The study of cells in horses encompasses the examination of various cell types and their functions within the equine body. Cells are the basic structural and functional units of life, and in horses, they contribute to numerous physiological processes, including growth, repair, and immune responses. Different cell types, such as red blood cells, white blood cells, and muscle cells, each perform specific roles that are vital for maintaining the health and homeostasis of the horse. This topic includes research on cellular mechanisms, cellular responses to disease or injury, and the application of cellular biology in equine medicine. This page compiles peer-reviewed research studies and scholarly articles that explore the structure, function, and significance of cells in equine biology and health.
Higgins AJ, Lees P.Arachidonic acid is a polyunsaturated fatty acid covalently bound in esterified form in the cell membranes of most body cells. Following irritation or injury, arachidonic acid is released and oxygenated by enzyme systems leading to the formation of an important group of inflammatory mediators, the eicosanoids. It is now recognised that eicosanoid release is fundamental to the inflammatory process. For example, the prostaglandins and other prostanoids, products of the cyclooxygenase enzyme pathway, have potent inflammatory properties and prostaglandin E2 is readily detectable in equine acute in...
Gunson DE, Halliwell RE, Minor RR.A 2-year-old female horse had large areas of hyperextensible, fragile skin that were interspersed with areas of normal skin. Affected skin tore easily and contained reduced amounts of dermal collagen. Collagen fibers were fragmented and disorganized, and in trichrome-stained sections, many fibers had abnormal red-stained centers. Electron microscopy showed that many collagen fibers had discrete foci of degradation in which the fibrils were fragmented, loosely packed, and widely separated by granular material. Collagen fibril fragments were present in secondary lysosomes in dermal fibroblasts, ...
Bouillant AM, Becker SA.The successive steps of maturation of seven retroviruses from five species of farm animals and one retrovirus from a mouse were compared in cell cultures. The viruses included three type C oncoviruses, one spumavirus, and three lentiviruses. Although members of the 3 subfamilies shared some gross morphologic features such as budding on plasma membranes, core, and surface projections, differences were noted in the ultrastructural detail of these features. Type C oncoviruses did not show any structural differentiation in identifiable form in the cytoplasm as opposed to characteristic features ob...
Lindberg R.A morphological study on equine granulomatous enteritis (EGE) in 13 horses is presented. All horses were young (1 to 5 years old) standardbreds. Based on gross pathology, cases were classified into those with diffuse (11 cases) and those with localized (2 cases) small bowel lesions. The granulomatous reaction in the gut was marked by diffuse and patchy infiltrates and distinct granulomas, composed of epithelioid cells, macrophages and lymphoid cells, and was devoid of necrosis. Salient accompanying features of the small bowel included lymphoid hyperplasia, peri-lymphatic and transmural inflamm...
Kitamura N, Yamada J, Calingasan NY, Yamashita T.Endocrine cells immunoreactive for somatostatin, gastrin, glicentin, glucagon, secretin, cholecystokinin, motilin and neurotensin were identified immunocytochemically in the gastrointestinal mucosa of the horse. Somatostatin-, glicentin- and glucagon-immunoreactive cells were very numerous in the cardiac and fundic regions of the stomach, whereas most gastrin-immunoreactive cells were confined to the pyloric region. Somatostatin-immunoreactive cells also were detected in all portions of the small intestine while gastrin-immunoreactive cells were confined exclusively to the upper portion and gl...
Hegre OD, Marshall S, Hickey GE.Plasma amine oxidase activities (benzylamine oxidase and spermine oxidase) were determined in the sera of a number of species of various ages. Benzylamine oxidase (BZO) activity, measured spectrophotometrically, was present in bovine, equine, and ovine species examined. Generally its activity in serum increased with the age of the animal. Spermine oxidase activity (SPO) was estimated by a bioassay of in vitro toxicity and did not necessarily correlate with BZO. Cytotoxicity in the presence of spermidine was found only in the sera of the ruminant species examined. Serum activity tended to rise ...
Gibson SJ, Polak JM, Anand P, Blank MA, Morrison JF, Kelly JS, Bloom SR.The distribution of VIP-immunoreactivity was studied in the spinal cord and dorsal root ganglia of 6 mammalian species. Immunoreactive fibres and cell bodies were most apparent in the dorsal horn, dorsolateral funiculus, intermediolateral cell columns and the area around the central canal. The distribution of VIP immunoreactivity was similar in all species studied, mouse, rat, guinea pig, cat, horse and the marmoset monkey. There were fewer VIP fibres in the dorsal horn of cervical and thoracic segments than in lumbosacral segments. Using radioimmunoassay this gradient increase was quantitativ...
