Topic:Embryo Transfer

Embryo transfer in horses is a reproductive technology that involves collecting a fertilized embryo from a donor mare and implanting it into a recipient mare, which then carries the pregnancy to term. This technique allows for the production of multiple offspring from a single mare within a breeding season. The procedure includes several stages: synchronization of the donor and recipient mares' estrous cycles, collection of the embryo, and its subsequent transfer to the recipient mare. Embryo transfer is utilized to enhance genetic dissemination, preserve valuable genetics, and manage breeding schedules. This page aggregates peer-reviewed research studies and scholarly articles that explore the methods, efficiency, and applications of embryo transfer in equine reproduction.
Marked blastomere herniation is the only post-thaw morphological characteristic associated with a reduced pregnancy rate for in vitro produced equine blastocysts.
Theriogenology    December 30, 2024   Volume 235 1-8 doi: 10.1016/j.theriogenology.2024.12.028
Arnold LC, Stout TAE, Beitsma MM, Deelen CC, Rizzo M, Leemans B, Galli C, Colleoni S, Lazzari G, Barandalla M, Claes ANJ.It is not known whether the likelihood of pregnancy after transfer of cryopreserved equine in vitro produced (IVP) blastocysts is related to post-thaw morphological characteristics, or could be impacted by other factors such as embryo biopsy prior to cryopreservation. This study examined associations between post-thaw morphological appearance of equine IVP embryos, and biopsy prior to slow-freezing, on the likelihood of pregnancy. Biopsy of IVP blastocysts was performed by harvesting cells herniated through the intra-cytoplasmic sperm injection (ICSI)-derived hole in the zona pellucida (ZP). M...
Birth of a Healthy Monozygotic Twin Foal with Hydrops and a Dead Co-Twin.
Veterinary sciences    December 13, 2024   Volume 11, Issue 12 649 doi: 10.3390/vetsci11120649
Peere S, Van den Branden E, Broothaers K, Polfliet E, Smits K, Govaere J.Transfer of in vitro-produced (IVP) equine embryos is associated with a higher incidence of monozygotic multiple pregnancies, but this remains a rarity. The outcome of monozygotic twin gestation is poor, and good management techniques are lacking. In addition, hydrops conditions are exceptional in horses, with hydroallantois occurring more frequently than hydramnion. Most hydrops conditions will lead to compromise or death of the foal and mare. A 6-year-old multiparous standardbred recipient mare, 332 days pregnant, diagnosed with a monozygotic twin and hydrops with a dead co-twin, gave birth ...
Improved embryo recovery through increased flushing media volume and the impact on unintended pregnancies in a single subject donor mare.
Journal of equine veterinary science    November 29, 2024   105246 doi: 10.1016/j.jevs.2024.105246
Sala-Ayala L, Cuervo-Arango J, Martinez-Bovi R.The case report discusses the implications of using different volumes of flushing media during embryo flushing (EF) procedures in a single donor mare that experienced multiple unintended pregnancies following negative EF outcomes. The study involved two sets of EF experiments over two years on a non-lactating Spanish purebred mare. The initial experiments (group 1, n=6) used 1L of flushing medium (Ringer´s Lactate), and subsequent tests (group 2, n=5) increased the volume to 2L, to assess whether a larger volume could enhance embryo recovery rates. After every EF procedure, no PGF2α was admi...
Periovulatory anticoagulant therapy enhances embryo recovery rates in superovulated mares.
Theriogenology    October 22, 2024   Volume 231 182-189 doi: 10.1016/j.theriogenology.2024.10.018
Rodrigues LT, Segabinazzi L, Frasson M, Dell'Aqua C, Carmo MT, Takahira RK, Alvarenga M, Papa F.Although protocols for superovulation have been described in horses, this technique has been discouraged due to the low embryo recovery rates in superovulated mares. The reason for these poor results is poorly understood, but the formation of a blood clot in the ovulation fossa following ovulations has been hypothesized. Therefore, this study aimed to assess the safety and effect of periovulatory anticoagulant therapy on embryo recovery of superovulated mares. In experiment 1, five mares were assigned to receive five anticoagulant treatments in a crossover design: intravenous injections of 150...
