Topic:Serological Surveys

Serological surveys for horses involve the collection and analysis of blood samples to detect the presence of antibodies or antigens related to infectious agents. These surveys are instrumental in understanding the epidemiology of diseases within equine populations, helping to identify exposure to pathogens such as viruses, bacteria, and parasites. Through serological testing, researchers can assess the prevalence of diseases, track their spread, and evaluate the effectiveness of vaccination programs. Common serological tests used in equine studies include the enzyme-linked immunosorbent assay (ELISA) and the virus neutralization test. This page compiles peer-reviewed research studies and scholarly articles that explore the methodologies, findings, and implications of serological surveys in equine health management.
Resistance of castrated male horses to attempted establishment of the carrier state with equine arteritis virus.
Journal of comparative pathology    November 1, 1994   Volume 111, Issue 4 383-388 doi: 10.1016/s0021-9975(05)80096-9
McCollum WH, Little TV, Timoney PJ, Swerczek TW.Twelve geldings all became infected when inoculated intranasally with the KY-84 strain of equine arteritis virus (EAV), a strain previously shown to be capable of establishing the carrier state in the stallion. With the exception of one animal that showed no effects other than pyrexia, all of the geldings developed clinical signs characteristic of equine viral arteritis (EVA). The geldings were febrile for varying periods within the range of 2-10 days after inoculation. Viraemia occurred from day 2 onwards, for periods varying from 9 to at least 19 days. Nasal shedding of virus began 2-4 days ...
Titers to Leptospira species in horses in Alberta.
The Canadian veterinary journal = La revue veterinaire canadienne    October 1, 1994   Volume 35, Issue 10 636-640
Lees VW, Gale SP.Sera from horses in Alberta, submitted to Agriculture and Agri-Food Canada for routine testing for equine infectious anemia from January 1987 to June 1989, were tested for antibody against 13 serovars of pathogenic Leptospira spp., using the microscopic agglutination test. The purpose of the study was to investigate the prevalence of serum titers to those serovars in horses in Alberta, and to analyze the associated risk factors. Descriptive statistics were compiled and logistic regressions were computed. Titers to L. interrogans serovars icterohaemorrhagiae, bratislava, copenhageni, and autumn...
Serological relationship between a donkey alphaherpesvirus (isolate M7/91) and equid herpesvirus type 1 and 4.
Journal of the South African Veterinary Association    June 1, 1994   Volume 65, Issue 2 64-66
Paweska JT, Gerdes T, Van Heerden J.Rabbit hyperimmune serum prepared against a donkey alphaherpesvirus isolate (M7/91), and against EHV-1 and EHV-4 was used to characterise the antigenic relationship between these 3 viruses. Serum from immunised rabbits was always more specific for homologous virus and showed different cross reactivity for heterologous virus. It was concluded that the immunologic relationship between the M7/91 isolate and EHV-1, was closer than that between this isolate and EHV-4. A serological survey of donkeys (n = 116) and horses (n = 57) revealed evidence of the presence of neutralising antibody to M7/91 in...
Use of the serum neutralisation test for equine viral arteritis with different virus strains.
The Veterinary record    May 28, 1994   Volume 134, Issue 22 574-576 doi: 10.1136/vr.134.22.574
Fukunaga Y, Matsumura T, Sugiura T, Wada R, Imagawa H, Kanemaru T, Kamada M.Serum cross neutralisation tests were conducted with a recent American isolate (84KY-A1) and a European isolate, (Wroclaw-2) and compared with the prototype and modified viruses of the Bucyrus strain of equine arteritis virus by using virus specific immune horse sera. The modified Bucyrus strain was neutralised and showed high neutralisation titres with all the immune sera. The prototype Bucyrus strain was also substantially neutralised, followed by the 84KY-A1 strain. As a result of the tests with the modified Bucyrus strain as the antigen, 20 seropositive horses were discovered among home-br...

Borrelia burgdorferi infection in UK horses.
Equine veterinary journal    May 1, 1994   Volume 26, Issue 3 187-190 doi: 10.1111/j.2042-3306.1994.tb04367.x
Carter SD, May C, Barnes A, Bennett D.Antibody levels (IgG and IgM) to Borrelia burgdorferi were measured in the sera and synovial fluids of UK horses. Western blotting against B. burgdorferi was also used on samples from seropositive horses. A low incidence of seropositivity was shown in horses from most parts of the UK. This increased in areas that have a high incidence of human and canine borreliosis (Norfolk and south coast). Leptospira infections of horses did not cause cross reactions in the B. burgdorferi ELISA. Most horses did not display clinical signs of Lyme disease. As with dogs and man, it is apparent that B. burgdorf...