Henderson LL, Warren JC.Estradiol 17 alpha-dehydrogenase (EC 1.1.1.148) and estradiol 17 beta-dehydrogenase (EC 1.1.1.62) from horse placenta have been purified to homogeneity. Both enzymes are localized in the microsomal fraction and are solubilized in 1.5% sodium cholate. The 17 alpha- and 17 beta-dehydrogenases are separated by selective elution from hydroxylapatite with 0.5 and 1.0 M potassium phosphate, respectively. Subsequent purification is achieved by two affinity-absorption steps using reactive blue 2-agarose and estriol hemisuccinate-Sepharose. Homogeneous estradiol 17 alpha-dehydrogenase has a specific ac...
Desmadril M, Mitraki A, Betton JM, Yon JM.The unfolding-folding transition of phosphoglycerate kinase induced by GuHC1 was studied at equilibrium. Various signals were used to follow the transition: fluorescence emission, difference spectra, circular dichroism and enzymatic activity. The non-coincidence of transition curves obtained from different structural parameters indicate a deviation from a two-state process. The view that structural domains behave as independent "folding units" is critically discussed.
Staczek J, Wharton JH, Dauenhauer SA, O'Callaghan DJ.Semipermissive, primary hamster embryo (HE) cells were morphologically transformed in vitro by infection with UV-irradiated equine cytomegalovirus (equine herpesvirus type 2; ECMV). Cell lines (designated EC-1-3) were established independently from foci and were shown to exhibit growth and biological properties typically associated with transformed cells: altered morphology, loss of contact inhibition, increased saturation density, decreased generation time, immortality in culture, normal growth in low concentrations of serum, colony formation in soft agar, and resistance to ECMV superinfectio...
Bailey E, Antczak DF, Bernoco D, Bull RW, Fister R, Guerin G, Lázary S, Matthews S, McClure J, Meyer J.The Second International Workshop on Lymphocyte Alloantigens of the Horse was held 3-8 October 1982. At this workshop, the 6 specificities identified at the first workshop were confirmed and an additional 5 new specificities were identified and given workshop nomenclature. Four of the new specificities, products of the ELA locus, were named ELA-W7, W8, W9, and W10. An additional specificity, designated ELY-2.1, is the product of a locus independent of the ELA locus. Cell isolation methods were compared at this workshop. Technical variation in methods clearly affected reactivity of many reagent...
Sławomirski J, Flieger S, Jastrzebski M, Boratyński Z.The studies carried out on 2 spinal cords of horses showed that cells of the medial motor nucleus (nucleus motorius medialis) are present in all neuromers of the lumbar and sacral segment of the spinal cord. It lies in the medial part of grey matter of the ventral column, neighbouring laterally and ventrally with cells of the lateral motor nucleus, whereas dorsally with cells of the nucleus of the ventral commissural horn. Along the nucleus numerous constrictions and intervals are found, which are connected with various numbers of nerve cells in particular cross-sections.
García-Sacristán A, Casanueva CR, Castilla C, Labadia A.The presence and types of alpha and beta adrenergic receptors in the urethra and prostate of the horse were studied in vitro using adrenergic agonist and antagonist drugs. The existence of these receptors was shown. This finding was based on the observation that the contractile action was mediated by adrenergic receptors of alpha-1 type, although in the prostate alpha-2 type receptors also participated. Relaxation in both tissues was controlled by receptors of the beta-2 type.
Lucke VM, Lane JG.The clinical and pathological aspects of two cases of C-cell (parafollicular cell) tumours of the thyroid are described. Both the horse and the pony presented with a paralaryngeal mass and a history of constant gulping. Ultrastructural examination of the tumours demonstrated that they were composed of C-cells containing typical, membrane-bound secretory granules. The pony is alive and well three years after surgery and the horse has raced successfully following removal of the tumour.
Flieger S, Sławomirski J, Boratyński Z, Jastrzebski M.Two medullae oblongatae of horses were cut into 15 microns cross-sections and stained according to the modified method of Nissel. The lateral motor nucleus lies in the lateral and median part of the ventral column of spinal cord grey matter. It adjoins medially nucleus motorius medialis of the spinal ventral column. Cells of this nucleus occur both along the whole lumbar and sacral segment of the spinal cord. In the lateral motor nucleus three cell groups are distinguished - median, basal and lateral. The latter is divided in some segments into subgroups - dorsal and ventral. Along the nucleus...

Williams IF, McCullagh KG, Silver IA.During tissue response to injury the glycoproteins fibronectin and Type III collagen are synthesized in increased amounts. We have studied the distribution of these molecules in the healing tendon at various times after injury by comparison with that of the major constituent of normal tendon, Type I collagen. Immunofluorescent localization demonstrated the presence of fibronectin throughout the tendon within one week after injury. Staining was found in the matrix, both around capillaries and around fibroblast-like cells. Fibronectin was still apparent in the healing tendon at one month after i...