A new hormonal protocol supports early development of in vitro-produced embryos after transfer to anoestrus mares.
Reproduction in domestic animals = Zuchthygiene    October 14, 2024   Volume 59 Suppl 3 e14622 doi: 10.1111/rda.14622
Muñoz-García CC, Luis-Calero M, Fernández-Hernández P, Pizarro-Hernández A, Fernández-González R, Gutiérrez-Adán A, Sánchez-Calabuig MJ....The present study aimed to evaluate whether primed anoestrus mares are suitable recipients for embryos produced by intracytoplasmic sperm injection (ICSI). Anoestrus was confirmed in four mares and daily doses of oestradiol benzoate (6 mg in total) over 5 days were administered; after 3 days of rest, oral altrenogest was administered at 0.088 mg/kg and embryos (1 to 5 embryos per mare; 15 in total) were transferred 3.5 days after progesterone onset. Uterine lavage was conducted 48 h after transfer. The results revealed an 80% embryo recovery rate, and among the retrieved embryos, 6...
Effect of refreezing extender on stallion sperm quality and embryo production after intracytoplasmic sperm injection.
Theriogenology    October 9, 2024   Volume 231 29-35 doi: 10.1016/j.theriogenology.2024.10.006
Brito LFC, Felix MR, Linardi RL, Martinez de Andino EV, Balamurugan NS, Hernández-Avilés C, Hinrichs K.Intracytoplasmic sperm injection (ICSI) is a valuable assisted reproduction technology in clinical practice, especially when semen availability is limited. Since the number of sperm required per ICSI cycle is much less than the number of sperm available in a standard straw of frozen semen, refreezing semen at lower sperm concentrations could yield multiple straws for ICSI use. However, there is little data on the effect of sperm refreezing on ICSI outcomes, especially on the effect of extender used for refreezing. The objective of the present study was to evaluate the effect of refreezing exte...
A novel method for equine embryo transfer from contaminated recipient mares into second healthy recipients for surviving embryos.
Journal of equine veterinary science    September 19, 2024   Volume 142 105200 doi: 10.1016/j.jevs.2024.105200
Derbala MK, Abu-Seida AM, El-Metwally AE, Asfour HAE.Embryo transfer (ET) is regarded as one of the reproductive methods for genetic improvement initiatives in the horse business. The current study aimed to increase the success rate of ET by re-flushing and re-transferring of embryos from contaminated recipient mares into second healthy recipients. The study involved 25 recipient mares that demonstrated embryonic vesicles inside intraluminal fluid (ILF) on ultrasound scanning 2 ± 0.5 days after ET. Seven recipient mares were left without re-flushing and re-transferring of embryos (control group) and 18 recipient mares (treated group) were subje...
Cervix-Deep Rectal Temperature Differential on the Day of Ovulation is Correlated With Embryo Recovery Results in Mares.
Reproduction in domestic animals = Zuchthygiene    August 31, 2024   Volume 59, Issue 9 e14716 doi: 10.1111/rda.14716
Funes J, Ismer A, Hoogewijs M, Wilsher S.Variations in temperature throughout the reproductive tract have been noted in many species. A recent study found the cervix-rectum temperature differential (CR-TD) in cattle was related to fertility. The present study aimed to assess the CR-TD in mares around the time of ovulation and relate it to embryo recover. Over 52 cycles, mares were inseminated with a fertile stallion and embryo recovery was undertaken on Day 7 post ovulation. Further 10 control mares were not inseminated. Rectal and cervical temperatures were measured using a precision thermometer on four or five occasions: the day of...