Identification of diagnostic antigens for South American Babesia caballi infections.
International journal for parasitology    April 1, 1994   Volume 24, Issue 2 255-258 doi: 10.1016/0020-7519(94)90034-5
Böse R, Peymann B, Barbosa IP.Sera from 60 horses held in breeding herd in Brazil were examined monthly by ELISA, immunofluorescence antibody test (IFAT) and Western blot. All foals had maternal antibodies detectable by ELISA and IFAT, and sero-conversion took place between the 2nd and 5th month of age. The 48 and 50 kDa antigens were recognized first in the course of infection. Of 79 sera taken after sero-conversion 78 reacted with the 48 kDa antigen, 76 with the 50 kDa, 50 with the 70 kDa, 54 with the 112 kDa, 72 with the 141 kDa antigen. In general, sera from horses older than 1 year reacted with all 5 diagnostic antige...
Serological and microbiological findings on 3 farms with equine leptospiral abortions.
Equine veterinary journal    March 1, 1994   Volume 26, Issue 2 105-108 doi: 10.1111/j.2042-3306.1994.tb04345.x
Williams DM, Smith BJ, Donahue JM, Poonacha KB.Blood and urine samples from horses on 3 central Kentucky horse farms with prior histories of leptospiral abortions were analysed. Blood samples were obtained from all available horses on each farm and tested for antibodies to 6 leptospira serovars. Urine samples were collected from non-gravid mares with serum antibody titres > or = 1:800 and examined for leptospires by dark-field microscopy, fluorescent antibody testing and culture. Adult horses had the greatest serological evidence of exposure to leptospira, followed by yearlings, then foals. Of horses with anti-leptospiral antibodies, 76...
[Rhinopneumonia and equine viral arteritis: seroepidemiological study in the northeast of Tunisia].
Archives de l'Institut Pasteur de Tunis    January 1, 1994   Volume 71, Issue 1-2 5-12
Ghram A, Chabchoub A, Turki I, Boussetta M, Ibn Amor H, Ghorbel A.A seroepidemiological survey was realized in the Nord-Est Tunisia to study the prevalence of complement fixing and neutralizing antibodies to equine rhinopneumonitis and viral arteritis of horse, respectively. Four hundred sera were tested, using complement fixation reaction and seroneutralization test. The results show that 8.75% of sera have antibodies to viral arteritis and only 1.25% are positive for equine rhinopneumonitis.

Escartin-Peña M, Bautista-Garfias CR.Sera from 41 horses and 159 donkeys, from twelve States of México, were tested to ascertain anti-Gasterophilus circulating antibodies by double immunodiffusion (DD), counterimmunoelectrophoresis (CIE), indirect haemagglutination (IH), thin layer immunoassay (TIA) and diffusion-in-gel ELISA (DIG-ELISA) methods using crude somatic antigen from third instar larvae of G. intestinalis (DeGeer). At necropsy, 33/41 horses and 24/159 donkeys were found to be parasitized by G. intestinalis and/or G. nasalis (L.). Gasterophilus intestinalis was the species most commonly found in the equines. Analysis o...
Circulation of eastern equine encephalitis, western equine encephalitis, Ilhéus, Maguari and Tacaiuma viruses in equines of the Brazilian Pantanal, South America.
Revista do Instituto de Medicina Tropical de Sao Paulo    July 1, 1993   Volume 35, Issue 4 355-359 doi: 10.1590/s0036-46651993000400009
Iversson LB, Silva RA, da Rosa AP, Barros VL.Neutralizing antibodies to EEE (6.7%), WEE (1.2%), ILH (26.6%), MAG (28.2%) and TCM (15.7%) viruses were found in sera of 432 equines of the Brazilian Pantanal, area where undiagnosed horse deaths are frequently observed. A 4-fold rise in CF titer to EEE virus was detected in acute and convalescent sera of an encephalitis horse sacrificed in 1992. Antibodies to EEE, ILH, MAG and TCM viruses were detected in horses less than 2 years old indicating recent circulation of these viruses in the Pantanal. The evidence of recent equine encephalitis associated with rising CF titer to EEE warrants a mor...
Serological evidence of equine arteritis virus in donkeys in South Africa.
The Onderstepoort journal of veterinary research    June 1, 1993   Volume 60, Issue 2 155-158
Paweska JT, Barnard BJ.This paper reports the first serological evidence of exposure of donkeys to equine arteritis virus. Seven hundred and thirty-four serum samples collected between 1989 and 1992 from donkeys in different areas of South Africa were examined for the presence of antibodies against this virus by a microneutralization test. Seventeen percent of serum samples tested positive. The distribution of seropositive animals varied from none in the western Cape Province and the Transvaal Highveld to 30% in the northern Transvaal. The country-wide distribution of serologically positive donkeys suggests a longst...