Stratil A, Tomásek V, Bobák P, Glasnák V.Homozygous horse transferrin (Tf O) is highly heterogeneous. In starch gel electrophoresis it gives at least 9 zones. Two main components (2a and 4b) were purified by rivanol and ammonium sulphate precipitation, DEAE-Sephadex chromatography and SP-Sephadex chromatography. Molecular weights of 75 200 and 80 500 for components 2a and 4b, respectively, were determined by sedimentation equilibrium ultracentrifugation. Amino acid compositions of the two components were similar, and there were no differences in the N-terminus (glutamic acid followed by glutamine) and the C-terminus (valine). Differe...
Leach DH, Oliphant LW.Annular gap junctions interiorized within cells of the stratum spinosum of the coronary border of the equine hoof were degraded by two methods. Some were autophagocytized and some appeared to fuse with lysosomes to form heterophagosomes. Structural changes of partially degraded annular gap junctions included increased density of the enclosed cytoplasm, formation of filamentous or membrane-like material within the annular gap junction, and disruption of the circular or oval profile of the gap junction. The annular gap junctions are apparently incorporated into the fully keratinized cells of the...
Frost ML, Colton SW, Wertz PW, Downing DT.The C34, C36, and C38 dienoic omega-lactones were isolated from sebum of the horse (Equus caballus) and the double bond positions were determined by stepwise chemical dissection and analysis of the fragments. The structures found could be formed by delta 9-desaturation at the C18-stage of fatty acid biosynthesis followed by a second delta 9-desaturation when the chains reached C24, C26, C28, C30, or C32 and then addition of one to seven 2-carbon units. These findings provide insight into the dimensions and organization of the endoplasmic reticulum in cells of the sebaceous glands.
Mengual R, Leblanc G, Sudaka P.The present study determines the characteristics of isotopic Na and lactate exchange under equilibrium conditions in horse kidney brush-border membrane vesicles. The influence of one solute (Na+ or lactate) on the isotopic exchange of the co-transported species (lactate or Na) was analyzed in detail. Analysis of the data suggests that Na and lactate interact sequentially with the carrier. The observed apparent symmetry between the activating effect of low Na concentrations and the inhibiting effect of high Na concentrations on the lactate exchange process suggests that the carrier functions ac...
Sayers DE, Theil EC, Rennick FJ.Cell-specific variations in apoferritin structure correlate with variations in iron metabolism that suggest functional specificity of the protein shell. Using EPR spectroscopy, we previously showed that vanadyl binds to specific sites on apoferritin, and that VO2+ binding is reduced by Fe(II) and Fe(III) (the natural substrates) and by metals known to influence iron storage (Chasteen, N. D., and Theil, E. C. (1982) J. Biol. Chem. 257, 7672-7677). Such observations suggest that the metal-binding site is important to apoferritin function and may define a location where the influence of cell-spec...
Dyer RM, Liggitt HD, Leid RW.A device was constructed from an equine nasogastric tube, polyethylene tubing, and a 3-way stopcock and used to lavage the lungs of anesthetized ponies. The technique was safe and atraumatic in that 6.4 to 19.7 X 10(7) purified alveolar macrophages were removed from the lungs without harm to the ponies or contamination of the samples with blood. Studies of these highly purified cell suspensions revealed a mean viability of 85% as assessed by eosin dye exclusion with a mean recovery (+/- SD) of 12.5 +/- 4.8 X 10(7) pulmonary alveolar macrophages/pony.
Magnuson NS, Decker DM, Perryman LE.The effect of adenosine, deoxyadenosine, guanosine, and deoxyguanosine on the growth rate of fibroblasts derived from normal horses, horses heterozygous for the severe combined immunodeficiency (SCID) trait (heterozygotes), and horses with SCID was studied. All four purines were found to inhibit growth in a dose-dependent manner, but only adenosine and deoxyadenosine were inhibitory at concentrations of less than 100 microM. No statistical difference in sensitivity to adenosine was detected between normal and SCID fibroblasts. Fibroblasts from SCID horses were, however, more sensitive to the g...
Huang Y, Ko WH, Chung YW, Wong PY.The patch-clamp recording technique was used to examine the properties of the K+ channels in cultured equine sweat gland epithelial cells. With symmetric K+ solutions (140 mM), a single population of K+ channels was identified with a slope conductance of 187 pS and a reversal potential of around 0 mV. The channel was selective for K+ over Na+. Channel activity was increased by membrane depolarization. A 10-fold increase in [Ca2+]i produced an approximate 60 mV negative shift in the open state probability (Popen)-voltage curve. Externally applied tetraethylammonium ions (TEA+) caused a rapid an...