Animal reproduction    August 5, 2024   Volume 21, Issue 3 e20240029 doi: 10.1590/1984-3143-AR2024-0029
Embryonic stem cells (ESCs) have proven to be a great model that faithfully recapitulates the events that occur during embryogenesis, making them a unique tool to study the cellular and molecular mechanisms that define tissue specification during embryonic development. Livestock ESCs are particularly attractive and have broad prospects including drug selection and human disease modeling, improvement of reproductive biotechniques and agriculture-related applications such as production of genetically modified animals. While mice and human ESCs have been established many years ago, no significa...

Pregnancy and Luteal Responses to Embryo Reinsertion following Embryo Flushing in Donor Mares.
Animals : an open access journal from MDPI    May 29, 2024   Volume 14, Issue 11 doi: 10.3390/ani14111605
Martínez-Boví R, Gaber JTH, Sala-Ayala L, Plaza-Dávila M, Cuervo-Arango J.The effect of embryo reinsertion immediately after embryo flushing was studied. In Experiment 1, eight mares were used during 32 cycles (8 cycles in each group). For the first two groups, inseminated mares were flushed 8 days after ovulation and prostaglandin F2α was not administered: in group EF-ET (embryo flushing and embryo transfer) the embryo was reinserted in the same donor mare, while in the EF group, no further procedure was performed. In the third group (ET), non-inseminated mares (recipients) received a Day 8 embryo. Progesterone concentration was measured before EF/ET and 72 h afte...
Effect of duration of estradiol exposure on embryo survival and endometrial gene expression in anestrous embryo recipient mares.
Theriogenology    May 28, 2024   Volume 226 1-9 doi: 10.1016/j.theriogenology.2024.05.039
Sant'Anna Monteiro da Silva E, Sanches Oquendo Júnior P, Gaspari Oquendo FM, Stout TAE, de Ruijter-Villani M, Rodrigues TS, Beletti ME....Previous studies indicate a positive correlation between the duration of estrus prior to ovulation and likelihood of pregnancy in embryo recipient mares. However, the mechanisms by which the duration of estrus before may affect fertility remains unclear. This study aimed to determine the effect of different durations of estradiol exposure, prior to progesterone administration, on embryo viability in anestrous recipient mares, and endometrial expression of genes thought to influence embryo survival. Three groups of anestrous recipient mares treated with different duration of estradiol were used...
The effect of uterine massage and number of embryo flushing attempts on embryo recovery in mares.
Theriogenology    May 15, 2024   Volume 224 94-101 doi: 10.1016/j.theriogenology.2024.05.017
Sala-Ayala L, Martínez-Boví R, Querol-Paajanen A, Cuervo-Arango J.The aims of this study were to determine the effect of the embryo flushing technique and the number of flushing attempts performed by operators of different experience on embryo recovery (ER). Ten non-lactating mares were inseminated with the same stallion in six cycles each (n = 60). Embryo flushing (EF) was performed 7-9 days after ovulation by three operators (OP; 20 EF cycles each): OP1 had performed >500 EF before the study, while OP2 and 3 had performed 0 EF. Each EF was performed with 2 flushing attempts (FA) using 1L of ringer's lactate "in-and-out" using two EF techniques: 1) ...

Journal of equine veterinary science    May 1, 2024   Volume 137 105076 doi: 10.1016/j.jevs.2024.105076
Sielhorst J, Koether K, Volkmann N, Blanco M, Vicioso R, Baade S, Kemper N, de Mestre AM, Sieme H.Little is known about the incidence and outcome of high-risk pregnancies in equine practice and clinical studies on spontaneous occurring placentitis cases and treatments are missing. Therefore, the aims of this retrospective field study were to (1) describe the incidence and severity of ultrasonographic assessed placental abnormalities (UPA) in 4,192 pregnancies on a large commercial warmblood stud farm in 2017 - 2019 and (2) characterize these UPA cases and their pregnancy outcome. UPA severity (Placental abnormality score (PSc) 1-3; low to high), nine treatment regimens (TM1-9) used in UPA ...