Miyamoto C, Takashima I, Karaiwa H, Sugiura T, Kamada M, Hashimoto N.To investigate the overall prevalence of chlamydial infections in light (i.e. non-draught) horses in Japan, 599 sera obtained from 12 localities in 1991 were tested for complement fixation antibodies. The mean antibody positive rates of the all sera were 15.2% (91/599) and the regional positive rates were higher in Honshu (19.1%, 48/251) and Kyushu (20.0%, 20/100) than in Hokkaido (9.3%, 23/248). In Honshu, the highest rate (56.0%, 28/50) was observed in Utsunomiya. Analysis of the positive rate in different age groups showed that the 2-5 years age-group had the highest prevalence of chlamydia...
Haemagglutination-inhibiting antibodies against African horse sickness virus in domestic animals in Nigeria.
Veterinary research    January 1, 1993   Volume 24, Issue 6 483-487
Baba SS, Olaleye OD, Ayanbadejo OA.A sero-epidemiological survey of African horse sickness (AHS) virus in 261 animals which included 96 camels, 81 horses, 80 dogs and 4 donkeys was carried out in Nigeria. The animals had no history of vaccination against AHS. Sera were tested by the haemagglutination-inhibition (HI) test for the presence of antibody against AHS virus. Of these, 77 (95.1%) horse, 4 (100%) donkey, 10 (10.4%) camel and 28 (35%) dog sera samples tested were recorded as positive. The prevalence of antibody in samples taken from horses in different regions was similar. The prevalence of antibody to AHS virus detected...
Renal dysfunction associated with infection of Leptospira interrogans in a horse.
Journal of the American Veterinary Medical Association    November 1, 1992   Volume 201, Issue 9 1391-1392
Divers TJ, Byars TD, Shin SJ.Renal failure associated with infection of Leptospira interrogans was detected in a horse. Fever, leukocytosis, pyuria, isosthenuria, and azotemia were suggestive of an inflammatory urinary tract disease. Despite persistent pyuria, no bacteria were found during routine microscopic examinations or bacteriologic culturing of urine. A fluorescent antibody examination of the urine was positive for L interrogans. Serologic testing during a 6-month period, supported an acute infection with L interrogans serovar pomona. Treatment with intravenously administered fluids and antimicrobials resulted in c...
Seroprevalence of antibodies to Borrelia burgdorferi in a population of horses in central Texas.
Journal of the American Veterinary Medical Association    October 1, 1992   Volume 201, Issue 7 1030-1034
Cohen ND, Heck FC, Heim B, Flad DM, Bosler EM, Cohen D.Four hundred sixty-nine serum samples were obtained from horses admitted to the internal medicine service of the Texas Veterinary Medical Center between Jan 1 and Dec 31, 1990. Serum samples were tested by ELISA for antibody to Borrelia burgdorferi. Of these 469 samples, 1 (0.2%) was repeatedly seropositive for the organism by ELISA. Confirmatory testing by protein immunoblot was negative. The observed seroprevalence was 0%; the upper bound of the 95% confidence interval was 0.6%. These findings indicate the evidence of infection with B burgdorferi is presently uncommon in horses in central Te...

Binepal VS, Wariru BN, Davies FG, Soi R, Olubayo R.A survey was carried out in horse, zebra, elephant, camel, sheep and goat and wild carnivore sera for virus-serum neutralising antibody to the nine type strains of African horse sickness virus. Antibody was found amongst the horse, zebra and elephant sera to all nine different strains. No antibody was detected in any sera from camels, sheep and goats. None was found in sera from hyaena and jackals in this series but had been detected earlier.
One-dimensional isoelectric focusing and immunoblotting of equine major histocompatibility complex class I antigens.
Animal genetics    January 1, 1992   Volume 23, Issue 2 87-95
Schuberth HJ, Anders I, Pape U, Leibold W.The cells of 60 randomly selected Hannoveranian warm-blooded horses were subjected to one-dimensional isoelectric focusing and immunoblotting with a cross-reacting monoclonal antibody (Bo 1) recognizing bovine class I antigens. The banding patterns were correlated with the serologically defined specificities of the ELA-A locus. ELA-A2 was correlated with four bands, while ELA-A5, ELA-W18, ELA-A6, ELA-A14 and ELA-A9 were correlated with a single band each. The complexity of the pattern and additional polymorphic bands which could not be correlated to any of the known ELA specificities may indic...
Prevalence of complement-fixing antibody to the African horse sickness virus in domestic animals in Nigeria.
