Subjecting horse spermatozoa to hypoosmotic incubation: effects of ouabain.
Theriogenology    February 1, 1997   Volume 47, Issue 3 765-784 doi: 10.1016/s0093-691x(97)00033-2
de la Cueva FI, Rigau T, Bonet S, Miró J, Briz M, Rodríguez-Gil JE.Although hypoosmotic tests are widely used to assess spermatozoal quality in different species, they have not been used extensively in the stallion. Moreover, the role of the Na (+)K (+), ouabain sensitive-ATP-ase in the response of equine sperm to hypoosmotic shock is not well understood. This study tests two hypotheses: 1) that equine spermatozoa will respond to a hypoosmotic medium by swelling of the tail, and 2) that addition of ouabain will increase the percentage of swollen sperm tails. Ejaculates from 3 stallions were collected with an artificial vagina and diluted in Kenney's medium (T...
Ultrasonographic evaluation of the equine placenta by transrectal and transabdominal approach in the normal pregnant mare.
Theriogenology    January 15, 1997   Volume 47, Issue 2 559-573 doi: 10.1016/s0093-691x(97)00014-9
Renaudin CD, Troedsson MH, Gillis CL, King VL, Bodena A.The objective of this study was to determine normal variations in the utero placental thickness during mid- and late gestation in the mare. Normal, healthy pregnant mares (n = 9) were examined monthly from 4 mo of gestation until parturition by transrectal and transabdominal ultrasonography. At each examination, the combined thickness of the uterus and the placenta (CTUP) was measured at the placento-cervical junction (transrectally) and at the uterine body or the uterine horns (transabdominally). In addition, the echogenicity of the amniotic and allantoic fluids was evaluated by transrectal u...
The influence of progesterone-induced proteins on glucose metabolism in early equine embryos.
Theriogenology    January 15, 1997   Volume 47, Issue 2 441-456 doi: 10.1016/s0093-691x(97)00003-4
Brück I, Anderson GA, Hyland JH.The influence of different maternal plasma progesterone concentrations on embryonic glucose metabolism was studied. Uterine flushes were obtained after treating ovariectomized mares (n = 3) with 0 (control), 100 or 200 mg progesterone daily for 7 d. A group of progesterone-induced proteins (PIP) of Mr approximately 20,000 were identified in flushes from progesterone treatments by SDS-PAGE but were not observed in control flushes. Progesterone-induced proteins were removed from half the pooled flush in each treatment group by Sepharose blue CL-6B. In a 3 x 2 factorial (progesterone treatments, ...
Production of free estrogens and estrogen conjugates by the preimplantation equine embryo.
Theriogenology    January 15, 1997   Volume 47, Issue 2 457-466 doi: 10.1016/s0093-691x(97)00004-6
Choi SJ, Anderson GB, Roser JF.In vitro production of free estrogens and estrogen conjugates by intact Day 12.5, 13.5 and 14.5 equine embryos was measured at 2-h intervals over a 24-h culture period. Production of free estrogens was higher for Day 14.5 than Day 12.5 embryos. Differences in production of conjugated estrogens were not significant, but a trend toward increased production with increased age of embryo was apparent. No trend toward increased free and conjugated estrogen production per cell was observed with age. Embryo diameter and number of cells increased with age but varied considerably within groups. The amou...
Scanning electron microscopy of the equine oviduct and observations on ciliary currents in vitro at day 2 after ovulation.
Theriogenology    November 1, 1996   Volume 46, Issue 7 1305-1311 doi: 10.1016/s0093-691x(96)00302-0
Ball BA.There are considerable differences between mammalian species in the distribution and activity of ciliated cells within the oviduct, and limited information is available concerning either the distribution or activity of cilia within the equine oviduct. Patterns of ciliary activity were characterized in the ampulla and isthmus of oviducts recovered at 2 d after ovulation from 10 mares, and scanning electron microscopy was used to examine regional differences in the distribution of cilia in oviducts from 3 of these mares. Based upon the motility of 15 microm latex microspheres, ciliary activity w...
Factors related to the time of fixation of the conceptus in mares.