The ability of donkey sperm to induce oocyte activation and mule embryo development after ICSI.
Theriogenology    February 2, 2024   Volume 218 200-207 doi: 10.1016/j.theriogenology.2024.02.002
Arroyo-Salvo C, Cogollo Villarreal MY, Clérico G, Flores Bragulat AP, Niño Vargas A, Castañeira C, Briski O, Alonso C, Plaza J, Zeledon JM....Members of the Equus genus exhibit a fascinating capacity for hybridization, giving rise to healthy offspring. Mules, resulting from the mating of a mare with a jack, represent the most prevalent equid hybrid, serving diverse roles in our society. While in vitro embryo production, particularly through Intracytoplasmic Sperm Injection (ICSI), has rapidly gained significance in domestic horses, the in vitro production in other equids remains largely unexplored. Utilizing donkey sperm for fertilizing horse oocytes not only addresses this gap but also provides an opportunity to investigate donkey ...
Cloned Foal Born from Postmortem-Obtained Ear Sample Refrigerated for 5 Days Before Fibroblast Isolation and Decontamination of the Infected Monolayer Culture.
Cellular reprogramming    January 23, 2024   Volume 26, Issue 1 33-36 doi: 10.1089/cell.2023.0076
Cortez JV, Hardwicke K, Grupen CG, Herrid M, Machaty Z, Vajta G.A 6-year-old mare, a valuable polo horse, died of complications following postcolic surgery. To preserve its genetics, ear skin samples were collected immediately after death and stored in an equine embryo transfer medium at 4°C for 5 days. After trypsin digestion, monolayer fibroblast cultures were established, but signs of massive bacterial infection were found in all of them. As an ultimate attempt for rescue, rigorously and repeatedly washed cells were individually cultured in all wells of four 96-well dishes. New monolayers were established from the few wells without contamination and us...

Morelli KG, Lourenço GG, Marangon VR, Feltrin IR, Imura Oshiro TS, Rodrigues da Silva IV, Pugliesi G.We aimed to evaluate the impact of corpus luteum (CL) and uterine characteristics accessed by B-mode and Color-Doppler ultrasonography in recipient mares at the time of embryo transfer (ET) on pregnancy outcomes. Recipient mares (n = 110), between days 3-9 after spontaneous ovulation, received a fresh embryo. Immediately before ET, the reproductive system was assessed by transrectal palpation for the following parameters: uterine tone (0-3), CL echogenicity (0-6), CL type (homogeneous, trabecular or anechoic center), luteal area (cm2), uterine echogenicity (0-3), uterine edema (0-3), luteal ...
Embryo Pulsing: Repeated Expansion and Contraction of In Vivo and In Vitro Equine Blastocysts.
Journal of equine veterinary science    July 8, 2023   Volume 128 104891 doi: 10.1016/j.jevs.2023.104891
de la Fuente A, Omyla K, Cooper C, Daels P, Meyers S, Dini P.Morphokinetic evaluation of embryo development has allowed the discovery of events occurring during blastulation. Here, we describe equine embryo pulsing, determined as continued expansion and contraction of both in vivo and in vitro produced blastocysts. Using time-lapse imaging, we demonstrated that pulsing starts during early blastocyst development of in vitro-produced embryos in horses. The median time for a complete contraction was 0.22h (0.08h-2h; min-max) where embryos reduced their sizes around 12.0% (median; 2.3%-27.0%) and the median time for an expansion was 3.3h (0.75-9.0h) where e...
Comparative reproduction of the female horse, elephant and rhinoceros: implications for advancing Assisted Reproductive Technologies (ART).