Beitrage zur tropischen Landwirtschaft und Veterinarmedizin    January 1, 1992   Volume 30, Issue 4 471-477
Baba SS, Akinyele HA, Olaleye OD.The occurrence of antibodies against the African horse sickness virus was investigated in 246 domestic animals (horses, donkeys, camels, dogs) in various regions of Nigeria by means of the complement-fixing rate. 34% of the sera tested were positive: 75% in donkeys, 68% in horses, 19% in camels, and 9% in dogs. Among the horses, those of 6 to 15 years of age had higher than average prevalence rates than the other age groups. Stallions from the northern regions had higher prevalence rates than mares generally and stallions from other regions. These findings are important for the epidemiology of...
Detection of adenovirus precipitating antibodies in the sera of Polo horses in Nigeria.
Revue d'elevage et de medecine veterinaire des pays tropicaux    January 1, 1992   Volume 45, Issue 1 21-22
Adeyefa CA, Durojaiye OA.Serum samples obtained from 107 Polo horses showing clinical signs of viral respiratory disease were tested for precipitating antibodies to adenovirus by agar gel precipitation test and counter-immunoelectrophoresis method. The results obtained demonstrate serological evidence of adenovirus infection in Polo horses in Nigeria. The counter-immunoelectrophoresis method was observed to be about 3 times more sensitive than the agar gel precipitation test with 19.3 vs 64.5%. It could thus be used to screen a large number of serum samples within a short period.
Equine monocytic ehrlichiosis (Potomac horse fever): a review.
Equine veterinary journal    November 1, 1991   Volume 23, Issue 6 400-404 doi: 10.1111/j.2042-3306.1991.tb03749.x
Mulville P.In the late 1970s, a new infectious disease in horses, involving acute enteritis, was recognised in the Potomac River area of Maryland, U.S.A. The causative agent was identified subsequently as a new species of rickettsial organism, later named Ehrlichia risticii. Since then, the disease has been reported in many other states, and in enzootic areas vaccination is common. Signs associated with the clinical disease included depression, fever, anorexia, decreased or absent intestinal sounds, profuse watery diarrhoea and laminitis. However, considerable variation in clinical manifestations has bee...
Attempts to immunoprotect adult horses, specifically pregnant mares, with commercial vaccines against clinical disease induced by equine herpesvirus-1.
Zentralblatt fur Veterinarmedizin. Reihe B. Journal of veterinary medicine. Series B    August 1, 1991   Volume 38, Issue 6 432-440 doi: 10.1111/j.1439-0450.1991.tb00892.x
Bürki F, Nowotny N, Oulehla J, Schmehlik O, Möstl K, Pallan C, Rossmanith E.In a project lasting 4 years more than 300 Lipizzans, around 180 of them adults, were vaccinated systematically against Equine Herpesvirus-1 (EHV-1) and representative groups thereof were serologically controlled for their antibody responses. In part, vaccination intervals recommended on packing slips were followed, in part other intervals, implicated by intermediary results, were used. A live virus vaccine proved ineffective if humoral antibodies were present. An oil-adjuvanted vaccine proved of highest antiviral immunogenicity, but after repeated revaccinations caused severe local reactions ...
Comparative evaluation of enzyme-linked immunosorbent assay (ELISA) for the serodiagnosis of dourine.
Veterinary parasitology    August 1, 1991   Volume 39, Issue 3-4 233-239 doi: 10.1016/0304-4017(91)90040-3
Wassall DA, Gregory RJ, Phipps LP.The detection of antibodies against Trypanosoma equiperdum in 689 equid sera was compared by enzyme-linked immunosorbent assay (ELISA), the complement fixation test (CFT) and an indirect immunofluorescent test (IIF). CFT was the least sensitive technique, susceptible to anti-complementary factors and the most technically demanding. IIF was more sensitive, but was only suitable for testing limited numbers of samples. In this study, ELISA was the most sensitive test, the least labour intensive and lends itself to a considerable degree of automation. It is suggested that ELISA would be relatively...
[Equine monocytic ehrlichiosis (EME), a review].
Tijdschrift voor diergeneeskunde    July 15, 1991   Volume 116, Issue 14 721-727
van der Kolk JH, de Groot J.Serological surveys showed that equine monocytic ehrlichiosis (EME) occurs in the USA, Canada and Europe. The causative agent is Rickettsia Ehrlichia risticii, isolated for the first time in 1984. The clinical features of the disease are sluggishness, anorexia, colic and fever, possibly followed by watery diarrhoea. Complications of an infection with E. risticii are laminitis and abortion. Colitis of the ascending colon may be observed at autopsy. Following a positive serological diagnosis (IgM ELISA) of EME, treatment with oxytetracycline can be initiated. It is also important to restore the ...