Theriogenology    November 1, 1996   Volume 46, Issue 7 1171-1180 doi: 10.1016/s0093-691x(96)00288-9
Gastal MO, Gastal EL, Kot K, Ginther OJ.The temporal relationships among day of conceptus fixation (cessation of mobility), conceptus diameter, uterine tone, uterine contractility, and myometrial and endometrial thickness of the middle and caudal segments of the uterine horns were assessed in 13 pony mares with fixation in the caudal segment of a uterine horn. The mean day of fixation (14.9 +/- 0.3) was established by 2-h mobility trials. Uterine tone increased (P < 0.0001) gradually over Days 11 to 21, whereas uterine contractility decreased (P < 0.0001) between Days 14 and 18. The diameter of the spherical embryonic vesicle ...
Direct transfer of equine blastocysts frozen-thawed in the presence of ethylene glycol and sucrose.
Theriogenology    November 1, 1996   Volume 46, Issue 7 1217-1224 doi: 10.1016/s0093-691x(96)00292-0
Hochi S, Maruyama K, Oguri N.The present study was designed to examine the suitability of ethylene glycol as a cryoprotectant for equine embryos. Blastocysts recovered nonsurgically from Day 6 donor mares were cryopreserved by conventional 2-step freezing in the presence of 10% ethylene glycol (EG), 10% glycerol (Gly), or 10% ethylene glycol + 0.1M sucrose (EG + Suc). After thawing, the EG and Gly were removed by a 6-step manner, and the EG + Suc was diluted to one fourth in the freezing straw. The postthaw blastocysts were transferred nonsurgically into the uteri of recipient mares on Days 4 to 7 after ovulation. Pregnan...
Seminal plasma affects membrane integrity and motility of equine spermatozoa after cryopreservation.
Theriogenology    October 1, 1996   Volume 46, Issue 5 791-797 doi: 10.1016/s0093-691x(96)00237-3
Aurich JE, Kühne A, Hoppe H, Aurich C.Effects of seminal plasma on post-thaw motility and membrane integrity of cryopreserved horse spermatozoa were investigated. Carboxyfluorescein diacetate staining was used for the assessment of sperm membrane integrity. Adding 30% of seminal plasma from stallions with high post-thaw sperm motility to ejaculates from stallions with low post-thaw sperm motility increased progressive motility from 24.0 +/- 1.6 to 34.5 +/- 1.9% (P < 0.05) and membrane integrity from 27.0 +/- 2.1 to 34.3 +/- 2.3% membrane-intact spermatozoa (P < 0.05). Conversely, the addition of seminal plasma from stallions...
SDS-PAGE characterization of the proteins in equine seminal plasma.
Theriogenology    September 1, 1996   Volume 46, Issue 4 579-591 doi: 10.1016/0093-691X(96)00210-5
Frazer GS, Bucci DM.The aims of this project were to document the protein profile of equine seminal plasma and determine the variability between stallions in the relative composition of proteins in the ejaculate. A single ejaculate was obtained from 14 stallions of varying breed and age. The gel fraction was removed by an in-line filter. The semen was centrifuged and the supernatant seminal plasma aspirated without disturbing the sperm pellet. The seminal plasma was recentrifuged and stored in cryovials at -70 degrees C. Samples were thawed, recentrifuged, assayed for protein concentration (BCA protein assay), di...
In vitro induction of acrosome reactions in stallion spermatozoa by heparin and A23187.
Theriogenology    April 15, 1996   Volume 45, Issue 6 1201-1210 doi: 10.1016/0093-691x(96)00075-1
Christensen P, Whitfield CH, Parkinson TJ.The ability of the glycosaminoglycan, heparin, and the calcium ionophore, A23187, to induce acrosome reaction in equine spermatozoa was assessed using semen from 3 warmblood stallions of known high fertility. After collection of semen, the spermatozoa were washed and incubated in vitro with heparin or A23187. Incubation periods were 0, 4, 6 or 8 h with 0, 1, 10 or 100 microg/ml heparin or 0, 10, 30 or 60 min with 0, 0.01, 0.1, 1 or 10 microM A23187, respectively. Acrosome reactions were determined by staining the spermatozoa with naphthol yellow S plus erythrosin B, and sperm viability was ass...
