Characterization of Berne virus genomic and messenger RNAs. From 380S particles of Berne virus (proposed family Toroviridae) one species of polyadenylated RNA was isolated. Using agarose gel electrophoresis its length was estimated as 20 kb or greater. When assayed under hypertonic transfection conditions genomic RNA was found to be infectious; RNase treatment destroyed the infectivity. Evaluation of equine locomotion during different degrees of experimentally induced lameness. I: Lameness model and quantification of ground reaction force patterns of the limbs. Force plate data from walking horses were used to evaluate the locomotion of a group of six horses during experimentally induced lameness in a forelimb or a hindlimb. By tightening or loosening screws in modified horseshoes the resulting pressure pain on the sole enabled induction and release of three different degrees of supporting lameness within a period of 2 h. The effect of dietary protein level on exercising horses. Six mature Quarter Horse mares were used in a crossover design to assess the effect of dietary protein level on metabolic response to exercise. After a 2-wk adaptation period to either a 12.9% (control) or an 18.5% CP (high-protein) diet, each mare performed a standard exercise test. The test consisted of a 15-min exercise period at 4.5 m/sec on a 9% grade motorized treadmill. Projectile dart foreign body in a horse. A 3-year-old Welsh pony mare was referred for evaluation of right hind limb lameness. The lameness developed 4 months after the pony had been captured by use of a projectile dart syringe to administer a tranquilizer. The lameness was attributable to a projectile dart foreign body. The dart was removed after induction of local anesthesia, and the lameness improved. The use of projected syringes has inherent risk, and clients should be advised before they are used.
Excision of cutaneous tumors in the horse using histologic guidance. A highly successful surgical technique for removing cutaneous tumors in humans was used in seven horses with cutaneous squamous cell carcinoma (n = 3) or sarcoid (n = 4). In humans, the complete underside of the tumor is evaluated histologically by processing horizontal sections from the tumor base, and orientation between the wound surface and the undersurface of the excised tumor is maintained by mapping both surfaces. Deletion analysis of the equine infectious anemia virus long terminal repeat revealed that sequences responsive to virus-specific transactivation are located within the region spanning the transcriptional start site (-31 to +22). In addition, an active exon of a trans-acting factor (tat) was identified downstream of pol and overlapping env (nucleotides 5264 to 5461). Activation by tat is accompanied by an increase in the steady-state levels of mRNA directed by the equine infectious anemia virus long terminal repeat.
Arthrodesis of the equine distal tarsal joints by perforated stainless steel cylinders. Perforated stainless steel cylinders filled with autogenous cancellous bone were implanted in the distal tarsal joints of four horses. Graft cell survival was poor two weeks after surgery in one horse. In two horses, at 10 months there was partial arthrodesis of the joints with incorporation of the implants into the osseous union. The implants were filled with vascularised woven bone. These two horses were sound nine months after surgery. One horse fractured its third tarsal bone and was still positive to a hock flexion test 12 months after surgery.
Partial arytenoidectomy in the horse with and without mucosal closure. Left-sided partial arytenoidectomy was performed in eight horses to evaluate healing. Four horses underwent conventional partial arytenoidectomy with suture apposition of the mucosa. Characterisation of IgE-mediated histamine release from equine basophils in vitro. In vitro IgE-mediated histamine release by equine blood basophils was characterised as the basis for a screening test for immediate hypersensitivity responses in horses. The responses are initiated by inducing agents that are capable of crosslinking or bridging the membrane-bound IgE molecules. The release process is complete within 40 mins. In vitro histamine release is dose-dependent, with a submaximal response at less or greater than the optimal dose of inducing agent. Curettage of subchondral bone cysts in medial femoral condyles of the horse. Fifty-one horses and unilateral or bilateral femorotibial arthrotomy with medial condyle bone cyst curettage. A total of 60 cyst were treated. Treated horses responded to the surgery three to 12 months postoperatively. In 42 horses lameness was reduced or eliminated, enabling them to return to an remain in their intended function. Of these 42 horses 35 are sound, four of which have not started training, and seven showed partial improvement, two of which had no long term follow up. Immunohistochemical study of the local humoral immune system of the equine respiratory mucosa. An indirect immunoperoxidase technique was used to demonstrate both free immunoglobulin and immunoglobulin-containing plasma cells of IgG, IgA, and IgM classes in the mucosa of the equine respiratory tract. The effect of detomidine (Domosedan) on the maintenance of equine pregnancy and foetal development: ten cases. Detomidine was administered throughout 10 pregnancies in eight mares. An intravenous injection of 20 micrograms/kg body weight was given weekly from Day 14 to Day 60 of gestation and thereafter every four weeks until parturition. Monitoring the effects of treadmill exercise on bone by non-invasive means during a progressive fitness programme. Six Standardbred horses underwent a 14-week training programme on a treadmill. The training schedule consisted of five weeks of slow work of six to 12 km/day at 5 m/sec.