Reproduction & fertility    July 1, 2023   Volume 4, Issue 3 doi: 10.1530/RAF-23-0020
Meuffels-Barkas J, Wilsher S, Allen WRT, Ververs C, Lueders I.Recent loss of rhinoceros subspecies has renewed interest in using more advanced assisted reproductive technologies (ART) in rhinoceroses and elephants. Currently, only semen collection, semen preservation and artificial insemination (AI) have been used repeatedly with success in these species. Although ovum pick-up (OPU) and intra-cytoplasmic sperm injection (ICSI) have been reported in rhinoceroses, the techniques are not yet optimised. In contrast, multiple ART applications are routinely used in the horse. Since elephant and rhinoceroses share some reproductive features with equids, we post...
Horse Somatic Cell Nuclear Transfer Using Zona Pellucida-Enclosed and Zona-Free Oocytes.
Methods in molecular biology (Clifton, N.J.)    April 12, 2023   Volume 2647 269-281 doi: 10.1007/978-1-0716-3064-8_15
Salamone D, Maserati M.Horse cloning by somatic cell nuclear transfer (SCNT) is an attractive scientific and commercial endeavor. Moreover, SCNT allows generating genetically identical animals from elite, aged, castrated, or deceased equine donors. Several variations in the horse SCNT method have been described, which may be useful for specific applications. This chapter describes a detailed protocol for horse cloning, thus including SCNT protocols using zona pellucida (ZP)-enclosed or ZP-free oocytes for enucleation. These SCNT protocols are under routine use for commercial equine cloning.
Cloning horses by somatic cell nuclear transfer: Effects of oocyte source on development to foaling.
Theriogenology    March 23, 2023   Volume 203 99-108 doi: 10.1016/j.theriogenology.2023.03.018
Cortez JV, Hardwicke K, Cuervo-Arango J, Grupen CG.The cloning of horses is a commercial reality, yet the availability of oocytes for cloned embryo production remains a major limitation. Immature oocytes collected from abattoir-sourced ovaries or from live mares by ovum pick-up (OPU) have both been used to generate cloned foals. However, the reported cloning efficiencies are difficult to compare due to the different somatic cell nuclear transfer (SCNT) techniques and conditions used. The objective of this retrospective study was to compare the in vitro and in vivo development of equine SCNT embryos produced using oocytes recovered from abatt...

The Journal of reproduction and development    January 31, 2023   Volume 69, Issue 2 125-128 doi: 10.1262/jrd.2022-125
Rajabi-Toustani R, Tsogtgerel M, Gao Y, Li C, Sakato M, Haneda S, Cheong SH, Nambo Y.Japanese native horses, which consists of 8 breeds, are threatened with extinction. Embryo transfer (ET) is used to reproduce endangered animals in various mammalian species. We aimed to perform ET using native ponies from Kiso and Hokkaido as donors and recipients, respectively. ET operation included long-distance transport of non-cryopreserved embryos from Nagano Prefecture to Hokkaido. Embryos were transported 1500 km over 9 h in a container maintained at 22°C. After transferring two embryos to two recipients, one mare delivered a healthy live foal. These results demonstrated that reciproc...
Time-lapse imaging and developmental competence of donkey eggs after ICSI: Effect of preovulatory follicular fluid during oocyte in vitro maturation.
Theriogenology    November 1, 2022   Volume 195 199-208 doi: 10.1016/j.theriogenology.2022.10.030
Flores Bragulat AP, Ortiz I, Catalán J, Dorado J, Hidalgo M, Losinno L, Dordas-Perpinyà M, Yánez-Ortiz I, Martínez-Rodero I, Miró J, Gambini A.Equus members exhibit very divergent karyotype, genetic plasticity, and significant differences in their reproductive physiology. Despite the fact that somatic cell nuclear transfer and intracytoplasmic sperm injection (ICSI) has gained relevance in the last few years in horses, few reports have been published exploring ovum pick up (OPU) and in vitro maturation (IVM) of cumulus-oocyte complexes (COCs) in donkeys. Yet, some donkey species and breeds are considered endangered, and these assisted-reproductive technologies could help to preserve the genetic of valuable individuals. In this study...