Testosterone secretion during early pregnancy in mares.
Theriogenology    April 15, 1996   Volume 45, Issue 6 1211-1219 doi: 10.1016/0093-691x(96)00076-3
Daels PF, Chang GC, Hansen B, Mohammed HO.We have characterized the testosterone secretion pattern during the first 80 d of pregnancy in mares and determined the sources that contribute to circulating testosterone levels during this period. Ten untreated, pregnant mares (Group 1), 10 altrenogest-treated, pregnant mares (Group 2), and 10 altrenogest-treated, pregnant mares in which the CL was eliminated by administration of PGF-2alpha on Day 16 (Group 3) were used in this study. Complete luteolysis occurred following PGF-2alpha administration in all mares in Group 3. Six of the 10 mares in Group 3 did not have an active CL until after ...
Differential suppression of endometrial prostaglandin F2alpha by the equine conceptus.
Theriogenology    February 1, 1996   Volume 45, Issue 3 541-546 doi: 10.1016/0093-691x(95)00401-s
Sissener TR, Squires EL, Clay CM.Prostaglandin F2alpha secretion by the uterine endometrium between Days 13 and 14 postovulation causes luteal regression in mares. A mechanism involving interruption or suppression of this secretion causes pregnancy to be maintained. The present study was designed to determine the age of the conceptus when maximal suppression of PGF2alpha secretion occurs. Mares were examined daily during estrus with ultrasonography (day 0 = day of ovulation). Conceptus tissues were recovered nonsurgically on Days 9 (n = 7), 12 (n = 5), 13 (n = 5), and 16 (n = 7) and uterine biopsies on Day 14. Both uterine an...
In vitro maturation and fertilization of equine oocytes recovered during the breeding season.
Theriogenology    February 1, 1996   Volume 45, Issue 3 547-560 doi: 10.1016/0093-691x(95)00402-t
Dell'aquila ME, Fusco S, Lacalandra GM, Maritato F.The aim of this study was to develope an efficient and reproducible procedure for in vitro maturation (IVM) and fertilization (IVF) in the horse. Cumulus-oocyte complexes (COCs) recovered from the ovaries of mares slaughtered during the breeding season were morphologically evaluated, and those showing a compact cumulus and homogeneously appearing cytoplasm were selected for culture. Effects on the maturation of estrous mare serum (EMS) versus estrous cow serum (ECS) as medium supplement were also evaluated (Experiment 1). In Experiment 2, the fertilization of in vitro matured oocytes with froz...
Effect of various extenders and taurine on survival of stallion sperm cooled to 5 degrees C.
Theriogenology    November 1, 1995   Volume 44, Issue 7 1039-1050 doi: 10.1016/0093-691x(95)00290-o
Ijaz A, Ducharme R.Stallion semen was diluted in five different extenders (dimitro-poulus onze (Dimitro's), Kenney's modified tryode (Kenney's), modified INRA82 (INRA82), egg yolk-citrate-taurine (Citrate) and EZ-Mixin) and evaluated for motility after cooling and storage at 5 degrees C for 0, 24, 48, 72 and 96 h. EZ-Mixin extender was used as control while 70 and 100 mM of taurine were added to Dimitro's, Kenney's and INRA82 to study its effect under conditions of storage at 5 degrees C and varying processing modifications. Motility in INRA82 was 57.0, 58.4, 61.1, and 56.1% after 24, 48, 72 and 96 h, respective...
Ultrasound-guided intrafollicular treatment in mares.
Theriogenology    November 1, 1995   Volume 44, Issue 7 1027-1037 doi: 10.1016/0093-691x(95)00289-k
Gastal EL, Kot K, Ginther OJ.A technique for intrafollicular treatment with a transvaginal ultrasound-guided injection needle was developed using equine chorionic gonadotropin (eCG) as the test substance. An injection was made into one growing follicle of a wave when the follicles were 20 to 23 mm. The treated follicles were injected with 1000 iu of eCG in 0.2 ml saline solution and control follicles were injected with 0.2 ml of the saline vehicle (10 mares per group, 1 follicle per mare). The injection system used an inner 25-gauge needle and an outer 20-gauge needle inserted together through the needle-guide channel of ...
