, using methods reported to yield serum fractions associated with neurotoxicity, and their components identified by liquid chromatography and spectroscopy. All isolates were found to contain cortisol and six isolates also contained a degradation product of an analgesic drug, dipyrone. However, no recognised neurotoxin was detected.

Use of the forced oscillating airflow technique to measure the resistance of the equine upper airway: effects of laryngoventriculectomy and laryngoplasty. Investigations were carried out to determine whether measurements of total respiratory resistance (TRR) made in resting animals could detect changes due to laryngeal hemiplegia. Cartilage breakdown in equine osteoarthritis: measurement of keratan sulphate by an ELISA system. Degradation of cartilage in osteoarthritis of man results in the release of sulphated glycosaminoglycans, particularly keratan sulphate, into tissue fluids. A study was made to evaluate these markers for osteoarthritis in the horse. Effects of exercise and adrenaline on equine erythrocyte ATP content. To investigate the claim that equine erythrocytes released from the spleen are older cells than those found at rest in the circulation, the 2,3, diphosphoglycerate (2,3 DPG), adenosine triphosphate (ATP) and creatine concentration of erythrocytes before and following splenic emptying were examined. Separation of equine bronchopulmonary lavage cells by density gradient centrifugation and expression of procoagulant activity in unpurified cells and cell subpopulations. Bronchopulmonary lavage was performed in 10 healthy horses and in 39 horses with chronic pulmonary disease. The predominant cell types were macrophages in healthy horses and neutrophils in severely diseased horses. Immunoreactive prostaglandin production by equine monocytes and alveolar macrophages and concentrations of PGE2 and PGF in bronchoalveolar lavage fluid. Because of their capacity to produce prostanoids, alveolar macrophages may play a part in the pathogenesis of chronic obstructive pulmonary disease (COPD). Effects of allopurinol on endotoxin-induced increase in serum xanthine oxidase in the horse. Using a modified bovine milk enzyme kinetic assay, xanthine oxidase activity of serum collected from 34 adult, healthy horses of both sexes was determined. Enzyme activity varied from 0 to 126 mU litre-1 with a mean of 44.95 +/- 21.05 mU litre-1. The optimal pH and temperature for maximal activity were 7.8 and 28 degrees C, respectively. Effect of clenbuterol on arterial oxygen tension in the anaesthetised horse. In horses in dorsal recumbency, spontaneously breathing oxygen, with halothane at a constant end-tidal concentration, the arterial oxygen tension (PO2) increased from 9.9 +/- 0.3 SEM kPa to 21.7 +/- 4.0 kPa with 0.8 micrograms kg-1 clenbuterol and to 29.1 +/- 3.8 kPa with 2.4 micrograms kg-1 clenbuterol. Age- and position-related heterogeneity of equine tendon extracellular matrix composition. The digital flexor tendons of the neonate and adult horse have been compared with respect to variation in extracellular matrix composition along their length. Two pepsin-sensitive, acetic acid soluble proteins, molecular weight (Mr) 52 kD (np 52) and Mr 54 kD (np 54), were prominent throughout the length of neonatal tendons. Quantitative and qualitative morphology of equine peripheral nerve: teased fibre studies. Single teased fibre studies were made on samples of the lateral palmar nerve from 16 horses of mixed age and size which had no evidence of neuromuscular disease. Significant proportions of abnormal fibres indicative of axonal degeneration/regeneration and demyelination/remyelination were identified. Liquid flow and capacity of the caecum and colon of the horse. The rate of flow of fluid from the caecum and from the large colon was measured in four Shetland-type ponies fed a hay diet. In two ponies with cannulas in the caecum and at the origin of the right ventral colon, a continuous intracaecal infusion of a solution of chromium EDTA was used and samples were obtained from the cannula at the origin of the right ventral colon. Serum lipids and lipoproteins in equine colic and grass sickness. One way protection between equid herpesvirus 1 and 4 in vivo. Two groups each of six sibling ponies were exposed to sequential infections with equid herpesvirus 1 or 4 (EHV-1 or EHV-4) at four or five month intervals. This study aimed to establish a clinically reproducible method of evaluation of sensory nerve conduction in the horse and to provide reference values in a group of normal horses. Use of erythrocyte fragility profiles for monitoring immune-mediated haemolysis in horses. The fragility of erythrocytes is easily demonstrated by their ability to withstand osmotic swelling and lysis in solutions of increasingly hypotonic saline. In healthy animals a plot of percentage haemolysis against increasing hypotonicity produces a sigmoid curve. Using the same data a derivative curve calculated from haemolytic increments shows a normal distribution of fragility within samples. Relationship between mitogen receptors in peripheral blood lymphocytes and blastogenic responses to mitogen. The relationship between the optimum concentration of mitogen which induces lymphocyte blastogenic response and the receptor occupancy by mitogen was investigated. The receptor occupancies which induced maximal blastogenic activity in equine, bovine and canine peripheral blood lymphocytes (PBL) were 31.1 per cent, 26.5 per cent and 38.4 per cent with phytohaemagglutinin-P, and 48.2 per cent, 17.9 per cent and 24.5 per cent with concanavalin A, respectively. Action of dexamethasone in an equine model of acute non-immune inflammation. In a crossover study in seven New Forest ponies the actions of dexamethasone, at a dose rate of 0.06 mg kg-1 administered intravenously, were compared with those of a placebo treatment. Pathways of lymph flow from superficial tissues in the legs of horses. Pathways of peripheral lymph flow from the legs in horses were studied with casts, and with light and electron microscopic techniques. Equine postanaesthetic myositis: a possible role for free radical generation and membrane lipoperoxidation. A method for the evaluation of total plasma antihydroxyl and antiperferryl activity is described. This method was applied to horse plasma obtained during halothane anaesthesia. In horses suffering from postanaesthetic myositis, a significant decrease in the antiperferryl activity was observed during anaesthesia particularly when the muscular compression produced by the weight of the horse was released. Immunohistochemical localisation of alpha 2-macroglobulin in the horse. A peroxidase antiperoxidase technique was used to identify alpha 2-macroglobulin in formalin-fixed sections of normal equine lung and liver and in tissue sections and bronchoalveolar lavage fluid from horses with various lung diseases.