Lower expression of the equine maternally imprinted gene IGF2R is related to the slow proliferation of hinny embryonic fibroblast in vitro.
Molecular biology reports    November 1, 2022   Volume 50, Issue 1 185-192 doi: 10.1007/s11033-022-07937-6
Wang X, Asgenbaatar N, Shen Y, Yi M, Zhao B, Ren H, Davshilt T, Ulaangerel T, Wang M, Burenbaatar A, Tian S, Li B, Dugarjav M, Bou G.Proliferation of embryonic fibroblasts under the same cell culture conditions, hinny embryonic fibroblasts (HiEFs) was slower than horse embryonic fibroblast (HEFs), donkey embryonic fibroblasts (DEFs) and mule embryonic fibroblasts (MuEFs). The imprinted genes IGF2 and IGF2R are important for cell proliferation. Therefore, we investigated whether the slower proliferation of HiEFs is related to an aberrant gene expression of IGF2 or its receptors or genes influencing the expression of the IGF2 system. Results: Real-time polymerase chain reaction, immunofluorescence and cell starving experiment...
Sperm factors associated with the production of equine blastocysts by intracytoplasmic sperm injection (ICSI) using frozen/thawed semen.
Theriogenology    October 17, 2022   Volume 195 85-92 doi: 10.1016/j.theriogenology.2022.10.014
Ramírez-Agámez L, Hernández-Avilés C, Varner DD, Love CC.Intracytoplasmic Sperm Injection (ICSI) using frozen/thawed sperm is a common procedure to obtain embryos from fertile or subfertile mares and stallions. Stallion-associated factors that impact the efficiency of ICSI have been studied less than those associated with the mare. Three experiments were conducted: Experiment 1: the effect of freezing extender composition and cryoprotectant; Experiment 2: the effect of sperm exposure to seminal plasma prior to freezing (ejaculated vs. epididymal sperm; two-freeze/thaw cycles each); and Experiment 3: the effect of sperm morphologic feature used for f...
Insulin-like growth factor system components expressed at the conceptus-maternal interface during the establishment of equine pregnancy.
Frontiers in veterinary science    September 13, 2022   Volume 9 912721 doi: 10.3389/fvets.2022.912721
Gibson C, de Ruijter-Villani M, Stout TAE.In many species, the insulin-like growth factors (IGF1 and IGF2), their receptors and IGF binding proteins play important roles in preparing the endometrium for implantation, and regulating conceptus growth and development. To determine whether the IGF system may contribute to conceptus-maternal interaction during equine pre-implantation development, we evaluated mRNA expression for IGF system components in conceptuses, and endometrium recovered from pregnant and cycling mares, on days 7, 14, 21 and 28 after ovulation. We also investigated expression of IGF1, IGF2 and their receptors 6 and 11 ...
Detection of Indiscriminate Genetic Manipulation in Thoroughbred Racehorses by Targeted Resequencing for Gene-Doping Control.
Genes    September 4, 2022   Volume 13, Issue 9 doi: 10.3390/genes13091589
Tozaki T, Ohnuma A, Nakamura K, Hano K, Takasu M, Takahashi Y, Tamura N, Sato F, Shimizu K, Kikuchi M, Ishige T, Kakoi H, Hirota KI, Hamilton NA....The creation of genetically modified horses is prohibited in horse racing as it falls under the banner of gene doping. In this study, we developed a test to detect gene editing based on amplicon sequencing using next-generation sequencing (NGS). We designed 1012 amplicons to target 52 genes (481 exons) and 147 single-nucleotide variants (SNVs). NGS analyses showed that 97.7% of the targeted exons were sequenced to sufficient coverage (depth > 50) for calling variants. The targets of artificial editing were defined as homozygous alternative (HomoALT) and compound heterozygous alternative (AL...