Sex diagnosis of equine preimplantation embryos using the polymerase chain reaction.
Theriogenology    October 1, 1995   Volume 44, Issue 5 619-627 doi: 10.1016/0093-691x(95)00242-z
Peippo J, Huhtinen M, Kotilainen T.A rapid and reliable method for sex determination of preimplantation-stage equine embryos has not been available. The aim of the present study was to find an enzyme which would distinguish sexes in the horse by finding a polymorphic restriction site between the ZFY and ZFX homologues amplified by the polymerase chain reaction (PCR). Altogether, 38 different restriction enzymes were tested using female and male DNA extracted from blood. The primers used for amplification were selected from conserved sequences between human ZFY and ZFX genes and mouse Zfy-1 and Zfy-2 genes. Nine enzymes cut the ...
Relation between stallion sperm binding to homologous hemizonae and fertility.
Theriogenology    October 1, 1995   Volume 44, Issue 5 751-760 doi: 10.1016/0093-691x(95)00254-6
Fazeli AR, Steenweg W, Bevers MM, van den Broek J, Bracher V, Parlevliet J, Colenbrander B.The hemizona assay (HZA) has been developed as a diagnostic test to predict the fertilisation potential of human spermatozoa. The aim of this study was to develop an HZA for stallion spermatozoa and to investigate a possible relationship between fertility and the outcome of the HZA in this species. Equine oocytes were obtained from ovaries collected at a slaughterhouse and by transvaginal, ultrasound-guided follicle aspiration. They were then denuded from cumulus cells and stored in salt solution at 4 degrees C until use. On the day of the experiments the oocytes were bisected, thus providing ...
M-mode ultrasonic assessment of equine fetal heart rate.
Theriogenology    October 1, 1995   Volume 44, Issue 5 609-617 doi: 10.1016/0093-691x(95)00241-y
Curran S, Ginther OJ.Embryonic heart rate was studied during 87 examinations in 56 mares on Days 25 to 174 post ovulation. Real-time B-mode using a stopwatch and M-mode ultrasonography were compared. For analyses of heart rates, data were grouped into weeks or months. For the stopwatch and M-mode techniques, respectively, heart rates averaged 123 and 133 beats per minute during Month 1, peaked during Month 3 at 172 or 196 beats, then gradually decreased over the duration of the study to 126 and 130 beats during Month 6. There was a significant increase in heart rate from Months 1 to 2 and Months 2 to 3 and a signi...
Fertility control using intrauterine devices: an alternative for population control in wild horses.
Theriogenology    October 1, 1995   Volume 44, Issue 5 629-639 doi: 10.1016/0093-691x(95)00243-2
Daels PF, Hughes JP.The purpose of this study was to develop a contraceptive method for feral horses. The feral horse population has increased significantly in recent years despite attempts to control numbers. As in most wild animal population control programs, contraceptive methods must be easy to apply, cause minimal disruption to the social structure and be fully reversible. In the present study, we tested the effectiveness of an intrauterine device (IUD) for fertility control in mares. Six mares were fitted with a silastic O-ring-shaped IUD on July 1 of Year 1. The IUD-treated mares were turned out with 12 no...
In vitro maturation of equine oocytes obtained from different age groups of sexually mature mares.
Theriogenology    September 1, 1995   Volume 44, Issue 4 461-469 doi: 10.1016/0093-691x(95)00218-w
Brinsko SP, Ball BA, Ellington JE.Oocytes were harvested from mare ovaries obtained at slaughter and were divided into 3 groups based on the age of the donor. The age groups consisted of young (2 to 7 yr), middle-aged (8 to 14 yr) and aged (>or=15 yr) mares. There were no differences between age groups in the proportions of follicles available for examination or the proportions of normal, abnormal or total oocytes collected. After 24 h of culture, the overall maturation rate to the second metaphase (MII) was 52.7%. Maturation rates for oocytes obtained from young and middle-aged mares were similar, but oocytes from aged mar...
Selection of follicles, preculture oocyte evaluation, and duration of culture for in vitro maturation of equine oocytes.
Theriogenology    May 1, 1995   Volume 43, Issue 7 1141-1153 doi: 10.1016/0093-691x(95)00086-n
Del Campo MR, Donoso X, Parrish JJ, Ginther OJ.Equine oocytes (n = 537) were collected from slaughterhouse ovaries (n = 118 mares) by scraping the internal follicular wall. Preculture record was made of the appearance of oocyte investments (no cumulus, corona radiata only, compact cumulus, expanded cumulus), appearance of cytoplasm (homogeneous, condensed heterogeneous/fragmented), and nuclear maturation stages (germinal vesicle, germinal-vesicle breakdown, metaphase I, metaphase II, degenerated). There was no difference between follicles > 30 mm and follicles < or = 30 mm in the preculture frequency distribution among the 5 nuclear ...
Morphology and location of attached follicular cumulus-oocyte complexes in horses, cattle and llamas.
Theriogenology    February 1, 1995   Volume 43, Issue 3 533-542 doi: 10.1016/0093-691x(94)00058-3
Del Campo MR, Del Campo CH, Mapletoft RJ, Ginther OJ.Morphology and location of the attached cumulus-oocyte complex (COC) were studied in slaughter-house ovaries in horses (49 follicles, 9 to 44 mm), cattle (68 follicles, 6 to 18 mm), and llamas (38 follicles, 3 to 14 mm). The expected point of ovulation was marked, using the ovulation fossa in mares and the center of the projecting follicular surface in cattle and llamas. A follicle was dissected from an ovary, and tissue was removed from the follicle until the COC became visible by transillumination. However, most llama follicles protruded prominently from the ovarian surface so that dissectio...
In vitro fertilization rate of horse oocytes with partially removed zonae.
Theriogenology    October 1, 1994   Volume 42, Issue 5 795-802 doi: 10.1016/0093-691x(94)90448-r
Choi YH, Okada Y, Hochi S, Braun J, Sato K, Oguri N.Frozen-thawed ejaculated stallion spermatozoa were preincubated for 3 h in BO medium containing 5 mM caffeine and then treated with 0.1 micro M calcium ionophore A23187 for 60 sec. Aliquots of the sperm suspension (final concentration 1-2 x 10(7)/ml) were added to the oocytes which had been matured in vitro for 32 h. In Experiment 1, there were 3 groups of oocytes; cumulus intact, denuded zona-intact, and zona-free. Cumulus cells were removed with 0.5% hyaluronidase and the zona pellucida with 0.1% protease. The oocytes were fixed 20 h after insemination with acetic acid:ethanol (1:3) and stai...
The effect of insemination volume on pregnancy rates of pony mares.
Theriogenology    September 1, 1994   Volume 42, Issue 4 571-578 doi: 10.1016/0093-691x(94)90374-r
Bedford SJ, Hinrichs K.It has recently been reported that large insemination volumes might affect fertility of mares. The results from these studies are confounded by other factors, however, such as inadequate number of spermatozoa in the inseminate. We conducted a study to test whether volume alone affects fertility when sufficient numbers of spermatozoa are present. Semen from one stallion was collected, extended at 50 x 10(6) spermatozoa/ml, and stored in a commercial semen cooling device for 18 to 30 h before insemination. Ten pony mares were assigned during estrus in random pairs to be bred every other day with...
Sperm-induced leukocytosis in the equine uterus.
Theriogenology    February 2, 1994   Volume 41, Issue 3 629-636 doi: 10.1016/0093-691x(94)90173-g
Kotilainen T, Huhtinen M, Katila T.The objective of this study was to investigate the inflammatory reaction induced in the equine uterus by insemination with fresh and frozen semen. Eleven groups (6 to 8 mares per group) were studied during 2 breeding seasons. The mares were inseminated using raw semen, frozen semen, extended fresh and frozen semen, concentrated fresh semen, seminal plasma and seminal extenders only. One group was bred naturally. Six hours after insemination, the uteri were flushed with 50 ml of phosphate-buffered saline (PBS). Seventeen out of 104 samples (16%) exhibited slight bacterial growth. Neutrophil con...
Follicular and FSH responses to parturition during the anovulatory season in mares.
Theriogenology    February 2, 1994   Volume 41, Issue 3 613-627 doi: 10.1016/0093-691x(94)90172-f
Ginther OJ, Baucus KL, Bergfelt DR.The ovaries of periparturient pony mares (n=9 to 16 parturitions per month for January to April) were scanned ultrasonically on the day of parturition, while those of postpartum and control mares (n=12) were examined at least twice weekly. Four mares had apparent lactational anovulation (incidence, 7%) that corrected spontaneously (1 mare) or within 14 d after the weaning of foals on August 10 (3 mares). All but 2 of the postpartum ovulations occurred after April 29; that is, parturition did not effectively stimulate ovulation in ponies foaling during the anovulatory season. Mean diameter of t...
Natural outcome and ultrasonic identification of equine fetal twins.
Theriogenology    January 1, 1994   Volume 41, Issue 5 1193-1199 doi: 10.1016/s0093-691x(05)80041-x
Ginther OJ, Griffin PG.The natural outcome of bilateral twins (one in each uterine horn) that were viable on Day 40 was studied in 15 pony mares. Fetuses were monitored by transrectal ultrasonography until the outcome was determinable. One fetus (2 mares) or both fetuses (8 mares) died (cessation of heartbeats) during Months 2 and 3. Loss of fetuses was externally observed in 4 additional mares during the late fetal stage (Months 8 to 11); 2 mares lost 1 fetus and 2 lost both fetuses. Birth of 2 live foals occurred in only 1 of 15 mares (7%) First day of detected apposition between portions of the 2 allantochorions ...
Binding of epidermal growth factor and transforming growth factor-alpha in mammalian preimplantation embryos.
Theriogenology    January 1, 1994   Volume 41, Issue 4 879-887 doi: 10.1016/0093-691x(94)90503-b
Fischer B, Rose-Hellekant TA, Sheffield LG, Bertics PJ, Bavister BD.Preimplantation embryos of the pig (Days 11 to 15), cow (Days 14 to 16), sheep (Day 14) and pony (Day 16) bind epidermal growth factor (EGF) specifically. Binding was not detected in embryos of the rabbit at Day 5 or 6 or the hamster at Day 3. Transforming growth factor-alpha displaced [(125)I] EGF in pig, cow and pony embryos almost as much as unlabeled EGF. The binding affinities of EGF ranged from 12 to 233 pM in pig and cow embryos. The range of species and binding features indicate that the EGF family may play a significant role in mammalian preimplantation development.
Pregnancies following transfer of equine embryos cryopreserved by vitrification.
Theriogenology    January 1, 1994   Volume 42, Issue 3 483-488 doi: 10.1016/0093-691x(94)90686-d
Hochi S, Fujimoto T, Braun J, Oguri N.The objective of this study was to investigate the in vitro and in vivo developmental abilities of equine embryos cryopreserved by vitrification. Twenty-eight embryos were recovered from Native pony and Thoroughbred mares at Days 5 to 7 by nonsurgical uterine flushing (detection of ovulation=Day 0). The vitrification solution contained 40% ethylene glycol, 18% Ficoll, and 0.3 M sucrose in PBS. The embryos were placed for 1 to 2 min in vitrification solution (Group 1) or following exposure to 20% ethylene glycol in PBS for 10 to 20 min (Groups 2 and 3). Single embryos were loaded in 0.25-ml str...
Post-thaw motility and longevity of motility of imipramine-induced ejaculates of pony stallions.
Theriogenology    January 1, 1994   Volume 42, Issue 3 475-481 doi: 10.1016/0093-691x(94)90685-c
McDonnell SM, Oristaglio Turner RM.Imipramine-induced ex copula ejaculates (11) and fractionated in copula ejaculates were collected from each of 5 pony stallions for freezing in 5-ml straws (6), using a modified Kenney glucose skim-milk extender (2). Initial post-thaw total and progressive motilities and daily post-thaw total and progressive motilities, as well as the number of days to reach 0 progressively motile spermatozoa, were also similar for the 2 methods of collection. The percentage of morphologically normal spermatozoa both before freezing and after thawing were also similar for in copula and ex copula ejaculates. Co...
